Summary The pattern of intercellular connections between germ line cells has been studied in follicles of the mutantdicephalic (dic), which possess nurse cell clusters at both poles. Staining of follicles with a fluorescent rhodamine conjugate of phalloidin reveals ring canals and cell membranes and thus allows us to reconstruct the spatial organization of the follicle. Each germ line cell can be identified by the pattern of cell-cell connections which reflect the mitotic history of individual cells in the 16-cell cluster. The results indicate that in both wild-type anddicephalic cystocyte clusters one of the two cells with four ring canals normally becomes the pro-oocyte. However, in some follicles (dicephalic and wild-type) oocytes were found with fewer or more than four ring canals. Indic follicles, one or several nurse cells may become disconnected from the other cells during oocyte growth at stage 9–10. Such disconnected cells cannot later on empty their cytoplasm into the oocyte. This, in turn, might be of consequence for the determination of axial polarity of the embryo. 相似文献

Frequent independent origins of environmental sex determination (ESD) are assumed within amniotes. However, the phylogenetic distribution of sex-determining modes suggests that ESD is likely very ancient and may be homologous across ESD groups. Sex chromosomes are demonstrated to be old and stable in endothermic (mammals and birds) and many ectothermic (non-avian reptiles) lineages, but they are mostly non-homologous between individual amniote lineages. The phylogenetic pattern may be explained by ancestral ESD with multiple transitions to later evolutionary stable genotypic sex determination. It is pointed out here that amniote ESD shares several key aspects with sequential hermaphroditism of fishes such as a lack of sex differences in genomes, biased population sex ratios, and potentially also molecular mechanism related to general stress responses. Here, it is speculated that ESD evolves via a heterochronic shift of the sensitive period of sex change from the adult to the embryonic stage in a hermaphroditic amniote ancestor. In this paper, we describe an efficient procedure for the purification of yeast phosphofructokinase. This procedure eliminates any time delay and enables to obtain an enzyme with minimum proteolytic alterations. The molecular weights of the oligomeric enzyme and of its constitutive subunits were both evaluated by means of several independent methods. However, the accuracy of each measurement was not sufficient to discriminate between an hexameric and an octameric structure of the enzyme oligomer. On the other hand, crosslinking experiments demonstrated the octameric structure of yeast phosphofructokinase. Obviously, some methods of molecular weight determination have led to erroneous results. In particular, our experiments show that the reliability of molecular weight determinations performed by gel filtration of native proteins must be considered with caution. 相似文献

High mountain grasslands offer multiple goods and services to society but are severely threatened by improper land use practices such as abandonment or rapid intensification. In order to reduce abandonment and strengthen the common extensive agricultural practice a sustainable land use management of high mountain grasslands is needed. A spatially detailed yield assessment helps to identify possible meadows or, on the contrary, areas with a low carrying capacity in a region, making it easier to manage these sites. Such assessments are rarely available for remote and inaccessible areas. Remotely sensed vegetation indices are able to provide valuable information on grassland properties. These indices tend, however, to saturate for high biomass. This affects their applicability to assessments of high-yield grasslands.The main aim of this study was to model a spatially explicit grassland yield map and to test whether saturation issues can be tackled by consideration of plant species composition in the modelling process. The high mountain grassland of the subalpine belt (1800 – 2500 m a.s.l.) in the Kazbegi region, Greater Caucasus, Georgia, was chosen as test site for its strong species composition and yield gradients.We first modelled the species composition of the grassland described as metrically scaled gradients in the form of ordination axes by random forest regression. We then derived vegetation indices from Rapid Eye imagery, and topographic variables from a digital elevation model, which we used together with the multispectral bands as predictive variables. For comparison, we performed two yield models, one excluding the species composition maps and one including the species composition map as predictors. Moreover, we performed a third individual model, with species composition as predictors and a split dataset, to produce the final yield map.Three main grassland types were found in the vegetation analysis: Hordeum violaceum-meadows, Gentianella caucasea-grassland and Astragalus captiosus-grassland. The three random forest regression models for the ordination axes explained 64%, 33% and 46% of the variance in species composition. Independent validation of modelled ordination scores against a validation data set resulted in an R2 of 0.64, 0.32 and 0.46 for the first, second and third axes, respectively. The model based on species composition resulted in a R2 = 0.55, whereas the benchmark model showed weaker relationships between yield and the multispectral reflectance, vegetation indices, and topographical parameters (R2 = 0.42). The final random forest yield model used to derive the yield map resulted in 62% variance explained and an R2 = 0.64 between predicted and observed biomass. The results further indicate that high yields are generally difficult to predict with both models.The benefit of including a species composition map as a predictor variable for grassland yield lies in the preservation of ecologically meaningful features, especially the occurrence of high yielding vegetation type of Hordeum violaceum meadows is depicted accurately in the map. Even though we used a gradient based design, sharp boundaries or immediate changes in productivity were visible, especially in small structures such as arable fields or roads (Fig. 6b), making it a valuable tool for sustainable land use management. Floral determination in the terminal bud of the short-day plant Nicotiana tabacum cv. Maryland Mammoth has been investigated. Plants grown continuously in short days flowered after producing 31.4±1.6 (SD) nodes while plants grown continuously in long days did not flower and produced 172.5±9.5 nodes after one year. At various ages, expressed as number of leaves that were at least 1.0 cm in length above the most basal 10-cm leaf, one of three treatments was performed on plants grown from seed in short days: 1) whole plants were shifted from short days to long days, 2) the terminal bud was removed and then rooted and grown in long days, and 3) the terminal bud was removed and then rooted and grown in short days. Whole plants flowered only when shifted from short days to long days at age 15 or later. Only rooted terminal buds from plants at age 15 or older produced plants that flowered when grown in long days. Only terminal buds from plants at age 15 or older that were rooted and grown in short days produced the same number of nodes as they would have produced in their original locations while buds from younger plants produced more nodes than they would have in their original locations. Thus, determination for floral development in the terminal bud, as assayed by rooting, is simultaneous with the commitment to flowering as assayed by shifting whole plants to non-inductive conditions.Abbreviations LD

Summary Phenotypic models of selection are used to determine the effect of facultative parthenogenesis on the production of males in a spatially variable environment when (i) sex determination is under strict genetic control, and (ii) when sex may be environmentally determined. The results show that when sex is under strict genetic control and there is some chance of maturing in isolation, selection favors a female-biased sex ratio. When sex can be environmentally induced by cues which indicate high density, selection favors a mixture of genetic and environmental control, such that half the individuals always become female and the other half become females when isolated and become males when not isolated. 相似文献

Summary Selection in the F3 generation for seed yield, fruiting branches/plant, effective pods/plant, and seed index (100-seed weight) was carried out in two chickpea crosses. Sixty F5 lines (15 lines/selection criterion) along with check variety were evaluated for seed yield in three distinct environments. The effects of selection criteria on yield stability was examined using linear regression approach and genotype-grouping technique. There were no differences between selection criteria for linear yield responses of F5 lines to different environments. Within all four selection criteria the lines showed similar linear responses. The non-linear component was relatively higher for lines selected for effective pods and seed index than lines selected for yield and fruiting branches. On the basis of mean yield and coefficient of variation across environments, the seed index was the least effective selection criterion for developing high yielding and stable lines. When the results of stability parameters and genotype-grouping technique were considered together, selection for yield and fruiting branches was highly effective for isolating stable and high yielding lines. 相似文献
