The epidermal growth factor receptor 1 (EGFR) is overexpressed in various malignancies and is associated with a poor patient prognosis. A small, receptor-specific, high-affinity imaging agent would be a useful tool in diagnosing malignant tumors and in deciding upon treatment and assessing the response to treatment. We describe here the affinity maturation procedure for the generation of Affibody molecules binding with high affinity and specificity to EGFR. A library for affinity maturation was constructed by rerandomization of selected positions after the alignment of first-generation binding variants. New binders were selected with phage display technology, using a single oligonucleotide in a single-library effort, and the best second-generation binders had an approximately 30-fold improvement in affinity (Kd = 5-10 nM) for the soluble extracellular domain of EGFR in biospecific interaction analysis using Biacore. The dissociation equilibrium constant, Kd, was also determined for the Affibody with highest affinity using EGFR-expressing A431 cells in flow cytometric analysis (Kd = 2.8 nM). A retained high specificity for EGFR was verified by a dot blot assay showing staining only of EGFR proteins among a panel of serum proteins and other EGFR family member proteins (HER2, HER3, and HER4). The EGFR-binding Affibody molecules were radiolabeled with indium-111, showing specific binding to EGFR-expressing A431 cells and successful targeting of the A431 tumor xenografts with 4-6% injected activity per gram accumulated in the tumor 4 h postinjection.  相似文献

The influence of various combinations of glycerol and lactose feed on the biosynthesis of two polyketide metabolites, lovastatin and (+)-geodin, by Aspergillus terreus ATCC20542 in a discontinuous fed-batch culture was presented. In these experiments lactose and/or glycerol were also used as the initial carbon substrates in the cultivation media. The application of glycerol feed, when lactose is the initial substrate, leads to the appreciable lovastatin concentration in the broth (122.4 mg l−1), nevertheless the abundant (+)-geodin level is at the same time obtained (255.5 mg l−1). The cultures with glycerol as the initial substrate and fed with lactose produce less lovastatin and (+)-geodin. The application of the various combined glycerol and/or lactose feeds allows for improving lovastatin production up to 161.8 mg l−1 and decreases (+)-geodin concentration to 98.7 mg l−1. The analysis of product formation rates and yield coefficients indicates that lovastatin is more efficiently produced on lactose, especially in the initial stages of the cultivation. Glycerol efficiently sustains fungal activity to form these polyketides in the late idiophase but it mainly favours (+)-geodin formation, if solely used in the feed. The feeds performed both with lactose and glycerol occur to be the most desired to maximise lovastatin and minimise (+)-geodin formation.  相似文献

Suspended BHK 21/13 cells were cultivated in 6M medium supplemented with PVP, bovine serum, LAH, YE, choline chloride and inositol at several concentrations. The maximum working capacity of the bioreactors was 400 ml and the experiments were run for 40 days. The growth promoting effects of each substrate were determined by calculation of generation numbers (n) in each culture. Viability testing and morphological detection of the cells were realized by the trypan blue exclusion method: As a result, the membrane protective effect of PVP, as suggested by some authors, was confirmed and it was estimated that the positive effects of PVP on cell propagation in cultures were due to its protective activities.  相似文献

Human Hikeshi (HsHikeshi) is a nuclear import carrier for Hsp70s and is required for cell survival after heat shock. The Hikeshi homolog in Schizosaccharomyces pombe (SpHikeshi/Opi10) localizes to the nuclear rim, interacts with the Hsp70 homolog Ssa2, and mediates its nuclear import in a reconstituted mammalian nuclear transport system. However, SpHikeshi/Opi10 is not required for heat stress response and survival after heat stress. Instead, SpHikeshi/Opi10 is required for the normal expression of stress response genes under optimal conditions and for cell growth during glucose deprivation. Here, the functions of SpHikeshi/Opi10 are discussed and compared to the functions of HsHikeshi.  相似文献

The effects of triamcinolone acetonide on the pattern of 45calcium efflux were investigated in cultured bone cells. Efflux was measured after equilibrating the cells with 45calcium for 24 hours and a short (3 hours) or a long (24 hours) preincubation with the hormone. The results were analyzed by fitting them to a model of three exponential terms, using a computer program based on the non-linear least square method. As reported previously the results indicated the presence of three exchangeable calcium pools which differ in their rates of calcium exchange. A short preincubation with the hormone (3 hours) caused the following changes: (1) A marked increase in the amount of exchangeable calcium in the “slow turnover” calcium pool (S3) which is probably in the mitochondria. (2) A lesser but significant increase in the amount of calcium in the “fast turnover” calcium pool (S2) which is probably the calcium in the cytosol. (3) An increase in the rates of calcium exchange between S2 and the medium (?20) and between S3 and S2 (?32). All these changes were transient. After 24 hours of preincubation with triamcinolone acetonide the amounts of calcium in S2 and S3 decreased below the control levels and the fluxes were not significantly different from those of the controls.  相似文献

Polyploidy is common in many plant species. Up to date, few studies were reported on photosynthesis and leaf anatomy of tetraploid black locust (Robinia pseudoacacia L.), which has considerable value for agriculture and forest. This study compared photosynthesis and leaf ultrastructure on two black locusts. The values of Pn and Ci in tetraploid were significantly higher than those in the corresponding diploids. Significantly lower stomata dimensions (6.0 μm in length and 2.4 μm in width) and tomatal density were observed in 4×. Leaf trichome density was statistically different between 2× and 4×. However, no substantial difference in the ultrastructure of chloroplasts and mitochondria between diploid and tetraploid was observed under any stress. These ultrastructural characteristics may contribute to tetraploid a better protection mechanism than diploid.  相似文献

Calcium efflux patterns were investigated in two clonal osteosarcoma cell lines, ROS 172 and ROS 23. Efflux was measured after equilibrating the cells with 45calcium in medium containing 1% or 10% serum. The results of 45calcium efflux were analyzed by fitting them to a model of three exponential terms, using a computer program based on the non-linear least square method. The results indicated the presence of three exchangeable calcium pools which differ in their rate constants of calcium efflux: a “very fast turnover” (S1) a “fast turnover” (S2) and a “slow turnover” (S3). Washing the labelled cells with sucrose solutions, at pH 7.8 and pH 4, removed most of the calcium localized in S1, indicating that this calcium is membrane bound. The parameters of calcium efflux in ROS cells were found to be different from those measured previously in cultured bone cells: (1) There was no difference between efflux patterns in cells incubated in medium containing 1% and 10% serum; (2) Rates of calcium fluxes were much lower in ROS cells than those in bone cells; and (3) The amount of calcium in S3 was very small.  相似文献
