Exposing Pseudomonas aeruginosa biofilm grown on the inner surface of Teflon and silicone tubes to UVC light (265 nm) from light emitting diodes (LED) has previously been shown to substantially reduce biofilm growth. Smaller UVC fluencies were required to disinfect Teflon tubes compared to silicone tubes. Light propagation enhancement in tubes can be obtained if the refractive index of the intra-luminal saline solution is higher than that of the polymer. This condition is achieved by using Teflon tubes with a low refractive index (1.34) instead of the polymers with a high refractive index (1.40–1.50) normally used for tubing in catheter production. Determining whether or not UVC light exposure can disinfect and maintain the intra-luminal number of colony forming units (CFUs) at an exceedingly low level and thus avoid the growth and establishment of biofilm is of interest. The use of UVC diodes is demonstrated to be a preventative disinfection treatment on tubes made of Teflon, which enhances the UVC light propagation, and on tubes made of a softer material, ethylene vinyl acetate (EVA), which is suitable for catheters but much less suitable for UVC light propagation. Simulating an aseptic breach (~103–104 CFU ml?1), the UVC disinfection set-up was demonstrated using tubes contaminated with planktonic P. aeruginosa. After the tubes (10–20 cm) were inoculated with the bacterial solution for 3 h, they were emptied and filled with saline solutions (0.9–20%). Next UVC fluencies (0–21 mJ cm?2) were applied to the tubes 3 h after inoculation. Colony counts were carried out on liquid samples drawn from the tubes the first day after UVC treatment and liquid and surface samples were collected and analyzed 3–4 days later. A fluence of approximately 1.0 mJ cm?2 was noted as being sufficient for no growth for a period of 3–4 days for the Teflon tubes. Determining the fluence threshold for the EVA tubes was not possible. Almost all of the UVC-treated EVA tubes were disinfected simply by filling the tubes with a saline solution. Direct UVC treatment of the contaminated EVA tubes revealed, however, that a fluence of 21 mJ cm?2 killed the bacteria present in the tubes and kept them disinfected for a period of 3–4 days.  相似文献

Algal proliferation has commonly been reported to occur on monuments, such as crypts, churches, and caves, as soon as artificial lighting is used. In this work we study the effects of UV-C irradiation on algae collected in different caves in Dordogne (southwest of France). First, the effect of UV-C irradiation was tested on algal cell suspensions during increasing exposure times. After treatment, the photosynthetic capacity was assayed using a polarometric method, and algal cell viability was then estimated using a Trypan blue test after a rest period of 15 h. UV-C irradiation was then studied on algal cells cultivated on a solid support consisting of pieces of calcareous stone. Drops of concentrated algal cells were inoculated on stone and exposed to UV-C radiation for 3, 6, or 9 h. After this irradiation, half of the samples were submitted to a high white light intensity (1400 ??mol m−2 s−1 of photosynthetically active radiation, PAR) for 6 h while the other half were incubated in the culture room. Subsequently, algal macroscopic parameters such as covering rate and colonized area were measured by macro photography. Both experiments led to the conclusion that UV-C irradiation has deleterious effects on photosynthetic parameters and growth of algal cells.  相似文献

Hermatypic (reef building) corals live in an environment characterized by high ambient levels of photosynthetically active radiation (PAR) and ultraviolet radiation (UVR). Photoadaptive mechanisms have evolved to protect the sensitive cell structures of the host coral and their photosynthetic, endosymbiotic zooxanthellae. Environmental stressors may destabilize the coral-zooxanthellae system resulting in the expulsion of zooxanthellae and/or loss of photosynthetic pigment within zooxanthellae, causing a condition known as bleaching. It is estimated that 1% of the world’s coral population is lost yearly, partly due to bleaching. Despite intensive research efforts, a single unified mechanism cannot explain this phenomenon. Although UVA and UVB cellular damage is well documented, UVC damage is rarely reported due to its almost complete absorption in the stratosphere. A small scale coral propagation system at the University of Maine was accidentally exposed to 15.5 h of UVC radiation (253.7 nm) from a G15T8 germicidal lamp, resulting in a cumulative surface irradiance of 8.39 × 104 J m−2. An experiment was designed to monitor the progression of UVC induced damage. Branch sections from affected scleractinian corals, Acropora yongei and Acropora formosa were submitted to histopathology to provide an historical record of tissue response. The death of gastrodermal cells and necrosis resulted in the release of intracellular zooxanthellae into the gastrovascular canals. Zooxanthellae were also injured as evidenced by pale coloration, increased vacuolization and loss of membrane integrity. The recovery of damaged coral tissue likely proceeds by re-epithelialization and zooxanthellae repopulation of gastrodermal cells by adjacent healthy tissue.  相似文献

It is an intriguing problem whether heat shock proteins (HSPs) play a protective role in UVC-induced cell death in human cells, and the problem has not been solved. To search for the HSPs involved in UVC resistance, gene expression profiles using cDNA array were compared between UVC-sensitive human RSa cells and their UVC-resistant variant AP(r)-1 cells. The expression levels of heat shock protein 27 (HSP27) were lower in RSa cells than in AP(r)-1 cells. RSa cells transfected with sense HSP27 cDNA showed slightly lower sensitivity to UVC-induced cell death than the control cells transfected with a vector alone and much lower sensitivity than RSa cells transfected with the antisense HSP27 cDNA. Furthermore, the removal capacities of the two major types of UVC-damaged DNA (thymine dimers and (6-4)photoproducts) in the cells with the up-regulation of HSP27 were moderately elevated compared with those in the control cells, while those in the cells with down-regulation were remarkably suppressed. These results suggest that HSP27 is involved in the UVC-resistance of human cells, at least those tested, possibly via functioning in nucleotide excision repair.  相似文献

Exposure of insects and higher orders of animals to UVC radiation has been shown to result in toxicity with a delayed expression. We have observed an immediate writhing response in slowly wandering third instar larvae of Drosophila melanogaster exposed to UVC radiation at doses that were not lethal. UVB and UVA radiation had no effect. Mutants for the visual and sensory systems appeared to respond normally, but CO2 anesthesia resulted in reversible inhibition, ebony and silver mutants, which affect different pathways in catecholamine metabolism, showed an absent to reduced response. Using UVC lasers, we were able to demonstrate a response in different regions of the larval body. Furthermore, a 193 nm laser that penetrates only 2–5 μm was able to induce a response but unable to kill the larvae. These results suggest a photochemical reaction occurs in the cuticle which produces free radicals that stimulate the nerves and muscle which are present immediately below the epidermis. Possible targets for the UVC radiation are catecholic compounds secreted and processed into the cuticle of third instar larvae just prior to pupariation whose primary function is to crosslink the protein and carbohydrate components. © 1996 Wiley-Liss, Inc.  相似文献

Abstract

A two-part study was designed to investigate the efficacy of using UVC to prevent biofouling in the context of ship hull coatings. The first study determined the frequency of UVC required for a coating that does not have any additives (epoxy). It was found that 1?min/day was effective at preventing hard fouling but not biofilm development. The second study addressed several variables: coating type (epoxy, copper, fouling release), frequency of UVC (no exposure, continuous exposure, 1min/6h, 1?min/day), and distance from the lamp (25 and 50?mm). Continuous UVC exposure resulted in no biofouling settlement but it did damage the copper coating. Intermittent UVC exposure was effective at preventing biofouling recruitment to both the copper and the fouling release coatings. Variations were observed with regards to the fouling composition, especially biofilms, sedimentary tubeworms and barnacles, suggesting tolerances within the community.  相似文献

Effects of fishing on marine communities are becoming increasingly evident, yet little is known of the spatial extent of impacts, particularly for multiple impacts distributed over broad scales. We tested the common perception that commercial and recreational fishing on inshore temperate reefs generate spatial impacts that diminish with distance from fisher access points. We collected data on harvested and non-harvested reef species using underwater visual censuses at 133 shallow rocky reef sites around Tasmania and tested for relationships between assemblage and species level indices of fishing impacts and distance to the nearest boat launching ramp. Slopes of size spectra of fish communities tended to decrease with distance from the nearest boat ramp, with this relationship apparently resulting from low numbers of large fish (> 30 cm TL) and a greater number of smaller fish (< 15 cm TL) at sites closest to access points. At the species level, relationships were evident either in the abundance of legal individuals or the mean size of harvested species with distance to the nearest boat ramp, except for rock lobster. Patterns for rock lobster differed when areas in which commercial or recreational fisheries dominated were considered separately from the statewide analysis. A pattern of increasing numbers of legal lobsters with increasing distance from boat ramps was observed, but only in the areas in which the recreational fishery dominated. Observed relationships in all species were consistent with greater fishing impacts at sites closest to boat ramps, with the exception of exploited wrasses. Banded morwong, which are subject to a live export fishery, appeared to be most affected by proximity to boat ramps. Conversely, no relationships were found between the abundance or size of the most frequently occurring non-harvested species and distance to boat ramps.These results support the hypothesis that greater fishing impacts occur at more accessible sites over the entire Tasmanian coastline. The variability of results among individual species are likely, at least in part, to be related to differences in fisheries characteristics such as vessel size and range, as well as the suitability of our methods for detecting impacts. The potential of such a pattern in fishing impacts to be evident in other locations will thus likely depend on characteristics of the particular fishes and fisheries.  相似文献

Despite increasing scientific and public concerns on the potential impacts of global ocean warming on marine biodiversity, very few empirical data on community-level responses to rising water temperatures are available other than for coral reefs. This study describes changes in temperate subtidal reef communities over decadal and regional scales in a location that has undergone considerable warming in recent decades and is forecast to be a 'hotspot' for future warming.
Plant and animal communities at 136 rocky reef sites around Tasmania (south-east Australia) were censused between 1992 and 1995, and again in 2006 and 2007. Despite evidence of major ecological changes before the period of study, reef communities appeared to remain relatively stable over the past decade. Multivariate analyses and univariate metrics of biotic communities revealed few changes with time, although some species-level responses could be interpreted as symptomatic of ocean warming. These included fishes detected in Tasmania only in recent surveys and several species with warmer water affinities that appeared to extend their distributions further south. The most statistically significant changes observed in species abundances, however, were not related to their biogeographical affinities. The majority of species with changing abundance possessed lower to mid-range abundances rather than being common, raising questions for biodiversity monitoring and management. We suggest that our study encompassed a relatively stable period following more abrupt change, and that community responses to ocean warming may follow nonlinear, step-like trajectories.  相似文献
