Alterations in the synthesis of melanin contribute to a number of diseases; therefore, the design of new tyrosinase inhibitors is very important. Mushroom tyrosinase (MT) is a metalloenzyme, which plays an important role in melanin biosynthesis. In this study, the inhibitory effect of a novel designed compound, i.e. 2-((1Z)-(2-(2,4-dinitrophenyl)hydrazin-1-ylidene)methyl)phenol, as a specific ligand which can bind to the copper ion of MT, has been assessed. The ligand was found to competitively inhibit both the cresolase and catecholase activities of MT, with small inhibition constants of 2.8 and 2.6?μM, respectively. Intrinsic fluorescence studies were performed to gain more information on the binding constants. Docking results indicated that the ligand binds to copper ions in the active site of MT via the OH group of the ligand. The ligand makes four hydrogen bonds with aspartic acid and one hydrogen bond with the histidine residue in the active site. Molecular dynamics results show that ligand binds to the MT via both electrostatic and hydrophobic interactions with its different parts.  相似文献

The Michaelis constant of catecholase in 3 cultivars of Dioscorea alata was determined. Oxidative browning was related to the activity of the enzyme in the yams. Inhibition by cysteine was competitive, while 2-mercaptoethanol caused an uncompetitive type of inhibition. On DEAE column chromatography a minor peak preceded the major peak in the fresh yam while a minor peak was eluted after the major peak in the stored yam.  相似文献

On gamma irradiation of potato tubers at sprout-inhibiting dose (10 krad) the cresolase activity showed a 45% increase while catecholase was reduced by 25%. This reduced the ratio of catecholase to cresolase from 11–12 in unirradiated to 5–6 in irradiated potatoes. Chlorogenic acid oxidation was enhanced by about 25% on irradiation. The increase in the oxidation of p-cresol corresponded with the production of diphenolic compounds. The process of activation of cresolase was slow, reversible and oxygen dependent. A comparative study of the isoenzyme pattern suggested that this activation was due to conformational change, rather than synthesis of new protein.  相似文献

Summary Diphenolases from Anoxybacillus kestanbolensis strains K1 and K4T, highly active against 4-methylcatechol were characterized in terms of pH- and temperature-optima, pH- and temperature-stability, kinetic parameters, and inhibition/activation behaviour towards some general polyphenol oxidase (PPO) inhibitors and metal ions. The temperature-activity optima, for Anoxybacillus kestanbolensis K1 and K4T catecholases in the presence of 4-methylcatechol, were 80 and 70 °C, respectively. Although catecholase from A. kestanbolensis K4T lost no activity after a period of 1 h incubation at its optimum temperature, the enzyme pH from K1 was stimulated by keeping at 80 °C. Both of the enzymes possessed pH optima at 9.5, and the pH-stability profiles showed that cathecholases from both preparations retained their activities at alkaline pH values. Both A. kestanbolensis K1 and K4T catecholase activities were totally inhibited by addition of 0.01 mM sodium metabisulphite, ascorbic acid and l-cysteine. 1 mM Mn2+ increased the activities of A. kestanbolensis K1 and K4T catecholases by 6.4- and 5.3-fold, respectively. These results indicate that both A. kestanbolensis K1 and K4T strains possess thermo- and alkalostable catecholases.  相似文献

The synthesis of metal complexes has vastly increased the scope of research for many scientists during the two last decades. Among these compounds, artificial tyrosinases, catecholases, proteases, and nucleases are some of the most studied due to their importance as modern tools in the fields of medicine, scientific research, and industry. Transition metals such as Zn2+, Cu2+, Fe3+, Co3+, Ni2+, and lanthanide ions are the most commonly used. Among these ions, copper complexes have been the focus of the majority of studies thanks to their significant activity in comparison with other ions. Studies of copper-based tyrosinases, catecholases, and nucleases have revealed some of the overarching factors affecting reactions of all three types, which has led to improved activity and efficiency for all. Key factors include proper core–core distance, (Cu?Cu distance 2.90–2.99?Å), suitable solvent, and ligands with proper hydrophobic structure and geometry. In the present investigation, we review and introduce the proposed mechanisms and the kinetically effective factors of natural catecholase, tyrosinase, and nuclease and their Cu-based synthetic mimics.  相似文献

The catecholase activities were routinely modeled using transition metal complexes as catalyst and in some case basic pH were used as a reaction condition. In this article, the catalytic aerobic oxidation of proxy substrate 3,5-di-tert-butylcatechol (DTBC) in methanol using triethylamine/diethylamine as catalyst was demonstrated as a functional mimic of catecholase activity. The kinetic manifestation of DTBC oxidation was explained as enzymatic substrate inhibition pattern in Michaelis-Menten kinetic model. The mechanistic insight of the aerobic oxidation of DTBC was further validated using various spectroscopic techniques and DFT methods.  相似文献

Effects of juglone on seedling growth of cucumber (Cucumis sativus cv. Beith Alpha) with respect to physiological and anatomical parameters were investigated. Growth parameters (seedling elongation, fresh and dry weights) were reduced by 1 mM juglone. Juglone also decreased chlorophyll a and b contents and reduced some anatomical tissues (xylem vessel and bundle radius of stem, stomata length and stomata number of the cotyledons). The anatomical changes in stem and cotyledon of the seedlings were related to growth inhibiting effect of juglone. On the other hand, increase in catecholase and tyrosinase activities by the effect of juglone were also recorded.  相似文献
