Summary The localization and orientation of cytoskeletal elements in developing cotton fibres were studied by the indirect immunofluorescence and the dry cleaving technique. Microtubules are transversely arranged to the cell axis, most probably in a flat helix, in the cortex of expanding fibres. Since the innermost deposited cellulose microfibrils always show primarily the same orientation it is postulated that the microtubules control the transverse deposition of the cellulose fibrils. Little further cell expansion takes place during secondary wall formation and the microfibril pattern corresponds to that of the cortical microtubules,e.g., in the steepness of their helicoidal turns. Microtubules with a length of 7–20 m were observed, probably they are longer. The importance of microtubule length on microfibril deposition is discussed. The density of microtubule packing is in the range of 8–14 m-1 as in other comparable cell types. In contrast to the microtubules, actin filaments are most likely longitudinally oriented during different phases of fibre development. The dry cleaving technique reveals numerous coated pits in the plasma membrane which are not crossed by microtubules. They seem to be linked to the latter by filamentous structures.  相似文献

Inhibition of endocytosis from coated pits by acidification of the cytosol   总被引:8,自引:0,他引:8  
Binding and endocytosis of the ligands transferrin, epidermal growth factor (EGF), and ricin were measured in a number of different cell lines after treatment of cells with compounds that react with SH-groups and under conditions where the cytosolic pH was lowered. N-ethylmalemide and diamide irreversibly inhibited endocytosis of all ligands tested, whereas low pH in the cytosol strongly inhibited endocytosis of transferrin and EGF. Data obtained by electron microscopy indicated that the formation of coated vesicles from coated pits is inhibited in acidified cells. Entry of ricin was much less affected, and ricin endocytosed under these conditions was able to intoxicate the cells. At low pH in the cytosol there was a calcium-dependent increase in the number of transferrin receptors at the cell surface. The increase was even larger in the presence of the calcium ionophore A23187, whereas it was completely blocked by the calmodulin antagonists trifluoperazine and W7. The results show that endocytosis from coated pits can be inhibited in a reversible way by acidification of the cytosol and they suggest that a second pathway of endocytosis exists, possibly involving formation of vesicles from uncoated areas of the membrane.  相似文献

The freeze-substitution method was applied toVicia hirsuta root hairs to test its effectiveness in improving preservation of the cell ultrastruture. Freeze-substitution almost certainly represents more faithfully the structure of the hair cell. A previously unreported ‘pyriformis’ vesicle is described. Also unique to freeze-substituted material are coated secretory vesicles; a smooth plasma membrane profile; mitochondrial ribosomes; long microfilament bundles which are associated with vesicles, mitochondria, coated pits and coated vesicles; microtubule-associated filaments; well-preserved coated vesicles and coated pits with enclosing rings; a pliciform nucleus. The results are discussed in context of previous reports using conventional fixation techniques.  相似文献

NAIR, M. N. B. AND MOHAN RAM, H. Y., 1989. Vestured pits and vestured vessel member walls in some Indian dicotyledonous woods. The woods of 144 taxa belonging to 38 families of angiosperms were examined for vestured pits and vestured vessel member walls using scanning electron microscopy. Vestured pits are present in 66 taxa (belonging to ten families) and vestured vessel member walls occur in only six taxa (belonging to three families). In Ehretiaceae and Euphorbiaceae vestures are present only in certain vessel members. In Wrighlia tinctoria , perforation plates containing vestures have been observed in addition to the presence of vestured pits. A classification of vestured pits based on their morphology and distribution is proposed by us. In all the types of vestured pits, vestures are present on the margin of the outer pit aperture or on the pit chamber wall. Occasionally, they are present in the pit canal, on the margin and in the vicinity of the inner pit aperture and rarely over the inner walls of the vessel members. The functions of vestured pits are not clear, although several suggestions are made. Whether or not these structures affect wood processing is not presently understood. It appears that vestured pits and vestured vessel member walls have diagnostic rather than phylogenetic value.  相似文献

The populations of tench from two gravel pit lakes of contrasting habitat type were studied. The population density of tench from St Peter's Lake, which has abundant aquatic vegetation was 176 individuals ha−1 (102.2 kg ha−1) compared to 0.3 individuals ha−1 (0.5 kg ha−1) in the Main Lake which has very little aquatic vegetation. The results of ageing tench by scales, opercular bones and otoliths were compared; all three structures gave similar results up to 9 years of age, but thereafter there were difficulties in ageing using scales and care was needed in counting annuli. The oldest tench caught was 15 years old. Growth of male and female tench was similar and fitted the von Bertalanffy model; for female tench L = 573 mm, K =0.122 and for male tench L = 586 mm, K =0.114. Recruitment was variable and strong year classes occurred in years with warm summers.  相似文献

The localization of thrombin receptors on mouse embryo (ME) cells was examined using electron microscope (EM) immunocytological techniques. ME cells were fixed with formaldehyde, prior to thrombin binding, and thrombin visualized on cell surfaces using affinity-purified antithrombin rabbit antibody and colloidal gold labeled anti-rabbit IgG. Colloidal gold particles were found in clusters on the surface of cells incubated with thrombin. There were approximately seven particles per cluster observed in thin sections with cluster diameters ranging from 70 to 200 nm. These clusters were not observed on cells incubated without thrombin. The total number of particles present on cells incubated with and without thrombin indicate that the colloidal gold labeling is approximately 98% specific for thrombin. Only four colloidal gold particles out of approximately 1,200 were associated with coated pits. Thus the thrombin receptor clusters do not appear to associate with coated membrane regions. To determine whether receptor-bound thrombin was internalized by receptor-mediated endocytosis, ME cells were incubated with 125I-thrombin and examined using EM autoradiography and the trypsin sensitivity of 125I-thrombin which was associated with the cells. In two types of experiments, where thrombin was incubated with cells at 4 degrees C and the temperature increased to 37 degrees C and where initial incubation was at 37 degrees C, the receptor-directed specific internalization proceeded at approximately the same rate as nonspecific internalization. These studies indicate that thrombin that binds to its receptors on ME cells is not rapidly internalized by receptor-mediated endocytosis.  相似文献

Drought induces an increase in a tree's vulnerability to a loss of its hydraulic conductivity in many tree species, including two common in western Canada, trembling aspen (Populus tremuloides) and balsam poplar (Populus balsamifera). Termed ‘cavitation fatigue’ or ‘air‐seeding fatigue’, the mechanism of this phenomenon is not well understood, but hypothesized to be a result of damage to xylem pit membranes. To examine the validity of this hypothesis, the effect of drought on the porosity of pit membranes in aspen and balsam poplar was investigated. Controlled drought and bench dehydration treatments were used to induce fatigue and scanning electron microscopy (SEM) was used to image pit membranes for relative porosity evaluations from air‐dried samples after ethanol dehydration. A significant increase in the diameter of the largest pore was found in the drought and dehydration treatments of aspen, while an increase in the percentage of porous pit membranes was found in the dehydration treatments of both species. Additionally, the location of the largest pore per pit membrane was observed to tend toward the periphery of the membrane.  相似文献

In several psammophine snake species, small sense-organ-like pits have been discovered occurring on the top of the head. These pits show a heretofore non-described structure. In numbers of one to four per head, they are present in Dromophis lineatus, Malpolon monspessulanus, several Psammophis species and Rhamphiophis rubropunctatus, but not in all individuals which is unrelated to sex. The reason for their sporadic occurrence remains obscure. Their equally obscure function is discussed in relation to the presumed functions of already known upper-head 'pits' in snakes. Phylogenetic analysis indicates that Lycastinae should be incorporated into a more broadly defined Maxillariinae. This is supported by several anatomical features, including the presence of sunken, glandular trichomes in both Lycastinae and Maxillariinae s.s. Until recently, these were known only from vegetative organs, but have since been reported from flowers of Maxillaria dichroma. One character currently used to distinguish between Lycaste and Sudamerlycaste is the distribution of floral trichomes. In this article, we test the reliability of this character, describe the floral micromorphology of Lycaste and Sudamerlycaste and investigate whether their flowers bear sunken hairs. Their floral micromorphology is compared with that of other genera currently assigned to Maxillariinae s.l. Flowers of Lycaste and Sudamerlycaste bear conical or obpyriform papillae and unbranched and unequally branched multicellular trichomes. Contrary to previous reports that trichomes are confined to the column in Sudamerlycaste, they also occur in the tepal axils. Labellar trichomes, although often present in Lycaste, are lacking in Sudamerlycaste. In Lycaste sections Lycaste and Aromaticae, floral trichomes tend to be unbranched, whereas section Intermediae has both unbranched and branched hairs. Branched hairs are more common in Sudamerlycaste. Some hairs are tracheoidal, pitted and lignified. These mainly occur in section Lycaste and, to a degree, in section Intermediae, but are absent from section Aromaticae and most species of Sudamerlycaste. Branched column hairs, present in Sudamerlycaste, are absent from all sections of Lycaste, and tracheoidal column hairs occur only in Sudamerlycaste. Sunken floral hairs are absent from both genera.
