The 16S-like ribosomal RNA genes from Mallomonas papillosa Harris et Bradley (Synurophyceae) and Tribonema aequale Pascher (Xanthophyceae) were sequenced and compared to those of other eukaryotes. Mallomonas is closely related to Ochromonas (Chrysophyceae) and supports the general hypothesis of a close phylogenetic relationship between the Synurophyceae and Chrysophyceae. Tribonema is specifically related to Costaria costata (C. A. Agardh) Saunders (Phaeophyceae) demonstrating an unexpected phylogenetic relationship between the Xanthophyceae and Phaeophyceae. Distance and parsimony analysis place these four chromophyte genera in a complex evolutionary assemblage that includes the Bacillariophyceae and Oomycetes but excludes the Dinophyceae. The close relationship between the chromophyte algae and the Öomycete fungi supports the hypothesis that protists with tripartite hairs form a natural assemblage.  相似文献

Tribonema bombycinum (Xanthophyceae), was examined. T. bombycinum shifted from vegetative cells to akinetes with starving by a prolonged batch culture, by culture with a diluted medium, or by culture with a single nutrient-deficient medium. In addition, akinetes developed by desiccation, but cold treatment at 4 C did not facilitate akinete formation. During starving, the vegetative cells, which had a large central vacuole in the protoplasm and thin cell walls, finally changed to akinetes, which had many small vacuoles and oil droplets in the protoplasm and thick cell walls. During akinete formation by starving, the freezing tolerance (LT50) increased gradually from −3 C in vegetative cells to far below −30 C in akinetes. When vegetative cells were subjected to equilibrium freezing, their size shrank greatly and aparticulate domains accompanied by fracture-jump lesions developed in the plasma membranes. Akinetes subjected to equilibrium freezing showed little shrinkage, and freezing-induced ultrastructural changes did not occur in the plasma membranes. The morphological changes in the process of akinete formation and the responses to equilibrium freezing resembled those of cold-acclimated terrestrial plants. 囊状黄丝藻在不同初始氮浓度条件下特殊的油脂积累规律   总被引:2,自引:0,他引:2  
对不同初始氮浓度条件下囊状黄丝藻(Tribonema utriculosum SAG22.94)的生长状况、油脂含量和脂肪酸组成与含量进行研究。结果显示,囊状黄丝藻在氮浓度为3.0 mmol/L时,获得生物质浓度最高,为6.39 g/L;氮浓度为18.0 mmol/L时获得总脂和总脂肪酸含量最高,分别为细胞干重的44.62%和42.21%;上述3个指标单位体积的产率均在氮浓度3.0 mmol/L时达到最高值,分别为0.538、0.209和0.206 g·L~(-1)·d~(-1)。在4种初始氮浓度条件下,囊状黄丝藻油脂和脂肪酸含量可随着氮浓度增加而增加。脂肪酸含量分析结果显示,该藻的主要脂肪酸为豆蔻酸(C14∶0)、棕榈酸(C16∶0)、棕榈油酸(C16∶1ω7)、花生四烯酸(C20∶4ω6)和二十碳五烯酸(C20∶5ω3,EPA)。其中棕榈油酸含量最高,占总脂肪酸含量的36.53%~50.08%。研究结果表明囊状黄丝藻在不同初始氮浓度条件下具有特殊的油脂积累规律,是一株具有重要应用价值的产油丝状微藻。  相似文献

丝状微藻黄丝藻Tribonema sp.具有抗浮游动物捕食、易收获、油脂含量高等优点,且其脂肪酸组分中含有丰富的棕榈油酸(Palmitoleic acid,PA)和二十碳五烯酸(Eicosapentaenoic acid,EPA),被认为是生产生物柴油和高附加值产品的重要原料。为了提高黄丝藻脂质生产效率,文中研究了不同浓度的氮(NaNO3:255–3060mg/L)、磷(K2HPO4:4–240mg/L)、铁((NH4)3FeC12H10O14:0.6–12mg/L)、镁(MgSO4:7.5–450mg/L)元素对黄丝藻FACHB-1786生长、油脂积累和脂肪酸组分的影响。结果表明,培养基中磷、铁、镁三种元素的浓度对黄丝藻生长具有显著影响,其中增加MgSO4浓度可显著提高黄丝藻的生物量,当MgSO4浓度增加至450 mg/L时,获得最大生物量为8.09 g/L,显著高于目前报道的有关黄丝藻自养条件下获得的生物量;氮元素浓度对黄丝藻的生长没有显著影响(P>0.05),但高浓度氮元素有利于黄丝藻脂质的积累;黄丝藻FACHB-1786在765 mg/L NaNO3、80 mg/L K2...  相似文献

The large subunit ribosomal RNA sequences from the heterokont algae Ochromonas danica, Nannochloropsis salina, and Tribonema aequale were determined. These sequences were combined with small subunit ribosomal RNA sequences in order to carry out a phylogenetic analysis based on neighbor-joining, maximum parsimony, and maximum likelihood methods. Our results indicate that heterokont fungi and heterokont algae each are monophyletic, and confirm that they together form a monophyletic group called ``stramenopiles.' Within the heterokont algae, the eustigmatophyte Nannochloropsis salina either clusters with the chrysophyte Ochromonas danica or forms a sister group to a cluster comprising the phaeophyte Scytosiphon lomentaria and the xanthophyte Tribonema aequale. The alveolates were identified as the closest relatives of the stramenopiles, but the exact order of divergence between the eukaryotic crown taxa could not be established with confidence. The chlorophyll-protein complexes of the yellow alga Synura petersenii (Chrysophyceae) and the yellow-green alga Tribonema aequale (Xanthophyceae) were studied. The sodiumdodecylsulfate/sodiumdesoxycholate solubilized photosynthetic membranes of these species yielded three distinct pigment-protein complexes and a non-proteinuous zone of free pigments, when subjected to SDS polyacrylamid gel electrophoresis. The slowest migrating protein was identical to complex I (CP I), the P-700 chlorophyll a-protein, which possessed 60 chlorophyll a molecules per reaction center in Tribonema and 108 in Synura. The zone of intermediate mobility contained chlorophyll a and carotenoids. The absorption spectrum of this complex was very similar to the chlorophyll a-protein of photosystem II (CP a), which is known from green plants. The fastest migrating pigment protein zone was identified as a light-harvesting chlorophyll-protein complex. In Synura this protein was characterized by the content of chlorophyll c and of fucoxanthin. Therefore this complex will be named as LH Chl a/c-fucocanthin protein. In addition to the separation of the chlorophyll-protein complexes the cellular contents of P-700, cytochrome f (bound cytochrome) and cytochrome c-553 (soluble cytochrome) were measured.
