Tri-1-alkynyltin compounds [R2Sn(CCR1)3 (1), R2 = Me, R1 = Me (a), nBu (b), tBu (c), Me3Si (d), 1-(1-cyclohexenyl) (e); R2 = Et, R1 = Me (a(Et)), nBu (b(Et)), tBu (c(Et)), SiMe3 (d(Et)); R2 = nBu, R1 = Me (a(Bu)), nBu (b(Bu))] were prepared, and their reactivity towards trialkylboranes Et3B (2) and iPr3B (3) in 1,1-organoboration reactions was studied. The first step in each reaction is an intermolecular 1,1-alkytoboration. Afterwards, intramolecular 1,1-vinyloboration or 1,1-alkyloboration compete with further intermolecular 1,1-alkyloboration. Various triorganotin cations (4-7), stabilized by intramolecular side-on coordination to the CC bond of an alkynylborate moiety, were detected as highly fluxional intermediates prior to rearrangement into heterocyclic systems such as stannoles (9-11), 1-stanna-4-bora-2, 5-cyclohexadienes (8, 12). The reactions between 1a or 1a(Bu) and an excess of Et3B (2) afford the tris(alkenyl)tin compounds 13 via threefold intermolecular 1, 1-ethyloboration. 13 rearrange to the 3-stannolenes (14a or 14a(Bu)). The intermediates and final products were characterized by multinuclear one- and two-dimensional 1H, 11B, 13C, 29Si and 119Sn NMR.  相似文献

The reaction between Zn(OAc)2 · 2H2O (1) and the 3-iminoisoindolin-1-ones H2NCNC(O)C6R1R2R3R4 (R1-R4 = H 2; R1, R4 = H, R2, R3 = Cl 3; R1, R3, R4 = H, R2 = Me 4) in EtCN at 70 °C for ca. 12 h affords the novel family of complexes [Zn{H2NCNC(O)C6R1R2R3R4}2(OAc)2] (R1-R4 = H 5; R1, R4 = H, R2, R3 = Cl 6; R1, R3, R4 = H, R2 = Me 7) in excellent (90% and 93% for 5 and 6, correspondingly) to good (64% for 7) yields. The isolated compounds were characterized by elemental analyses (C, H, N), IR, NMR and ESI+-MS. X-ray diffraction data for 2 and 5 indicate that both free (2) and ligated (5) 3-iminoisoindolin-1-ones exist in the zwitterionic form.  相似文献

The incorporation of a carboxylic acid within in a series of 3-amido-4-aryl substituted piperidines (represented by general structure 32) led to the discovery of potent, zwitterionic, renin inhibitors with improved off-target profiles (CYP3A4 time-dependent inhibition and hERG affinity) relative to analogous non-zwitterionic inhibitors of the past (i.e., 3). Strategies to address the oral absorption of these zwitterions are also discussed within.  相似文献

Starting from the efficient hexahydropyridoindole antioxidant stobadine, a series of carboxymethylated tetrahydro- and hexahydropyridoindole derivatives was synthesized and tested for the inhibition of aldose reductase, an enzyme involved in the etiology of diabetic complications. In vitro inhibiton of rat lens aldose reductase was determined by a conventional method. Kinetic analysis of (2-benzyl-2,3,4,5-tetrahydro-1H-pyrido[4,3-b]indole-8-yl)-acetic acid (5b) and (2-phenethyl-2,3,4,5-tetrahydro-1H-pyrido[4,3-b]indole-8-yl)-acetic acid (5c), the most potent compounds in this series with activities in micromolar range, showed uncompetitive inhibition. In addition to the importance of the acidic function, the inhibition efficacy was highly influenced by the steric conformation of the lipophilic aromatic backbone when comparing tetrahydro- and hexahydropyridoindole congeners. Selectivity with respect to the closely related aldehyde reductase was determined by measuring the corresponding inhibitory activities. Antioxidant action of the novel compounds was documented in a DPPH test and in a liposomal membrane model, oxidatively stressed by peroxyl radicals. The presence of a basicity center at the tertiary nitrogen, in addition to the acidic carboxylic function, predisposes these compounds to form double charged zwitterionic species, a characteristic which may remarkably affect their pH-lipophilicity profile. For compounds 5b and 5c, a maximal distribution ratio in a system comprised of 1-octanol/phosphate buffer was recorded near the neutral physiological pH, the region where the isoelectric point lies. Molecular docking simulations into the ALR2 active site performed for the zwitterionic species provided an explanation for the observed structure–activity relationships and the calculated parameters were in agreement with characteristic differences in the stereoelectronic profiles of the tetrahydro- versus hexahydropyridoindoles. ‘Drug-likeness’ of the novel aldose reductase inhibitors was assessed by applying the criteria of Lipinski’s ‘rule of five’.  相似文献
