The present study was designed to acquire further understanding of the differences in the immune response of mice orally (OS) or subcutaneously (SS) sensitized to Nematospiroides dubius. Two immunosuppressive agents and skin tests were utilized in this regard.Rabbit antimouse thymus serum (RAMTS) and cyproheptadine (antihistamine-antiserotonin) were similarly effective in suppressing the immune response of subcutaneously sensitized mice. When compared to normal rabbit serum and SS control (sensitized, untreated) animals, observations of the intestinal tunica muscularis in the immunosuppressed SS groups revealed granulomatous lesions in which fewer eosinophils enveloped the sequestered parasite. Cyproheptadine was more successful than the other treatments in interrupting the immune expulsion of N. dubius from orally sensitized mice, but this was only at a borderline significance level. The size and intensity of the active cutaneous anaphylactic skin tests in OS mice injected with an adult or larval antigen, was greater than the response elicited in SS mice or uninfected control mice injected with the same preparations. Similarly, the reaction in subcutaneously sensitized mice exceeded that observed in the noninfected controls.  相似文献

The effects of histamine and an antihistamine on the number and fecundity of adult Trichinella spiralis and on trichinous enteritis measured by determining myeloperoxidase activity in the small intestine of the host on days 7, 9 and 11 postinfection (PI) were examined during primary infections of the CD-1 Swiss white mouse. In mice receiving oral doses of histamine the fecundity of adult worms decreased, the number of adult worms was unaffected and enteritis was elevated above that seen with untreated mice or mice receiving oral doses of saline alone. In mice injected intramuscularly with antihistamine after day 7 PI fecundity of adult worms increased, the number of adult worms remained the same and enteritis decreased compared to untreated mice receiving intramuscular injections of saline. As the concentration of histamine present in the incubation medium (0.01, 0.1, 10, 25, 50 or 100 mg%) was increased above 0.01 mg% the fecundity of adult worms during culture in vitro decreased. As the concentration of antihistamine present in incubation medium (0.01, 0.1, 10 or 100 mg%) was increased above 0.1 mg% the fecundity of adult worms during culture in vitro decreased.  相似文献

Summary Antimuscarinic side-effects, which include dry mouth, tachycardia, thickening of mucus possibly sedation, of the antihistamines limited the usefulness of these drugs. The advent of newer agents has reduced the sedative effect of the antihistamine. The data presented here show that one of the newest antihistamines, desloratadine, and a first generation drug, diphenhydramine, are both competitive inhibitors of muscarinic receptor mediated slowing of the heart as measured using a Langendorff preparation. Both agents have apparent sub-micromolar affinities for the muscarinic receptor. Two other agents, cetirizine and fexofenadine, do not interact with muscarinic receptors in the heart at the concentrations used in this study. Structural similarities of the drugs suggest that substitution of a group with a high dipole moment or charge on the side chain nitrogen decreases the binding with muscarinic receptors. We conclude that of the compounds tested fexofenadine and cetirizine have little or no interaction with muscarinic receptors.  相似文献

Hormonal imprinting usually takes place perinatally at the first encounter between the developing receptor and its target hormone, determining the future binding capacity of the receptor for life. Molecules similar to a hormone can cause faulty imprinting also with life-long consequences. Hormone production of the imprinted cell is also durably influenced. In cytogenic organs imprinting can also be provoked in adulthood. At present the effect of a single terfenadine treatment in adult rats on the histamine content of peritoneal cells (lymphocytes, mast cells and the monocyte-macrophage-granulocyte group), white blood cells (lymphocytes, granulocytes, monocytes) and thymic lymphocytes was studied 3 weeks after treatment to clarify the effect of prolonged treatment with an antihistamine in adulthood. The cells were studied by flow cytometric analysis. Peritoneal mast cells contained significantly more and thymic lymphocytes significantly less histamine than controls. In the other cells the differences were not significant. The results support earlier observations on the effect of antihistamines on mast cell histamine release (inhibition) and call attention to the fact that this effect is durable (hormonal imprinting provoked in adults).  相似文献

Antihistamines, which are commonly used to treat allergic reactions, are known for their side effects, which contribute to weight gain. It is hypothesized that simultaneous Brillouin elastography and Raman spectroscopy can be used to detect changes in adipose tissue associated with a prolonged intake of desloratadine, a commonly used second generation antihistamine. White and brown adipose tissue samples were excised from adult rats following 16 weeks of daily administration of desloratadine. It was found that the prolonged intake of desloratadine leads to an increase in Brillouin shift in both adipose tissue types. Raman spectra indicate that antihistamine use reduces protein-to-lipid ratio in brown adipose tissue but not white adipose tissue, indicating the effect on adipose tissue is location-dependent.  相似文献
