采用反相高效液相色谱法和电喷雾质谱法对我国野生笃斯越橘中花青素组分进行了分离和分析.制备野生越橘花青素的粗提物,经大孔树脂柱层析得到花青素样品,其得率为0.72%.用香草醛法测得其总黄烷醇含量为95%.采用Alltima C18反相色谱柱(250 mm×4.6 mm,5 μm),以2 %甲酸水溶液(A)和2 %甲酸/甲醇溶液(B)作为流动相,分2个梯度对花青素组分进行洗脱,将其主要洗脱峰在电喷雾正离子模式下进行分析,根据质谱的准分子离子[M+H](m/z)检测其聚合度.结果表明,我国野生越橘花青素的提取物被展开成8个主要的洗脱峰,以峰面积计算,主要洗脱峰总含量达85%.经质谱分析,判断为2个单体,3个二聚体,1个三聚体,2个四聚体.结果表明,我国野生笃斯越橘中含有较丰富的花青素,低聚体是其中的主要成分.本文首次报道了我国大兴安岭野生笃斯越橘花青素的组成情况,对于开展其后续的结构、功能、质量检测以及生产开发研究都具有一定的参考价值.  相似文献

使用响应面法优化了紫甘薯花色苷的水解条件.使用高效液相色谱-多波长检测器测定花青素含量.采用3因素3水平Box-Behnken中心组合设计,优化了水解时间、盐酸浓度和水解温度对矢车菊素和芍药素峰面积之和的综合影响.结果显示,紫甘薯花色苷的最佳水浴水解条件为:盐酸浓度4.54 mol/L,水浴温度100℃,水解时间45.8 min.在最优条件下进行验证试验,响应值为理论值的96.5%,说明该响应面法优化方法可行.  相似文献

Kinase Suppressor of Ras (KSR) is a molecular scaffold that interacts with the core kinase components of the ERK cascade, Raf, MEK, ERK to provide spatial and temporal regulation of Ras-dependent ERK cascade signaling. Interruption of this mechanism can have a high influence in inhibiting the downstream signaling of the mutated tyrosine kinase receptor kinase upon ligand binding. Still none of the studies targeted to prevent the binding of Raf, MEK binding on kinase suppressor of RAS. In that perspective the cysteine rich C1 domain of scaffold proteins kinase suppressor of Ras-1 was targeted rather than its ATP binding site with small ligand molecules like flavones and anthocyanidins and analyzed through insilico docking studies. The binding energy evaluation shows the importance of hydroxyl groups at various positions on the flavone and anthocyanidin nucleus. Over all binding interaction shows these ligands occupied the potential sites of cysteine rich C1 domain of scaffold protein KSR.  相似文献

Radiation-induced lung injury (RILI) is a potentially fatal and dose-limiting complication of thoracic radiotherapy. This study was to investigate the protective effects of grape seed pro-anthocyanidins (GSPs), an efficient antioxidant and anti-carcinogenic agent, on RILI. In our study, it was demonstrated that acute and late RILI was ameliorated after GSPs treatment possibly through suppressing TGF-β1/Smad3/Snail signalling pathway and modulating the levels of cytokines (interferon-γ, IL-4 and IL-13) derived from Th1/Th2 cells. In addition, a sustained high level of PGE2 was also maintained by GSPs treatment to limited fibroblast functions. As shown by electron spin resonance spectrometry, GSPs could scavenge hydroxyl radical (•OH) in a dose-dependent manner, which might account for the mitigation of lipid peroxidation and consequent apoptosis of lung cells. In vitro, GSPs radiosensitized lung cancer cell A549 while mitigating radiation injury on normal alveolar epithelial cell RLE-6TN. In conclusion, the results showed that GSPs protects mice from RILI through scavenging free radicals and modulating RILI-associated cytokines, suggesting GSPs as a novel protective agent in RILI.  相似文献

为获取紫背天葵(Cynura bicolor DC.)叶片中花青素种类及其合成调控基因等信息,该试验以紫背天葵叶背面紫色以及经处理叶背面几乎全绿(对照)的叶片为材料,进行转录组测序分析,同时进行6个相关差异表达基因的qRT-PCR分析和6种花青素苷元的HPLC检测,以揭示紫背天葵特有的花青素苷元及其合成调控关键基因信息。结果表明:(1)在紫背天葵中共获得14个花青素苷元及32个花青素合成调控基因信息,其中表达量差异显著下调的4个基因为Pg(c11692)、Cy(c42112)、ANS(c38551)和3GT(c9064),表达量差异显著上调的2个基因是D FR(c35961)和3GT(c20283)。(2)qRT-PCR检测结果显示,上述6个基因在2种紫背天葵叶中的表达趋势(上调或下调)与转录组测序结果完全一致,但转录组测序检测到的表达趋势差异倍数比qRT-PCR检测结果更加明显。(3)HPLC分析显示,紫背天葵叶中均未检测到Dp、Pt、Pn及Mv等4类花青素苷元,但紫背天葵叶中富含Cy花青素苷元,且背面紫色的叶中Cy类花青素苷元含量(62.21 mg/kg)显著高于绿色叶对照(6.86 mg/kg);背面紫色和全绿叶中的Pg花青素苷元含量均低于0.43 mg/kg。研究推测,Cy和Pg花青素苷元在绿叶紫背天葵(对照)中含量显著降低可能是因为存在1个ANS和1个3GT正调控以及1个DFR和1个3GT负调控所致。  相似文献

一品红苞片花色素的分离及初步鉴定   总被引:3,自引:0,他引:3  
用紫外-可见光分光光度计、高效液相色谱(HPLC)和质谱(MS)技术对一品红(Euphorbia pulcherrima)红色苞片中的花色素提取液进行了初步鉴定.一品红花色素的甲醇溶液分别在270、340和520 nm处有3个吸收峰;在440 nm吸光度与可见光最大吸收波长520 nm吸光度的比值为0.29;花色素的甲醇溶液中加入AlCl3后发生红移,再加入HCl后发生蓝移;色素溶液在紫外光下无荧光;色素样品经液相色谱分离后在270 nm检测有5个比较明显的吸收峰;质谱中得到595、611、381、571和589等对应的分子离子峰;花色素酸解液高效液相色谱图谱和鼠李糖、葡萄糖的出峰时间一致.由这些结果可推断一品红花色素样品中主要含有5种组分:矢车菊花色素芸香苷、飞燕草花色素芸香苷、飞燕草花色素苯甲酰基葡糖苷、矢车菊花色素苯甲酰基葡糖苷和一种未知成分.  相似文献
