The oxidized B chain of insulin was used as a simple model for further consideration of limited proteolysis with low substrate:enzyme ratios. With low B chain:trypsin ratios, the ordinarily slower cleavage rate of the -Lys29-Ala30 bond essentially equaled the cleavage saturation rate of the -Arg22-Gly23 bond. This led to the disappearance of octapeptide which ordinarily forms most rapidly. Heptapeptide and alanine, formed mainly by cleavage of the octapeptide, decreased somewhat at high enzyme relative levels. Trypsin added to B chain formed a single chromatographic peak. 相似文献

1. Larvae of antlions (Neuroptera: Myrmeleontidae) and wormlions (Diptera: Vermileonidae) display a convergently evolved sit‐and‐wait hunting strategy of building pitfall traps in sandy areas. This study investigated a sympatric population of antlions and wormlions in the lowland rainforest of Borneo for substrate moisture, particle size and temperature preferences. It was hypothesised that these animals would show different preferences regarding these microhabitat traits. 2. The results showed that antlions had a higher aversion to moisture compared with wormlions, but that wormlions had a higher preference for small‐particle sand. Furthermore, thermal preferences in antlions and wormlions were significantly different, with antlions choosing higher temperatures. 3. The detected differences between antlions and wormlions might contribute to their niche partitioning in the mixed Bornean population and thus facilitate coexistence of these animals. It is possible that the hotter and dryer microhabitat edges are preferred by antlions. 相似文献

The mechanical properties of cells are influenced by their microenvironment. Here we report cell stiffness alteration by changing the cell substrate stiffness for isolated cells and cells in contact with other cells. Polydimethylsiloxane (PDMS) is used to prepare soft substrates with three different stiffness values (173, 88 and 17 kPa respectively). Breast cancer cells lines, namely HBL-100, MCF-7 and MDA-MB-231 with different level of aggressiveness are cultured on these substrates and their local elasticity is investigated by vertical indentation of the cell membrane. Our preliminary results show an unforeseen behavior of the MDA-MB-231 cells. When cultured on glass substrate as isolated cells, they are less stiff than the other two types of cells, in agreement with the general statement that more aggressive and metastatic cells are softer. However, when connected to other cells the stiffness of MDA-MB-231 cells becomes similar to the other two cell lines. Moreover, the stiffness of MDA-MB-231 cells cultured on soft PDMS substrates is significantly higher than the stiffness of the other cell types, demonstrating thus the strong influence of the environmental conditions on the mechanical properties of the cells. 相似文献

Supplementation of the growing substrate by nitrogenous additives has been known to improve the production of oyster mushroom (Pleurotus ostreatus (Jacq. ex Fr.) P. Kumm. (1871)). However, the application of nano-additives has not been reported in such cultivation yet. The study investigated the effect of nano-urea added in two different doses (3 g and 5 g per kg substrate), once (at spawning or after first flush) or twice (at spawning and after first flush) to the growing substrate consisting of wheat straw and spent oyster substrate (1:1, w/w). Results showed that the application of nano-urea once has induced the highest number of mushroom flushes (four flushes) despite the dose applied. Contrarily to early findings, where high doses of nitrogen have caused inhibition of mushroom growth and production, nano-urea application has had better effects when applied twice. With 5 g/kg, it induced the shortest period between the first and the third flush (15 days). With 3 g/kg, it resulted in the highest biological and economic yields at the third flush (332.7 g/bag and 283.1 g/bag respectively), in total (973.4 g/bag and 854.0 g/bag respectively), the highest biological efficiency (109.6%), and pileus diameter/stipe length ratio (2.8). Methanogenic populations were investigated in subsaline Laguna Potrok Aike sediments, southern Argentina. Microbial density and activity were assessed via cell count and in situ ATP detection for the last ~11K years. Methanogen phylogenetics highlighted species stratification throughout depth, whereas CO2 reduction was the major pathway leading to methane production. Organic substrates, characterized using pore water analysis, bulk organic fractions and saturated fatty acids, showed a clear link between sediment colonization and initial organic sources. Concentrations and δ13C compositions of methane and fatty acids provided final evidence of a microbial imprint on Holocene organic proxies in the most colonized intervals. 相似文献

Substrate particle size affects pit building decision and pit size in the antlion larvae Euroleon nostras (Neuroptera: Myrmeleontidae) 总被引:2,自引:0,他引:2
Abstract. The larvae of the antlion Euroleon nostras are pit-builders, constructing pitfall traps in loose sand. The number of pits and the pit diameter are recorded when larvae are kept in substrates with different particle sizes. The most convenient pit-building sand fractions are two fractions with fine sand (≤ 0.23 mm; 0.23–0.54 mm). The largest pits are constructed in sand with a particle size of 0.23–0.54 mm. In this sand fraction, larvae of all three instars most readily build pits. No pits are constructed in sand with a particle size greater than 1.54 mm. First- and second-instar larvae avoid building pits in substrates of particle size 1–1.54 mm, but third-instar larvae construct pits in this sand fraction. It is assumed that the antlion is capable of distinguishing between substrate types and this hypothesis is tested by giving larvae the choice of building a pit in one of four particle-size fractions. Larvae of all three instars prefer to build pits in the fraction with a particle size of 0.23–0.54 mm. A quantitative model of ammonia volatilization from the calcareous soil uppermost 1-cm layer was developed and tested. The
model accounts for the following processes: ammonium-ammonia equilibration in the soil solution, cation exchange between calcium
and ammonium which results in ammonium distribution between soil liquid and solid phases, nitrification of dissolved ammonium,
distribution of ammonia between liquid and gaseous phases and diffusion of gaseous ammonia in the soil air.
The combined effect of various characteristics such as soil pH, cation exchange capacity, water capacity and nitrification
rate on ammonia losses from various soil types have been studied. The model was validated against experimental results of
ammonia losses from different soils for its use as a predicting tool.
The model shows that most of ammonia losses can be explained by the interactive effect of high soil pH and low cation exchange
capacity. Computations show increased ammonia volatilization with decreasing soil water capacity. Increasing fertilizer application
rate has a small effect on percentage of ammonia losses. Increased nitrification rate and shorter “lag” period of nitrification
reduce ammonia losses considerably. Good agreement was obtained between model calculations and experimental results of ammonia

The influence of vertical nutrient transport on epilimnetic phosphorus was studied comparatively in four calcareous Wisconsin lakes during 1972. In the two lakes with steep metalimnetic nutrient gradients (Mendota, Delavan), upward fluxes by entrainment and eddy diffusion exceeded all other influxes combined; here epilimnetic total-P concentrations increased during period of high windpower and thermocline migration, and decreased during comparatively stagnant intervals. In the two lakes lacking upper metalimnetic P gradients (Green, Fish), higher windpower had little or no effect on epilimnetic phosphorus.In each of the four lakes epilimnetic P declined in early summer until a quasi steady-state was achieved between P influxes and P sedimentation. In Mendota and Delavan steady-state featured higher concentrations of total-P, and much higher epilimnetic concentrations of particulate-P and chlorophyll, than in Green and Fish Lakes — mainly on account of the high fluxes of molybdate-reactive, biologically-available P through the seasonal thermocline. The flux analysis illustrates why mean lake TP concentration after ice-out is an inconsistent measure (not sufficient statistic) for estimating nutrient potential and chlorophyll standing crops during the following summer in stratified lakes. 相似文献

Effect of simulated forest fire on the availability of N and P in mediterranean soils 总被引:4,自引:0,他引:4
The effect of soil burning on N and P availability and on mineralization and nitrification rates of N in the burned mineral
soil was studied by combustion of soils in the laboratory.
At a fire temperature of 600°C, there was a complete volatilization of NH4 and a significant increase of pH, from 7.6 in the unburned soil to 11.7 in the burned soil. Under such conditions ammonification
and nitrification reactions were inhibited. Less available P was produced immediately after the fire at 600°C, as compared
to P amount produced at 250°C. Burning the soils with plants caused a decrease in NH4-N and (NO2+NO3)-N concentrations in the soil as well as a reduction in ammonification and nitrification rates. Combustion of soil with plants
contributed additional available P to the burned soil.
The existence of a non-burned soil under the burned one played an important role in triggering ammonification and nitrification
