We report a new approach for molecular sex identification of extant Ursinae and Tremarctinae bears. Two Y‐specific fragments (SMCY and 318.2) and one X‐specific fragment (ZFX) are amplified in a multiplex PCR, yielding a double test for male‐specific amplification and an internal positive control. The primers were designed and tested to be bear‐specific, thereby minimizing the risk of cross‐amplification in other species including humans. The high sensitivity and small amplicon sizes (100, 124, 160 base pairs) facilitate analysis of non‐invasively obtained DNA material. DNA from tissue and blood as well as from 30 non‐invasively collected hair and faeces yielded clear and easily interpretable results. The fragments were detected both by standard gel electrophoresis and automated capillary electrophoresis.  相似文献

ABSTRACT

Cave bears have disappeared from the Alps from different altitudes at different times. The temporal progression of the HDEL (Height Dependent Extinction Line) – a compilation of the geologically most recent radiocarbon dates per altitude level – is not consistent with the general cooling of the temperatures from about 45 ka BP. The cave bear sites of the Northern Alps with the most recent radiocarbon ages are not situated in the lowlands but in caves in altitudes of 1,500 m to 1,700 m above sea level (a.s.l.).

Cave bears fed almost exclusively on herbs and leaves. It was assumed that with the general cooling in the OIS 3 since about 45 ka BP also the migration of the alpine elements into the lowlands took place. It could be recognized that the populations in the lower situated cave bear site became earlier extinct than the cave bear population in the higher altitudes.

With new radiocarbon dates, done at the Curt-Engelhorn-Center Archaeometry at the Reiss-Engelhorn-Museen in Mannheim (Germany), the HDEL can be determined much more precisely and the causes of gradual extinction are also better understood.  相似文献

The brown bears of coastal Alaska have been recently regarded as comprising from one to three distinct genetic groups. We sampled brown bears from each of the regions for which hypotheses of genetic uniqueness have been made, including the bears of the Kodiak Archipelago and the bears of Admiralty, Baranof and Chichagof (ABC) Islands in southeast Alaska. These samples were analysed with a suite of nuclear microsatellite markers. The 'big brown bears' of coastal Alaska were found to be part of the continuous continental distribution of brown bears, and not genetically isolated from the physically smaller 'grizzly bears' of the interior. By contrast, Kodiak brown bears appear to have experienced little or no genetic exchange with continental populations in recent generations. The bears of the ABC Islands, which have previously been shown to undergo little or no female-mediated gene flow with mainland populations, were found not to be genetically isolated from mainland bears. The data from the four insular populations indicate that female and male dispersal can be reduced or eliminated by water barriers of 2–4 km and 7km in width, respectively.  相似文献

Brown bears have lost most of their range on the European continent. The remaining western populations are small, isolated and highly endangered. The Dinaric-Pindos brown bear population is the western-most stable population and the fourth largest in Europe. It has been recognized as a potential source for recolonization of populations whose survival is at risk. Indeed, several translocations of Dinaric bears to Italy, Austria and France have recently been made. Despite the importance of the Dinaric bear population, its genetic status remains poorly understood. Using tissue samples from 156 hunted or accidentally killed Dinaric bears in Croatia, this study analysed genetic diversity at 12 microsatellite loci, as well as population structure and past reductions in size. In addition, a subset of 59 samples was used to assess diversity of the mitochondrial DNA control region. The results indicate that Dinaric bears have high nuclear genetic diversity, as compared to other extant brown bear populations, despite genetic evidence of a bottleneck caused by past persecutions. However, haplotype diversity was low, probably as a result of male-biased dispersal and female philopatry. Not surprisingly, no evidence of population sub-structure was found using nuclear markers, as the bear habitat has remained continuous and the highway network has been built only recently. Management should focus on maintaining habitat connectivity and keeping the effective population size as large as possible. In addition, when removing individuals, care should be taken not to further deplete the population of rare haplotypes. A coordinated transboundary management of the entire Dinaric-Pindos brown bear population should be a priority for its long-term conservation.  相似文献

The benefits to captive animals of environmental enrichment (EE) are widely recognized. Few studies have, however, studied how to maximise the effect of EE. One issue with EE programs seems to be habituation to the enrichment device. To study the effect of habituation to EE, 14 captive sloth bears (Melursus ursinus) were subjected to two different EE treatments. Treatment one presented EE (logs with honey containing holes) for five consecutive days, whereas treatment two presented EE on intermittent days for five days. Intermittent presentations tended to reduce habituation toward the EE. Both consecutive and intermittent presentations significantly reduced stereotypies; however, the consecutive presentations had a longer‐lasting effect. Explorative behaviors increased in both treatments, consistent with earlier findings that EE increase levels of natural behaviors. Other behaviors were unaffected by the EE presentations. The results show that intermittent presentation of EE objects may secure the interest of the animals, but continuous access to enrichment may be more efficient in reducing stereotypies in the long run. Abstract We developed a snare for collection of black bear (Ursus americanus) hair that obtained a unique hair sample at each snare site, improved the quantity of collected hair compared to barbed-wire corrals, and was easy to deploy over a wide range of topographical features and habitat conditions. This device allowed us to implement intensive sampling methodology needed in mark-recapture experiments with minimal effort. By improving the quantity of hair collected, we also lowered the potential for bear identification errors at the lab. During 2003–2004, bears in 2 study areas triggered snares 1,104 times, which resulted in the collection of 981 hair samples. Of the samples we collected, 79% (775) produced valid genetic data. In 2003, 454 samples identified 79 genetically distinct individuals, and 321 samples identified 86 genetically distinct individuals in 2004. Analysis of capture-recapture data indicated that capture probabilities were affected by heterogeneity among individuals and behavioral responses, but showed little evidence of time effects. Consequently, we used the Pollock and Otto (1983) estimator for model Mbh to estimate abundance with reasonably good precision (CV: 12–14%). Density on the Steamboat and Toketee, Oregon, USA, study areas over the 2-year period averaged 19 bears/100 km2 and 22 bears/100 km2, respectively. Average capture and recapture probabilities over the 2 years of the study were 30% and 63%, respectively, indicating a trap-prone behavioral response. Knowledge of bear densities on the Steamboat and Toketee study areas will enable managers to set hunting quotas, advise land management agencies on habitat issues, and create a baseline database to assist in the long-term monitoring of bear trends in a changing landscape.  相似文献

The welfare of captive bears became a big issue of concern in Poland when a case of a bear being ill-treated became a high-profile case in the media. This case created a challenge to verify, study, and understand the main problems associated with bear keeping so that zoos could significantly improve the conditions in which they keep bears or ensure they keep bears at the minimum required standards. The results presented here are from 1 of the few countrywide studies of captive bear conditions conducted in all the captive institutions in Poland that keep bears. Thirteen institutions kept bears at the time of the study (2007–2009), including 54 individuals of 5 species. Major welfare problems were identified, and the results have been used to challenge zoos to address the changes required and focus the government's attention on areas that require legislative improvement.  相似文献

Initial appraisals of the status of endangered large-mammal populations may have to depend on indices of population trend. Such indices may possibly be improved by using auxiliary variables. Various models were studied for populations of the Florida manatee (Trichechus manatus latirostris), Yellowstone grizzly bear (Ursus arctos horribilis), and Hawaiian monk seal (Monachus schauinslandi). Several criteria for checking validity of the fitted models were considered, and the simple R2 criterion appears to provide useful comparisons. Multiple regression models overestimated the rate of change of the East Coast manatee population as determined from three other sources (a covariance model, a non-linear model, and the rate estimated from reproductive and survival data). A multiple regression model for grizzly bears using three auxiliary variables exhibited a fairly high R2 (0.84) and appeared to provide a better fit than did a non-linear model. A beach count index for Hawaiian monk seals seemed to be unreliable for year-to-year comparisons in contrast to total population counts and estimates from a capture-recapture method. The use of auxiliary variables for checking and improving trend index data appears feasible and well worthwhile.  相似文献
