The first-year zoology course at University College Cardiff is based on a self-instructional system using printed texts, audiovisual programmes, and integrated practical work. Each course ‘Unit’ has in addition a ‘General Session’ in the lecture theatre and structured tutorial work; some Units have associated fieldwork. The students work in a specially designed teaching laboratory and proceed at their own preferred time and pace. Each aspect of the course and its organization is described, with emphasis on the modifications to the original scheme which have been introduced during its first three years of existence in the light of experience.  相似文献

Little information is available on the variability of the dynamics of the actual and observed root respiration rate in relation to abiotic factors. In this study, we describe I) interactions between soil CO2 concentration, temperature, soil water content and root respiration, and II) the effect of short-term fluctuations of these three environmental factors on the relation between actual and observed root respiration rates. We designed an automated, open, gas-exchange system that allows continuous measurements on 12 chambers with intact roots in soil. By using three distinct chamber designs with each a different path for the air flow, we were able to measure root respiration over a 50-fold range of soil CO2 concentrations (400 to 25000 ppm) and to separate the effect of irrigation on observed vs. actual root respiration rate. All respiration measurements were made on one-year-old citrus seedlings in sterilized sandy soil with minimal organic material.Root respiration was strongly affected by diurnal fluctuations in temperature (Q10 = 2), which agrees well with the literature. In contrast to earlier findings for Douglas-fir (Qi et al., 1994), root respiration rates of citrus were not affected by soil CO2 concentrations (400 to 25000 ppm CO2; pH around 6). Soil CO2 was strongly affected by soil water content but not by respiration measurements, unless the air flow for root respiration measurements was directed through the soil. The latter method of measuring root respiration reduced soil CO2 concentration to that of incoming air. Irrigation caused a temporary reduction in CO2 diffusion, decreasing the observed respiration rates obtained by techniques that depended on diffusion. This apparent drop in respiration rate did not occur if the air flow was directed through the soil. Our dynamic data are used to indicate the optimal method of measuring root respiration in soil, in relation to the objectives and limitations of the experimental conditions.  相似文献

Two species of Melissa are currently present in Sardinia: Melissa officinalis L. and Melissa romana Mill . Our research can only count on a few supported evidences (as reported in Flora Italiana and Moris) and some notes on new stations in Sardinia that give us some information about morphology, distribution, bioecological, and ethnobotanical characteristics of both species. In this paper, we present the results of the research about morphological aspects of Mromana vs. Mofficinalis, and their essential oils in different stations at different phenological periods. Moreover, we compared the essential oil of Mromana with the one obtained from Mofficinalis growing in the few naturalized stations still present in Sardinia. The most evident morphological differences between the two entities are the long‐stalked capitate glandular trichomes, shorts and inclined capitate trichomes, and peltate hairs. The chemical composition of essential oil presents several significant differences between the species. In fact, oils show that in none of the phenological stages, M. romana recalls in its composition M. officinalis. Major distinctions are also evident between dry and fresh plants, and among essential oils distilled in different seasons.  相似文献

Response surface methodology was used to optimize pectin recovery from lemon by-product using an acidified date juice as extraction solution. When enriched in pectin, this latter can be useful for preparation of date-lemon jelly. The effects of three parameters namely temperature, pH and extraction time, on pectin extraction were studied. The fitted mathematical model allowed us to plot response surfaces as well as isoresponse curves and to determine optimal extraction conditions. Results clearly indicated that the temperature was the main factor influencing the pectin yield which increased with temperature and time or decreasing pH. The selected optimal conditions were: temperature 84.34 °C; extraction time 3 h 34 min and pH 2.8. These conditions yielded about 11.21% of pectin versus 10.89% for the predicted value.  相似文献

Estimates of root and soil respiration are becoming increasingly important in agricultural and ecological research, but there is little understanding how soil texture and water content may affect these estimates. We examined the effects of soil texture on (i) estimated rates of root and soil respiration and (ii) soil CO2 concentrations, during cycles of soil wetting and drying in the citrus rootstock, Volkamer lemon (Citrus volkameriana Tan. and Pasq.). Plants were grown in soil columns filled with three different soil mixtures varying in their sand, silt and clay content. Root and soil respiration rates, soil water content, plant water uptake and soil CO2 concentrations were measured and dynamic relationships among these variables were developed for each soil texture treatment. We found that although the different soil textures differed in their plant-soil water relations characteristics, plant growth was only slightly affected. Root and soil respiration rates were similar under most soil moisture conditions for soils varying widely in percentages of sand, silt and clay. Only following irrigation did CO2 efflux from the soil surface vary among soils. That is, efflux of CO2 from the soil surface was much more restricted after watering (therefore rendering any respiration measurements inaccurate) in finer textured soils than in sandy soils because of reduced porosity in the finer textured soils. Accordingly, CO2 reached and maintained the highest concentrations in finer textured soils (> 40 mmol CO2 mol−1). This study revealed that changes in soil moisture can affect interpretations of root and soil measurements based on CO2 efflux, particularly in fine textured soils. The implications of the present findings for field soil CO2 flux measurements are discussed. This revised version was published online in June 2006 with corrections to the Cover Date.  相似文献

The waters around Bimini (25° 43.70′ N, 79° 18.00′ W) provide an ideal nursery location for juvenile lemon sharks, Negaprion brevirostris, but this habitat is threatened by the development of a large resort. Since 1999 the North Sound (NS) has been subjected to intermittent periods of dredging, the most intensive of which was in March 2001. Possible effects from the development up to June 2006 were investigated by: comparing growth rates of juvenile lemon sharks in the NS, Sharkland (SL) and South Bimini (SB) nurseries between 1995-2005 using before-after, control-impact (BACI) analysis; analyzing survival of juvenile lemon sharks in the NS and SL between 1995–2006; and by comparing habitat structures in the NS and SB nurseries in 2003 and 2005. BACI analysis detected no statistically significant difference between the growth rates of juvenile lemon sharks in the three nurseries before and after the impact date of March 2001. However, a reduction in the survival rate of juvenile lemon sharks in the NS after March 2001 was statistically significant, including a 23.5% decline in first-year survival. Habitat structure of the NS in 2003 and 2005 also varied with the mean percentage cover of the seagrass Thalassia testudinum declining by 17.7% since 2003. Our results indicate a correlation between the development thus far and a decline in the survival rates of juvenile lemon sharks and changes in the habitat structure of the NS. To elucidate further information regarding potential effects of the resort development on juvenile lemon sharks in the NS nursery, we suggest several future research directions.  相似文献

In the field in south-central Sweden, we tested by randomised, standardised methodology the potential anti-tick repellent activity of two concentrations of MyggA Natural spray (containing PMD) (4.2 and 3.2 g/m2) and one of RB86 (with 70% neem oil containing azadirachtin) (3 g/m2) to host seeking nymphs of Ixodes ricinus. Each substance was applied separately to 1 m2 cotton flannel cloths. Nymphal ticks on the cloths, pulled over the vegetation, were recorded at 10-m stops. Nymphal numbers recorded differed significantly between treated cloths [4.2 or 3.2 g MyggA Natural spray/m2 and 3 g RB86/m2] and the untreated control (df = 3, chi2 = 112.74, P < 0.0001). Nymphal numbers also differed significantly among collectors (df = 3, chi2 = 15.80, P < 0.001). Repellency of treated cloths, i.e., 4.2 or 3.2 g MyggA Natural spray/m2 and 3 g RB 86/m2 declined from day 0 (i.e. the day of impregnation) to day 3 after impregnation from 77 to 24%, 58 to 16% and 47 to 0.5%, respectively. This study suggests that all three treatments have significant repellent activities against I. ricinus nymphs.  相似文献

Lemon balm (Melissa officinalis L.; Lamiaceae) is a well-known medicinal plant mainly due to two groups of compounds, the essential oil and the phenylpropanoid derivatives. The prominent phenolic compound is rosmarinic acid (RA), an ester of caffeic acid and 3,4-dihydroxyphenyllactic acid. RA shows a number of interesting biological activities. Rosmarinic acid synthase (RAS; 4-coumaroyl-CoA:hydroxyphenyllactic acid hydroxycinnamoyltransferase) catalyses the ester formation. Cell cultures of M. officinalis have been established in order to characterise the formation of RA in an important diploid medicinal plant. RAS activity as well as the expression of the RAS gene are closely correlated with the accumulation of RA in suspension cultures of M. officinalis. The RAS cDNA and gene (MoRAS) were isolated. The RAS gene was shown to be intron-free. MoRAS belongs to the BAHD superfamily of acyltransferases. Southern-blot analysis suggests the presence of only one RAS gene copy in the M. officinalis genome. The enzyme was characterised with respect to enzyme properties, substrate preferences and kinetic data in crude plant extracts and as heterologously synthesised protein from Escherichia coli.  相似文献

目的观察柠檬提取物对细菌生物膜的消除作用,对柠檬提取物抑制多重耐药金黄色葡萄球菌的机制进行初步研究,以及对环境中富集的病毒消除作用的研究。方法采用高分子滤膜法制备金黄色葡萄球菌生物膜后,加入柠檬提取物分别作用1、2和3h,通过细菌菌落计数判定不同生物膜的细菌存活数;采用SDS—PAGE电泳法,观察在柠檬提取物作用下菌体蛋白质的变化;EcoRⅠ酶切金黄色葡萄球菌DNA,观察酶切图谱的改变;在低渗环境下动态观察柠檬提取物在不同作用时间对细菌细胞壁的影响;通过柠檬提取物对病毒作用后RNA量的变化,说明柠檬提取物具有杀灭病毒的作用。结果柠檬提取物对生物膜中的细菌具有杀菌作用,且随作用时间的延长而逐渐增强;柠檬提取物对细菌蛋白质的组成和表达量均有一定影响,同时细菌DNA的EcoRⅠ酶切图谱发生改变;柠檬提取物作用15min时细菌呈现明显的胞壁缺损现象,即细菌个体胀大,呈现大球形;经柠檬提取物作用后,RNA病毒被杀灭。结论结果显示柠檬提取物对耐药金黄色葡萄球菌所形成的生物膜具有明显抑制作用;在柠檬提取物作用下,菌体蛋白合成量发生改变,同时对细菌的DNA和细胞壁有明显的影响。柠檬提取物处理病毒悬液后,病毒RNA量显著减少,说明病毒RNA结构被破坏。  相似文献

Because fine roots tend to be concentrated at the soil surface, exposure to dry surface soil can have a large influence on patterns of root growth, death and respiration. We studied the effects of arbuscular mycorrhizas (AM) formation on specific root length (SRL), respiration and mortality of fine roots of bearing red grapefruit (Citrus paradisi Macf.) trees on Volkamer lemon (C. volkameriana Tan. & Pasq.) rootstock exposed to drying soil. For each tree, the fine roots were removed from two woody lateral roots, the roots were surface sterilized and then each woody root was placed in a separate pair of vertically divided and independently irrigated soil compartments. The two split-pot systems were filled with sterilized soil and one was inoculated with arbuscular mycorrhizal fungi (Glomus etunicatum/G. intraradices). New fine lateral roots that emerged from the woody laterals were permitted to grow inside the pots over a 10-month period. Irrigation was then removed from the top compartment for a 15-week period. At the end of the study, roots inoculated with AM fungi exhibited about 20% incidence of AM formation, whereas the uninoculated roots were completely void of AM fungi. Arbuscular mycorrhizal roots exhibited lower SRL, lower root/soil respiration and about 10% lower fine root mortality than nonmycorrhizal roots after 15 weeks of exposure to dry surface soil. This study demonstrates the feasibility of examining mycorrhizal effects on the fine roots of adult trees in the field using simple inexpensive methods.  相似文献
