Summary The occurrence of plasmodesmata in the graft interfaces of two heteroplastic grafts (Impatiens walleriana onImpatiens olivieri andHelianthus annum onVicia faba) has been studied. For both systems two types of intercellular strand are described: 1. Continuous plasmodesmata interconnecting the cells of stock and scion and 2. half plasmodesmata traversing the wall part of one partner cell without connection to the abutting cell. Single strands or branched forms occur in both types of plasmodesma. In the case of half plasmodesmata, branchings with extended median nodules predominate. The distribution of half and continuous plasmodesmata varies with the different areas of a graft interface: in the region of bridging vascular tissues most cell connections are continuous. In areas where cortex or pith-derived callus cells and those of misaligned tissues (cortex/vascular tissue; cortex/pith; pith/vascular tissue) match, discontinuous strands predominate.Branched half plasmodesmata also occur in presumably fused walls between related callus cells; they are typical structures secondarily formed in non-division walls.The results are discussed with regard to compatibility/incompatibility phenomena in heterografts and the development and function of interspecific cell bridges.  相似文献

Summary Leydig cells in testes of adult rats were selectively destroyed by a single intraperitoneal injection of ethane dimethane sulphonate. Four days later rats were made unilaterally cryptorchid and 1, 2 and 4 weeks later the histology of the testes was examined by light microscopy and morphometry. After induction of unilateral cryptorchidism, the volume of abdominal compared to scrotal testes was reduced by 45–60% due to rapid impairment of spermatogenesis in abdominal testes. Leydig cells were not present in either scrotal or abdominal testes in the 1-week unilateral crytorchid group. A new generation of foetal-type Leydig cells was observed in scrotal testes of the 2-week unilateral crytorchid group although their total volume per testis estimated by morphometry, was small, being approximately 1 l. In contrast, the abdominal testis exhibited a remarkable proliferation of foetal-type Leydig cells (total volume per testis, 16 l) which predominantly surrounded the peritubular tissues of the seminiferous tubules. A similar morphology and pattern of Leydig cell development was observed in scrotal and abdominal testes of the 4-week unilateral cryptorchid group where total Leydig cell volume was 7 l vs 21 l, respectively. The results show that regeneration of a new population of Leydig cells occurs more rapidly in the abdominal testis than in the scrotal testis of the same animal. These observations suggest the possibility that augmentation of Leydig cell growth is mediated by local intratesticular stimulatory factors within the abdominal testis. Development of new Leydig cells from the peritubular tissue provides circumstantial evidence that the seminiferous tubules and in particular the Sertoli cells, are a likely source of agents that stimulate the growth of Leydig cells.  相似文献

Zusammenfassung Der Verlauf der Sehbahn und die Lokalisation der optischen Zentren wurden bei Zonotrichia leucophrys gambelii (nordamerikanischer Ammernfink) nach einseitiger Augenexstirpation mit den Techniken von Nauta-Fink-Heimer, Bodian und Bielschowsky erforscht. Die Untersuchungen erstreckten sich über einen Zeitraum von 3 bis zu 120 Tagen nach der Operation. Zonotrichia leucophrys gambelii besitzt ein für Vögel typisches visuelles System. Die Hauptmasse der Optikusfasern endet im Stratum griseum et fibrosum superficiale des Tectum opticum. Weitere zentrale Endgebiete sind: Nucleus geniculatus lateralis, Nucleus lateralis anterior, Nucleus superficialis synencephali, Nucleus externus, tectales Grau und Nucleus ectomamillaris als Kern der basalen optischen Wurzel. Alle Fasern werden im Chiasma opticum total gekreuzt, auch der Tractus isthmo-opticus, ein efferentes Bündel, dessen Ursprung im Nucleus isthmo-opticus zu finden ist. Dieses efferente Fasersystem läßt sich im Stumpf des durchtrennten N. opticus noch 120 Tage nach der Operation gut versilbern. Eine direkte Verbindung von Retina und Hypothalamus war lichtmikroskopisch nicht nachweisbar. Neurosekretorisch aktive Zellen des Hypothalamus können zwar einen engen räumlichen Kontakt mit den optischen Fasern haben, Synapsen sind aber an diesen Stellen nicht zu erkennen. Es werden passagere Opticusfasern beschrieben, die auf dem Weg zum Nucleus lateralis anterior und Nucleus superficialis synencephali den Hypothalamus durchsetzen.
Neurohistological and experimental studies of the visual system in Zonotrichia leucophrys gambelii
Summary The course of the optic pathways and the positions of the optic centers have been investigated with unilaterally enucleated white-crowned sparrows, Zonotrichia leucophrys gambelii, using the techniques of Nauta-Fink-Heimer, Bodian, and Bielachowsky. The investigation involved birds examined 3–120 days after enucleation. The white-crowned sparrow has a typically avian visual system. The major bundles of optic fibers terminate in the stratum griseum et fibrosum superficiale of the tectum opticum. Further terminal areas are the nucleus geniculatus lateralis, nucleus lateralis anterior, nucleus superficialis synencephali, nucleus externus, the tectal gray, and the nucleus ectomamillaris of the basal optic root. There is a complete crossing of all fibers in the chiasma, including those of the tractus isthmo-opticus, an efferent bundle with its origin in the nucleus isthmo-opticus. This efferent fiber system can be well impregnated in the stump of the sectioned optic nerve up to 120 days after the operation. No direct connection between the retina and hypothalamus could be demonstrated by light microscopy. Hypothalamic neurosecretory cells can occur in close contact with optic fibers but no synapses have been recognized. Some optic fibers pass through the hypothalamus enroute to the nucleus lateralis anterior and the nucleus superficialis synencephali.

Summary The phenomenon of interspecific incompatibility between various wild tuber-bearing and closely related non-tuber-bearing Solanum species was studied. One area of investigation included an examination of possible protein interactions in the incompatibility reaction using SDS electrophoresis. Pollen tube inhibition and morphology were examined in conjunction with biochemical analysis. Two sets of crosses were examined: interspecific tuber-bearing species crosses and interspecific tuber-bearing × non-tuber-bearing species crosses. These crosses had consistent pollen tube inhibition in the upper one-third of the style. The upper third of the styles of incompatibly pollinated, compatibly pollinated, and unpollinated styles was studied under fluorescence microscopy to observe pollen tube growth and morphology. Interspecific tuber-bearing × non-tuber-bearing species crosses demonstrated consistent pollen tube inhibition just below the stigma with frequent pollen tube swelling and bursting and extensive callose deposition along the pollen tube wall. Interspecific tuber-bearing species crosses had pollen tube inhibition further down the style with pollen tube tip tapering and extensive callose deposition. Stylar proteins of the lower two-thirds of the styles were analyzed with SDS electrophoresis. No unique protein differences were found to be specifically associated with the interspecific incompatibility reaction in this portion of the style.Cooperative investigation of the U.S. Department of Agriculture, Agricultural Research Service, and the Wisconsin Experiment Station. The influence of different pollen donors on seed formation was investigated in three populations ofEspeletia schultzii that differ in environmental conditions and life history characteristics. Self pollen and pollen from different donors (< 15m apart) within each population was used in a diallel design in order to test the genetic base of seed set variation. Three measures of seed formation were used: (1) achene number; (2) proportion of filled achenes (fruits) that distinguishes between achenes with seeds and empty achenes; (3) proportion of aborted seeds that distinguishes between viable and aborted seeds. Self-pollinations resulted in empty achenes. Achene number did not vary between the different pollen donors. A bimodal pattern of filled achenes was found in two populations in two consecutive years. On the other hand, a unimodal pattern was found in crosses between more distant donors (> 30m). These patterns seems to be the results of a sporophytic incompatibility system. Seed abortion was highest at the higher elevations and seems to be correlated with elevation rather than with any genetic effect.  相似文献

The shoot growth of compatible ( Prunus persica L. Batsch cv. Springtime grafted on Prunus cerasifera L. Ehrh cv. Myrobolan P2032) and incompatible ( Prunus persica L. Batsch cv. Springtime grafted on Prunus cerasifera L. Ehrh cv. Myrobolan P18) peach/plum grafts was observed over a period of 100 days after grafting under controlled conditions. Leaf and root activities were determined by studying carbon assimilation and partitioning, leaf mineral contents and water relations. Shoot length and leaf number were not significantly affected in the incompatible combination during the first 55 days after grafting, but then, shoot growth rate was significantly reduced. Final total dry weights of the shoot were similar in both graft combinations. The incompatible combination did not show any water stress. Soluble sugar and starch contents increased in the leaves of the incompatible combination, accounting for about 36% of the increase of leaf dry weight per unit area. Photosynthesis was affected by the compatibility of the grafts. Leaf nitrogen content (% dry weight) fell in the incompatible graft combination 65 days after grafting. However, nitrogen content on an area basis was not affected. The possibility of nitrogen stress is discussed.  相似文献

Inbreeding effects and incompatibility relationships were examined in strains of the egg parasitoid Trichogramma nr brassicae (Hymenoptera: Trichogrammatidae) from southeastern Australia. Crosses between strains provided weak evidence of incompatibility in a few cases. However sex ratio in crosses within strains tended to be more female-biased than in crosses between strains. Inbreeding was imposed for four generations (F>0.59) of sib mating. The fitness of inbred strains was compared to that of outbred strains generated by crossing the inbred strains. No effects of inbreeding were found for any of the four female traits examined (fecundity, body length, head width and hind tibia length), indicating that T. nr. brassicae is not subjected to inbreeding depression. Inbreeding effects were also not found for male mating success as expected for the haploid sex. There were differences among strains for all traits apart from fecundity, indicating heritable variation. Strain differences for fitness measures were uncorrelated with wasp size. The potential use of inbreeding in the quality control of Trichogramma for mass-release is discussed. Inbreeding may be a useful tool in minimising the effects of laboratory adaptation, thereby extending the useful life of a strain.  相似文献

Paternal genome loss (PGL) during early embryogenesis is caused by two different genetic elements in the parasitoid wasp, Nasonia vitripennis. Paternal sex ratio (PSR) is a paternally inherited supernumerary chromosome that disrupts condensation of the paternal chromosomes by the first mitotic division of fertilized eggs. Bacteria belonging to the genus Wolbachia are present in Nasonia eggs and also disrupt paternal chromosome condensation in crosses between cytoplasmically incompatible strains. Cytoplasmic incompatibility Wolbachia are widespread in insects, whereas PSR is specific to this wasp. PGL results in production of male progeny in Nasonia due to haplodiploid sex determination. The cytological events associated with PGL induced by the PSR chromosome and by Wolbachia were compared by fluorescent light microscopy using the fluorochrome Hoescht 33258. Cytological examination of eggs fertilized with PSR-bearing sperm revealed that a dense paternal chromatin mass forms prior to the first metaphase. Quantification of chromatin by epifluorescence indicates that this mass does undergo replication along with the maternal chromatin prior to the first mitotic division but does not replicate during later mitotic cycles. Contrary to previous reports using other staining methods, the paternal chromatin mass remains condensed during interphase and persists over subsequent mitotic cycles, at least until formation of the syncytial blastoderm and cellularization, at which time it remains near the center of the egg with the yolk nuclei. Wolbachia-induced PGL shows several marked differences. Most notable is that the paternal chromatin mass is more diffuse and tends to be fragmented during the first mitotic division, with portions becoming associated with the daughter nuclei. Nuclei containing portions of the paternal chromatin mass appear to be delayed in subsequent mitotic divisions relative to nuclei free of paternal chromatin. Crosses combining incompatibility with PSR were cytologically similar to Wolbachia-induced PGL, although shearing of the paternal chromatin mass was reduced. Wolbachia may, therefore, block an earlier stage of paternal chromatin processing in the fertilized eggs than does PSR. © 1995 Wiley-Liss, Inc.  相似文献
