Delocalized chemiosmotic coupling of oxidative phosphorylation requires that a single-value correlation exists between the extent of Δ\?gmH+ and the kinetic parameters of respiration and ATP synthesis. This expectation was tested experimentally in nigericin-treated plant mitochondria in single combined experiments, in which simultaneously respiration (in State 3 and in State 4) was measured polarographically, FΔψ (which under these conditions was equivalent to Δ\?gmH+) was evaluated potentiometrically from the uptake of tetraphenylphosphonium+ and the rate of phosphorylation was estimated from the transient depolarization of mitochondria during State 4-State 3-State 4 transitions. The steady-state rates of the different biochemical reactions were progressively inhibited by specific inhibitors active with different modalities on various steps of the energy-transducing process: succinate respiration was inhibited competitively with malonate or noncompetitively with antimycin A, or by limiting the rate of transport into the mitochondria of the respiratory substrate with phenylsuccinate; Δ\?gmH+ was dissipated by uncoupling with increasing concentrations of valinomycin; ADP phosphorylation was limited with oligomycin. The results indicate generally that when the rate of respiratory electron flow is decreased, a parallel inhibition of the rate of phosphorylation is also observed, while very limited effects can be detected on the extent of Δ\?gmH+. This behavior is in marked contrast to the effect of uncoupling where the decreased rate of ATP synthesis is clearly due to energy limitation. Extending previous observations in bacterial photosynthesis and in respiration by animal mitochondria and submitochondrial particles the results indicate, therefore, that respiration tightly controls the rate of ATP synthesis, with a mechanism largely independent of Δ\?gmH+. These data cannot be reconciled with a delocalized chemiosmotic coupling model.  相似文献

Bacteria have been isolated from shoot tips of symptomless globe artichoke plants. These were identified as Pseudomonas fluorescens, Pseudomonas marginalis, Pseudomonas spp., Serratia liquefaciens, Enterobacter agglomerans/Erwinia, Agrobacterium radiobacter, an unidentified member of Rhizobiaceae and another classified in the “corynebacteria” group. The most frequently isolated species was P. fluorescens, biovars II and III. The endogenous character of these bacteria was studied in plants growing in vitro and in the open field. P. fluorescens, P. marginalis, S. liquefaciens and E. agglomerans/Erwinia caused symptoms in plants growing in vitro, but only P. fluorescens biovar II and P. marginalis produced symptoms in plants growing in open fields. Differences in pathogenicity were observed on inoculated plants growing in vitro or in the open field. This suggests that several endophytic bacterial species may be responsible for the high levels of contaminants found during the micropropagation of globe artichoke.  相似文献

Artichoke yellow ringspot nepovirus (AYRV) was isolated from stunted broad bean plants in Crete showing leaf mottling and deformation and wrinkling of seed testas. AYRV identification was based on host range, behaviour during purification, electron microscopy and serology. AYRV-broad bean isolate was seed-transmitted in broad bean, and embryo infection appeared to be a prerequisite for virus transmission to seedlings. Transmission through pollen was not detected in broad bean plants, although pollen grains were externally contaminated by the virus. Ovule infection occurred in a high proportion of seed and was independent of pollination.  相似文献

Summary Tracheary element differentiation in cultured explants of pith parenchyma isolated from heads of romaine lettuce (Lactuca sativa L. var. Romana) was strongly inhibited by concentrations of gentamicin sulfate recommended for tissue culture media (50 to 100 μg/ml). Similar results were obtained with cultured explants of Jerusalem artichoke tuber (Helianthus tuberosus L.). Callus formation was suppressed in the presence of increasing levels of gentamicin sulfate in both tissue systems. Plant tissue culture media employed in studies on cell division and xylem differentiation should be supplemented with this antibiotic in concentrations of 10 μg/ml or less according to these results.  相似文献

Epichlorohydrin-triethanolamine (ECTEOLA)-cellulose films (paper and cloth) have been found to bind Saccharomyces cerevisiae cells which were able to develop metabolically active colonies on the surface of the films. Unmodified cellulose films also bound the yeast but to a lesser extent. Film fermenters were constructed by coiling a double layer of the cloth and copper screen and vertically placing the resulting cartridge into a column. These film fermenters were able to convert the sugars (14%) in the hydrolysate of a Jerusalem artichoke tuber into ethanol, with 90% of the theoretical yield after 6 h of fermentation. The bound yeast produced ethanol at a specific rate of 1.0 g ethanol per g cell per hour.  相似文献

The phytochemical composition in two Tunisian globe artichoke cultivars (bracts, leaves, and floral stems) was evaluated in the plant byproducts. The results indicated that the bracts contain the highest levels of total phenols, o‐diphenols, and flavonoids, whereas tannins seem to be more abundant in the leaves. Bracts from the ‘Violet d'Hyères’ cultivar possessed more total phenols (160.8 mg/g DW), flavonoids (64.9 mg/g DW), and anthocyanins (15.3 μg/g DW) than the ‘Blanc d'Oran’ bracts (134.5 mg/g DW, 51.2 mg/g DW, and 8.3 μg/g DW, resp.). Sixty‐four volatile compounds were identified in the headspace of globe artichoke material, particularly in the bracts. The volatile profile showed that sesquiterpene hydrocarbons and non‐terpene derivatives were the main volatiles emitted by the bracts in both cultivars. These results suggest that globe artichoke byproducts might represent a potential source of natural compounds, which could be used as nutraceuticals or as ingredients in the design of functional foods.  相似文献

(Ⅰ)对荨麻科楼梯草属的苞片和小苞片的形态进行了全面研究。(Ⅱ)该属原始群疏伞楼梯草组的雄聚伞花序苞片在每花序为15~90枚,纸质,绿色,狭卵形、狭三角形或条形,长0.5~4 mm,扁平,无任何突起,而与楼梯草族的冷水花属和赤车属的聚伞花序苞片极为相似,因此,上述各种形态可以视为楼梯草属苞片的原始特征,并据此观察到总苞苞片的以下演化趋势:(1)近等大,形状相似,在花序托边缘轮生形成一层→排列为二层,外层2苞片对生,较大,内层苞片较小,形状稍不同;(2)狭卵形,狭三角形或条形→宽卵形或宽三角形,或扁半圆形→由于长度强烈缩小,宽度增大而最终消失;(3)扁平→顶端兜形→船形→船形,顶端突起成细筒;(4)无任何突起↗背面有1龙骨状突起,或有1~6条纵肋或狭翅 ↘顶端具短到长的角状突起→背面顶端之下具角状突起;(5)分生→基部合生→由于长度强烈缩小,宽度强烈增大,总苞苞片合生成一横条形狭片;(6)在数目上,由每花序的7~45枚,一方面增加到50~180枚,另一方面则减少到5枚以下。同时,观察到小苞片形态以下演化趋势:(1)膜质,半透明,白色↗具褐色线纹或呈褐色→呈黑色 ↘薄膜质,透明,无色;(2)扁平→顶端兜形→船形;(3)无任何突起→顶端或在背面顶端之下具角状突起;(4)在数目上,由每花序的7~45枚一方面增加到100或数百枚,甚至达1千到数千枚,另一方面则减少到5枚以下,甚至到0枚。上述演化趋势有助于了解属下各级分类群的演化水平。  相似文献

Ephedra sinica Stapf (commonly, Chinese ephedra) grows mainly in Mongolia, Inner Mongolia, and other regions of northern China. The stems of Chinese ephedra have been called the world’s oldest medicine; other uses remain to be reported. Among the Mongolian people, in addition to medical applications, the fleshy bracts of the seed cones (or “fruit”) are traditionally used as food, fresh or dried, in several ways; for example, the fruit is used as a refined sugar, a cheese condiment, and a tea substitute. In this paper, we analyze the fruit’s nutritional components: moisture, crude protein, crude fat, crude fiber, carbohydrate, ash, pectin, vitamins, fatty acids, mineral elements, and amino acids. Ethnobotanical and nutritional component information may indicate that the plant has potential for development as a special food plant  相似文献

用含有不同浓度(0~400μmol/L)Cd(NO3)2的Hoagland营养液处理砂培的菊芋。处理50d后,测定植物体内镉积累量以及过氧化物酶(POD)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)活性,并对POD同工酶进行电泳分析。发现在Cd50~100μmol/L浓度内,随着镉浓度的升高,菊芋根和叶中镉的积累量显著增加,而随后积累量的增加有所减少。根和叶中MDA含量显著上升,说明镉引起了膜脂过氧化。0~100μmol/LCd处理,根和叶中POD活性随Cd浓度增加而增强,而在200~400μmol/LCd处理下有所减弱。根和叶SOD活性在50~200μmol/LCd处理下随Cd浓度增加而增强,而在400μmol/LCd处理下SOD活性明显受到抑制。根和叶CAT活性随Cd浓度升高而增强。电泳结果显示,POD同工酶变化明显,镉诱导出一条新酶带LP10。菊芋POD同工酶可以作为镉污染的土壤的生物指示剂。  相似文献
