On the mechanism of speed and altitude control in Drosophila melanogaster   总被引:1,自引:0,他引:1  
ABSTRACT The total power output of tethered flying Drosophila melanogaster in still air depends on translational velocity components of image flow on the eye, whereas the orientation of the average flight force in the midsagittal plane of the fly is widely independent of visual input (Götz, 1968). The fly does not seem to control the vertical and the horizontal force component independently. Freely flying flies nevertheless generate different ratios between lift and thrust, simply by changing the inclination of their body. By the combined adjustment of the body angle and the total power output a fly appears to be able to stabilize height and speed (David, 1985). Here a possible mechanism is proposed by which the appropriate torque about the transverse body axis could be generated. Translational pattern motion influences the posture of the abdomen and the plane of wing oscillation. Thus the position of the centre of gravity relative to the flight force vector is changed. When abdomen and stroke plane deviate from an equilibrium state, a lever is generated by which the force vector will rotate the fly about its transverse axis.  相似文献

蝮蛇抗栓酶治疗特发性突发性感音神经性耳聋的临床研究   总被引:1,自引:0,他引:1

我院自1991年3月至1992年11月底共收治特发性突发性感音神经性耳聋(突聋)80例,随机分成治疗组和对照组。治疗组40例,44耳,经蝮蛇抗栓酶治疗,治愈率47.7%,有效率81.8%。对照组40例,48耳,经用低分子右旋糖酐、丹参、病毒唑等药物综合治疗,治愈率25%,有效率50%,治愈率及有效率,治疗组明显高于对照组,经统计学处理,有显著性差异(P<0.05,P<0.01)。据血液流变学资料分析,治疗组治疗后全血低切粘度,高切粘度等五个指标明显下降。研究表明,蝮蛇抗栓酶治疗突聋,疗效高,无明显毒副作用,为治疗突聋开辟了新途径。  相似文献

We examined the subcellular localization and function of several Cx26 mutants that exhibit both sensorineural deafness and various skin disease phenotypes. To facilitate these aims, all Cx26 mutants were tagged at the carboxyl-terminal with green fluorescent protein (GFP), which has previously been shown not to affect Cx26 transport, assembly or function. In this article we focus on two point mutations (R75W and ΔE42) that occur in the first extracellular loop region of Cx26, a region hypothesized to be critical for correct hemichannel docking between contacting cells. In gap junctional intercellular communication (GJIC)-deficient HeLa cells, both R75W-GFP and ΔE42-GFP were transported to the cell surface and assembled into gap junction-like structures. Neither R75W-GFP nor ΔE42-GFP formed gap junctions that were permeable to Lucifer Yellow suggesting they are loss-of-function mutations. We also examined the phenotype of these two mutations in a rat epidermal keratinocyte (REK) cell line that is capable of undergoing differentiation. Using antibodies against several members of the connexin family reportedly expressed by epidermal keratinocytes, we found these cells endogenously expressed Cx43 and Cx26 but not Cx30, Cx32, or Cx37. When expressed in REK cells, similar to in HeLa cells, R75W-GFP and ΔE42-GFP were assembled at the cell surface into structures that resembled gap junctions. Future experiments will examine the effect of the Cx26 mutants on the function and differentiation of these epidermal keratinocytes.  相似文献

The human deafness‐pigmentation syndromes, Waardenburg syndrome (WS) type 2a, and Tietz syndrome are characterized by profound deafness but only partial cutaneous pigmentary abnormalities. Both syndromes are caused by mutations in MITF. To illuminate differences between cutaneous and otic melanocytes in these syndromes, their development and survival in heterozygous Microphthalmia‐White (MitfMi‐wh/+) mice were studied and hearing function of these mice characterized. MitfMi‐wh/+ mice have a profound hearing deficit, characterized by elevated auditory brainstem response thresholds, reduced distortion product otoacoustic emissions, absent endocochlear potential, loss of outer hair cells, and stria vascularis abnormalities. MitfMi‐wh/+ embryos have fewer melanoblasts during embryonic development than their wild‐type littermates. Although cochlear melanocytes are present at birth, they disappear from the MitfMi‐wh/+ cochlea between P1 and P7. These findings may provide insight into the mechanism of melanocyte and hearing loss in human deafness‐pigmentation syndromes such as WS and Tietz syndrome and illustrate differences between otic and follicular melanocytes.  相似文献

Several methods are currently available for selection when conducting sperm cryopreservation, however, these methods might cause different degrees of damage on sperm DNA. The aim of the this study is to compare the effects of storage at −80 °C (in ultra-low temperature refrigerator) and at −196 °C (in liquid nitrogen) on sperm DNA damage, thus to provide a reference for choosing the right method according to different aims. We randomly collected 28 semen samples from college students of Chongqing city. The samples stored at −80 °C were neat semen samples and the samples stored at −196 °C were mixed with additional cryoprotectants. Each sample was subjected to two freezing-thawing cycles, and the sperm DNA damage levels of fresh and thawed samples were measured by single cell gel electrophoresis (SCGE) and sperm chromatin structure assay (SCSA). Both SCGE and SCSA assays showed cryopreservation induced significant damage to sperm DNA. However, storage at −196 °C lead to more severe damage to sperm DNA than storage at −80 °C measured by SCSA. Sperm DNA damage increased simultaneously with the higher frequency of freezing-thawing cycles. We concluded that storage of neat semen samples at −80 °C had milder damage to sperm DNA than storage at −196 °C mixed with cryoprotectants. To avoid additional sperm DNA damage, repeated freezing and thawing should be prevented.  相似文献

Here we propose a bio-MEMS device designed to evaluate contractile force and conduction velocity of cell sheets in response to mechanical and electrical stimulation of the cell source as it grows to form a cellular sheet. Moreover, the design allows for the incorporation of patient-specific data and cell sources. An optimized device would allow cell sheets to be cultured, characterized, and conditioned to be compatible with a specific patient's cardiac environment in vitro, before implantation. This design draws upon existing methods in the literature but makes an important advance by combining the mechanical and electrical stimulation into a single system for optimized cell sheet growth. The device has been designed to achieve cellular alignment, electrical stimulation, mechanical stimulation, conduction velocity readout, contraction force readout, and eventually cell sheet release. The platform is a set of comb electrical contacts consisting of three-dimensional walls made of polydimethylsiloxane and coated with electrically conductive metals on the tops of the walls. Not only do the walls serve as a method for stimulating cells that are attached to the top, but their geometry is tailored such that they are flexible enough to be bent by the cells and used to measure force. The platform can be stretched via a linear actuator setup, allowing for simultaneous electrical and mechanical stimulation that can be derived from patient-specific clinical data.  相似文献

流速对红鳍银鲫幼鱼游泳状态的影响   总被引:7,自引:0,他引:7  
在28℃水温下,采用特制的鱼类游泳行为测定装置,研究体重(125.94±13.87)g的红鳍银鲫(Barbodes schwanenfeldi)幼鱼在0.0m/s(对照组)和0.1m/s、0.3m/s、0.5m/s3种流速下的游泳行为。结果表明,从0.0~0.3m/s,红鳍银鲫幼鱼平均趋流率和摆尾频率均随着流速的增加而增大,而0.5m/s组在90min内随时间延长而下降。红鳍银鲫游泳状态明显受到所处流速的影响,在静水对照组以"逆流前进"和"顺流而下"为主,两者共占总观察时间的98%以上;各流速组均以逆流静止为主,且随着流速的增大,逆流静止所占时间比例从45.8%增加至81.3%,而逆流前进所占时间比例由24.1%减至5%以下;逆流后退所占时间比例以0.1m/s组最大,为16.4%;顺流而下的比例随着流速增大先减小后增大,3个流速组依次为13.7%、2.1%和10.9%。红鳍银鲫幼鱼的游泳速度(V)和摆尾频率(TBF)在逆流前进及逆流静止两种游泳状态下呈线性正相关,而在逆流后退和顺流而下两种状态下两者没有显著相关。  相似文献

Abstract Myogenic contractions of the heart of the female blood-feeding insect, Rhodnius prolixus (Stål), are inhibited by crude extracts of testes applied directly to isolated dorsal vessels. Dorsal vessels were observed with a stereo microscope and heart beats timed with a stopwatch. In normal Rhodnius saline, hearts contract at 14.8 ± 7.1 beats per minute (n= 45). Crude extracts of the testes and the two male reproductive accessory organs (the opaque and transparent accessory glands) were prepared from previously frozen tissue by homogenizing 5–20 glands in a small glass homogenizer containing Rhodnius saline, centrifuging for 5 min at 2 000 g, and collecting the supernatant. Testes extract as low as 1.0 glands per mL inhibit contractions whereas crude extracts of the opaque or transparent accessory glands have no consistent effect. We refer to this cardiac inhibitor as rhodtestolin (Rhodnius testis inhibitory factor), and discuss its possible effects on the female during copulation.  相似文献

The time needed for juvenile cod Gadus morhua to recover from handling stress, and also whether observer presence or haemoglobin genotype influenced recovery time, were investigated. Time to recovery after handling was found to be 1·5 h based on opercular beat frequencies and behaviour. No effects of haemoglobin genotype or observer presence were found.  相似文献
