The collapse of the Classic Maya state is investigated from an ecological perspective. Settlement and palynological data from the Maya center of Copan, Honduras, are presented which indicate that substantial clearing of the upland pine forest had occurred prior to and during the abandonment of that urban center. A comparative use- rate analysis suggests that the increased clearing of pine was primarily caused by demands for domestic fuel wood by an expanding urban population. This forest mismanagement is directly linked to accelerated erosion rates which are considered primary elements in the collapse of the Maya state.  相似文献

The mode of antitumor action of rHu-TNF was elucidated in BALB/c mice bearing Meth A fibrosarcoma 7 days after transplantation with respect to time course, dose-response relationships and selectivity of the effects. The maximal cytotoxic effect on tumor cells revealed by inhibition of DNA synthesis and maximal lesional effect on tumor vasculature revealed by change in blood pool-size in the tissue were detected at 30 min and I h after administration of rHu-TNF, respectively. The dose-response relationship between cytotoxic and tumoricidal effects of rHu-TNF was irrespective of administration route. ED50s of these antitumor effects afteri.v. administration of rHu-TNF were about 50 times as high as ED50s afteri.t. administration. ED50 ofi.t. given rHu-TNF for vascular effect was about 20 times as high as that for cytotoxicity while ED50 ofi.v. rHu-TNF for vascular effect was only 2–3 times as high as that for cytotoxicity. The whole body autoradiographies with [125I] HSA giveni.v. to see the blood influx into tumor tissue and [14C]thymidine given i.v. to see DNA synthesis in the whole body after administration of rHu-TNF revealed that the distribution of radioactivity was markedly changed in the tumor alone without any detectable change in other whole body tissues.In conclusion, thein vivo antitumor effect of rHu-TNF giveni.t. ori.v., appears to be exerted through the direct action on Meth A sarcoma rather than indirectly on tumor vasculature. Under present conditions, the effect of rHu-TNF in the whole body tissues seems rather selective on cells and vasculature of the tumor.  相似文献

Small marine pelagic fish and the threat of fishing; are they endangered?   总被引:1,自引:0,他引:1  
Evidence of the collapse and recovery of major marine fisheries for pelagic fish species is reviewed, distinguishing the influence of fishing compared with natural (environmental) effects. In one only of the best documented cases (Icelandic spring-spawning herring) has the stock failed to reappear, after 20 years. Several others, e.g. California sardine, have persisted at 1/000th or possibly less of their peak size for some years before beginning to recover. Fishing has been the main cause of collapse in most but not all cases, due initially to the escalation of catchability as stock size decreases–a phenomenon characteristic of fisheries for pelagic species due to their shoaling habit, ease of detection and vulnerability to modern fishing methods. It is concluded that although the threat of fishing to the continuity of the species is remote, excessive depletion (although avoidable by firm and timely management) is potentially able to cause temporary disappearance of local stock and disruption of the ecosystem.  相似文献

Since cytoplasmic Ca2+ levels are reported to regulate neurite elongation, we tested whether calcium-activated kinases might be necessary for growth cone motility and neurite elongation in explant cultures of goldfish retina. Kinase inhibitors and activators were locally applied by micropipette to retinal growth cones and the responses were observed via phase-contrast videomicroscopy. In some cases, growth rates were also quantifed over several hours after general application in the medium. The selective inhibitors of protein kinase C, calphostin C (0.1–1 μM) and chelerythrin (up to 50 μM), caused no obvious changes in growth cones or neurite elongation, and activators of PKC (phorbols, arachidonic acid, and diacylglycerol) also were generally without effects, although phorbols slowed the growth rate. Inhibitors of protein kinase A and tyrosine kinases also produced no obvious effects. The calmodulin antagonists, calmidazolium (0.1 μM), trifluoperazine (100 μM), and CGS9343B (50 μM), however, caused a reversible growth cone arrest with loss of filopodia and lamellipodia. The growth cone became a club-shaped swelling which sometimes moved a short distance back the shaft, leaving evacuated filaments at points of strong filopodial attachments. A similar reversible growth cone arrest occurred with the general kinase inhibitors: H7 at 200 but not at 100 μM, and staurosporine at 100 but not 10 nM, suggesting possible involvement of a calmodulin-dependent kinase (camK) rather than PKC. The selective inhibitor of camKII, KN-62 (tested up to 50 μM), produced no effects but the specific myosin light-chain kinase (MLCK) inhibitors ML-7 (3–5 μM) and ML-9 (5–10 μM) reversibly reproduced the effect, suggesting that MLCK rather than camKII is necessary for growth cone motility. The MLCK inhibitors' effects both on growth cone morphology and on F-actin filaments (rhodamine-phalloidin staining) were similar to those caused by cytochalasin D (5 μM), and are discussed in light of findings that inhibiting MLCK disrupts actin filaments in astrocytes and fibroblasts. 1994 John Wiley & Sons, Inc.  相似文献

The validity of Connell's intermediate disturbance hypothesis in phytoplankton communities was tested on data from a hypertrophic, shallow lake, Hjarbæk Fjord, Denmark.The present data from Hjarbæk Fjord demonstrate the difficulties in distinguishing stress from disturbance in a phytoplankton community, and show that great changes in the phytoplankton community can take place within few days.A collapse of blue-green algae in late June 1986 caused remineralization of nutrients and resulted in a rapid increase of fast-growing small chlorococcal green algae and phytoplankton species diversity, without any external disturbances acting on the lake. External disturbances in the form of wind action and brackish water intrusion occurred several days after the onset of these events. Carbon depletion and pH 11.0 were severe stress factors on the phytoplankton community. They were induced by calm, warm weather, but eventually acted as a kind of disturbance to the normally well circulated lake.  相似文献

The isolated, 101-residue long C-terminal (so called F2) fragment of the beta chain from Escherichia coli tryptophan synthase was shown previously to fold into an ensemble of conformations that are condensed, to contain large amounts of highly dynamic secondary structures, and to behave as a good model of structured intermediates that form at the very early stages of protein folding. Here, solvent perturbations were used to investigate the forces that are involved in stabilizing the secondary structure (monitored by far-UV CD) and the condensation of the polypeptide chain (monitored by dynamic light scattering) in isolated F2. It was observed that neither the ionic strength, nor the pH (between 7 and 10), nor salts of the Hofmeister series affected the global secondary structure contents of F2, whereas some of these salts affected the collapse slightly. Addition of trifluoroethanol resulted in a large increase in both the amount of secondary structure and the Stokes radius of F2. Conversely, F2 became more condensed upon raising the temperature from 4 to 60 degrees C, whereas in this temperature range, the secondary structure undergoes significant melting. These observations lead to the conclusion that, in isolated F2, there is no coupling between the hydrophobic collapse and the secondary structure. This finding will be discussed in terms of early events in protein folding.  相似文献

This study evaluates trepanations from five well‐contextualized prehistoric sites in the south‐central highlands of Andahuaylas, Peru. The emergence of trepanation in this region coincides with the collapse of the Wari Empire, ca. ad 1000. Thirty‐two individuals from Andahuaylas, AMS radiocarbon dated to the early Late Intermediate Period (ca. ad 1000–1250), were found to have 45 total trepanations. Various surgical techniques were being employed concurrently throughout the region. Scraping trepanations evinced the highest survival rate; circular grooving, drilling and boring, and linear cutting were far less successful. Evidence of perioperative procedures like hair shaving, poultice application, and possible cranioplasty use aimed to ensure the survival of a trepanation recipient. Postmortem trepanations, also present in Andahuaylas, were likely executed on corpses as a means of better understanding cranial anatomy and improving techniques. Similarities in trepanation patterns throughout the region attest to common motivations to engage in surgery. Although moderate physical head trauma seems to be the impetus for intervention in many cases of trepanation, other motivations included physiological and possibly psychosomatic factors. Nevertheless, treatment was not for everyone. In Andahuaylas, trepanations were withheld from subadults, females, and those individuals who practiced cranial modification. 目的:探讨磁共振体素内不相干运动扩散加权成像(intravoxel incoherent motion diffusion weighted MR Imaging,IVIM-DWI)在肺癌所致肺不张的影像诊断中的初步应用。方法:选取独立肺段肺不张病变患者43例,其中肺癌致肺不张31例(肺癌组),局部炎症致肺不张12例(炎症组)。所有患者在药物治疗或手术前均行3.0T磁共振IVIM-DWI检查,b值取0、50、100、200、400、600、800、1000 s/mm~2,分别测量灌注分数(perfusion fraction,f)、真实扩散系数(diffusion coefficient,D)以及关注相关扩散系数(diffusion coefficient from the perfused compartment,D*),对上述参数值进行相关统计学分析,并依据受试者工作特征曲线(Receiver Operating Characteristic,ROC)对各参数的评价效能进行分析。结果:肺癌组的D值、D*值以及f值分别为(0.64±0.16)×10~(-3)mm~2/s、(19.77±6.16)×10~(-3)mm~2/s以及(29.62±9.74)%,而炎症组的D值、D*值以及f值分别为(0.67±0.14)×10~(-3) mm~2/s、(21.14±8.32)×10~(-3)mm~2/s以及(47.62±11.46)%;经比较,肺癌组D值、D*值与炎症组无统计学差异(P0.05),而二者f值差异显著,有统计学意义(P0.01)。f值最佳阈值为38.42%,此时曲线下面积(Area Under Curve,AUC)为0.94,诊断肺癌的特异度为0.89,敏感度为0.93,阳性预测值为0.92,阴性预测值为0.89。结论:磁共振IVIM-DWI技术在肺癌所致肺不张的影像诊断中具有一定的应用价值。  相似文献

Free‐living marine nematode communities of the Larsen B embayment at the eastern Antarctic Peninsula were investigated to provide insights on their response and colonization rate after large‐scale ice‐shelf collapse. This study compares published data on the post‐collapse situation from 2007 with new material from 2011, focusing on two locations in the embayment that showed highly divergent communities in 2007 and that are characterized by a difference in timing of ice‐shelf breakup. Data from 2007 exposed a more diverse community at outer station B.South, dominated by the genus Microlaimus. On the contrary, station B.West in the inner part of Larsen B was poor in both numbers of individuals and genera, with dominance of a single Halomonhystera species. Re‐assessment of the situation in 2011 showed that communities at both stations diverged even more, due to a drastic increase in Halomonhystera at B.West compared to relatively little change at B.South. On a broader geographical scale, it seems that B.South gradually starts resembling other Antarctic shelf communities, although the absence of the genus Sabatieria and the high abundance of Microlaimus still set it apart nine years after the main Larsen B collapse. In contrast, thriving of Halomonhystera at B.West further separates its community from other Antarctic shelf areas.  相似文献
