Abstract Colonies of Streptomyces antibioticus were studied by transmission and scanning electron microscopy. The micrographs show that substrate mycelium growth takes place among an intercellular material and this mycelium is covered by a surface film. This structure could be a boundary between the aerial mycelium and the substrate mycelium.  相似文献

Abstract Streptomyces antibioticus strain TÜ 99, from which a wide variety of active compounds had been isolated previously, was reinvestigated using an HPLC photoconductivity screening system. Four new compounds were isolated, characterized and their constitutions determined. All four were α,β-unsaturated γ-lactones; the most abundant compound 3 (C10H16O4), as well as compound 1 (C9H14O4) had a hydroxy group at C(5) of the lactone ring. The four lactones showed antibiotic activity against Pseudomonas aeruginosa and also a weak inhibition of the chitinase from Serratia marcescens .  相似文献

Abstract A cosmid clone from an oleandomycin producer, Streptomyces antibioticus , contains a large open reading frame encoding a type I polyketide synthase subunit and an oleandomycin resistance gene ( oleB ). Sequencing of a 1.4-kb DNA fragment adjacent to oleB revealed the existence of an open reading frame ( oleP ) encoding a protein similar to several cytochrome P450 monooxygenases from different sources, including the products of the eryF and eryK genes from Saccharopolyspora erythraea that participate in erythromycin biosynthesis. The oleP gene was expressed in Escherichia coli as a fusion protein to a maltose-binding protein. Using polyclonal antibodies against this fusion protein it was observed that the synthesis of the cytochrome P450 was in parallel to that of oleandomycin. The cytochrome P450 encoded by the oleP gene could be responsible for the epoxidation of carbon 8 of the oleandomycin lactone ring.  相似文献

Two genes, orf6 and orf9 located in the L-oleandrose sugar biosynthetic gene cluster of Streptomyces antibioticus Tü99. NovU has been characterized as C-5 methyltrnaferase involved in noviose biosynthetic pathway. We have cloned and heterologously expressed the orf6, orf9, and novU genes in S. venezuelae YJ003-OTBP1. This established the function of orf6 and orf9 as 4-ketoreductase and 3-epimerase, respectively. All of analytical data of the noviosylated 10-deoxymethynolide also is in support of proving their functions. Furthermore biosynthetic pathway 5,5-gem-dimethyl-6-deoxyglucose (TDP-Lnoviose) has been proposed.  相似文献

Streptomyces antibioticus tyrosinase was kinetically characterized after purification by PEG-8000/phosphate phase partitioning and ammonium sulfate fractionation using tert-butylcathechol (TBC) and dopamine. The enzyme showed an optimal pH at 6.5 and a KM of 1.2 mM and 8.4 mM, respectively. The effect of several modulators was studied on this Gram-positive bacterium tyrosinase. In addition, previously undescribed characterization of apparent inhibition and activation of a bacterial tyrosinase using different kinds of cyclodextrins was carried out. When a hydrophobic substrate of S. antibioticus tyrosinase, in this case, tert-butylcatechol was used, a marked substrate sequestrant effect was observed in the presence of hydroxypropyl-β-cyclodextrins (OH-β-CDs) and gamma cyclodextrins (γ-CDs). This sequestrant effect was due to the complexation of TBC into the CD cavity. Moreover, the effect of some hydrophobic inhibitors in the presence of OH-β-CDs and γ-CDs was studied using dopamine, a hydrophilic substrate of S. antibioticus tyrosinase. Increasing concentrations of CDs in the presence of inhibitors like hexestrol or hinokitiol, were able to reactivate the inhibited enzyme to reach the non-inhibited level, as a result of the complexation of these inhibitory compounds in the hydrophobic core of the CDs. This dual effect of CDs as apparent inhibitor and activator has never before described being observed in bacteria.  相似文献

Simocyclinones, a novel group of angucyclinone antibiotics, are produced by Streptomyces antibioticus Tü 6040. The compounds show antibacterial and antitumor properties. In submerged cultivation, the production of simocyclinones is strongly dependent on the carbon and nitrogen sources used in a chemically defined medium. Productivity of distinct components and diversity of simocyclinone compounds are influenced by the medium composition. Four series of simocyclinone compounds were detected by high-performance liquid chromatography (HPLC) diode array detector (DAD) and HPLC electrospray ionization (ESI) mass spectrometry (MS) analysis, isolated and the structures determined by nuclear magnetic resonance (NMR) techniques. Under optimized conditions, simocyclinone D8 was produced in an amount of 300 mg l−1 and simocyclinone C4 in a concentration up to 50 mg l−1. Abstract Differences in plaque size of phages developing in Streptomyces antibiotics on several media were mainly determined by their adsorption ability to mycelium obtained at different stages of the bacterial growth-curve. This is due to the exposure of phages to the exocytoplasmic endodeoxyribonuclease produced by this bacterium at the middle-end of the exponential phase.  相似文献

13-1 是从魔芋根际土壤中筛选得到的一株革兰氏阴性生防细菌, 抑菌试验结果表明它对7种植物病原细菌和8种病原真菌具有拮抗作用。形态和常规生理生化特征鉴定, 发现该菌株与溶杆菌属Lysobacter 特征很相近。Biolog 鉴定和16S rDNA基因序列分析表明, 菌株13-1与已报道的抗生素溶杆菌Lysobacter antibioticus DSM 2044(=ATCC29479)同源性高达99%。二者在所建系统发育树中处于同一分枝, 将13-1菌株鉴定为抗生素溶杆菌(L. antibioticus)。  相似文献
