BACKGROUND: The germination of both pollen and fern spores results in the emergence of a cell-pollen tube from pollen, rhizoid from spore-that grows in a polar fashion, primarily at its apical end. In both of these tip-growing cells, the delivery of secretory vesicles to the growing end is guided in part by a calcium gradient, with calcium entering at the tip where it is most highly concentrated. The similarities between the two systems extend beyond tip-focused calcium gradients to encompass signalling pathways and elements including calmodulin, nitric oxide, annexins and Rop-GTPases. SCOPE AND AIMS: This review is limited to those pathways and elements that function similarly in fern and pollen systems based on currently available evidence. The aim is to illustrate the common mechanisms by which tip growth occurs, facilitate further investigations into this area, and examine the implications for the evolutionarily conserved control of tip growth. CONCLUSIONS: The interplay of calcium, nitric oxide and other effectors in both pollen and fern spores suggests certain signalling pathways became important regulators of germination and growth early in the evolution of land plants. Both large- and small-scale comparative genomic methods have shown to be promising in their ability to find new and relevant comparisons for further research. Cross-species comparisons may serve to speed up this process by highlighting both basic pathways and system-specific deviations.  相似文献

Movement patterns of Alaska harbor seal pups were studied using satellite telemetry during 1997–2000. Mean tracking duration was 277.3 d (SD = 105.8) for Tugidak Island pups ( n = 26) and 171.2 d (108.3) for Prince William Sound (PWS) pups ( n = 27). Movements were similar for males and females and were largely restricted to the continental shelf. Multiple return trips of > 75 km from the natal area and up to ∼3 wk duration were most common, followed by movements restricted to <25 km from the natal area; one way movements from the natal site were rare. Distances moved and home range sizes remained relatively stable or increased gradually from July through winter, then decreased markedly through spring. Monthly movements (maximum distance from tagging location, mean distance from haul-outs to at-sea locations, and home range size) were significantly greater for Tugidak vs . PWS pups. Six of seven pups from each region that traveled farthest and were tracked the longest had returned to their tagging site when their last location was recorded, indicating philopatry or limited dispersal during their first year of life. Seal pups exhibited similar movement patterns in the distinct habitats of the two regions, but differed in the spatial extent of their movements.  相似文献

Seven prey species ( n total > 2,700) were fed to seven captive male Pacific harbor seals ( Phoca vitulina richardii ) in 177 experimental meals to quantify biases associated with scat analysis and current consumption models. Hard parts from an individual meal were recovered in an average of 3.8 ± 1.8 scats (range 1–10; mean ± SD). Overall, 57.7 ± 33.2% of otoliths and 89.5 ± 15.5% of squid beaks were recovered. Recovery rates varied, and prey with smaller, fragile otoliths were recovered in lesser quantities than prey with larger, robust otoliths. Recovery rates of all prey except pink salmon were improved by a mean of 31.7% when all diagnostic structures were included in estimates. Estimated recovery of pink salmon was 9.5 times that fed seals based on the all-structure technique. Mean length reduction of recovered otoliths was 20.4 ± 10.1%. Correction factors calculated from average length reduction improved length estimates for all fish species. Grade-specific length correction factors (gLCFs) reduced variability in all of the estimates and significantly improved estimates of prey with highly eroded otoliths including Pacific hake and shortbelly rockfish. The Biomass Reconstruction (BR) model accurately predicted biomass consumption within 4% of known consumption, whereas estimates based on frequency of occurrence were inaccurate.  相似文献

Adrenal function in harbor seals (Phoca vitulina richardii) was evaluated using adrenocorticotrophic hormone (ACTH) stimulation tests and fecal cortisol levels. The effect of ACTH administration on plasma cortisol and aldosterone levels in five free-living and 14 rehabilitated harbor seal pups was determined using enzyme immunoassay and radioimmunoassay, respectively. In free-living seals, injection of ACTH caused a significant increase in mean plasma cortisol but not of mean aldosterone levels 60 min postinjection. In these seals, mean initial plasma aldosterone was significantly higher than initial levels in rehabilitated seals, while initial cortisol levels were similar. Of the rehabilitated seals, eight died with adrenal cortical necrosis associated with herpesvirus inclusions, while six lived to be released. In the seals that were released, both mean initial cortisol levels and response to ACTH decreased through rehabilitation. In the seals that died, mean initial cortisol and response to ACTH increased through rehabilitation. The differences between initial cortisol levels in seals that lived and those that died were significant at weeks two and four of rehabilitation but not at the week of admission. There was considerable individual variation in initial plasma aldosterone levels and responses to ACTH, although initial aldosterone levels were significantly higher in rehabilitated seals that died than in seals that lived. Seals with adrenal necrosis associated with herpesvirus infection did not have decreased adrenal hormone responses to ACTH. Differences between initial hormone levels and responses to ACTH in different groups of seals may be associated with differing stress levels. Fecal cortisol assays were not a useful method of assessing adrenal function in these seals, as measured levels did not correlate with plasma cortisol levels.  相似文献

The endogenous localisation of peroxidase and hydrogen peroxide (H2O2) was detected when gametophytes of the fern, Ceratopteris richardii, were exposed to the plant pathogenic fungi Sclerotium rolfsii and Sclerotinia sclerotiorum and Phytophthora infestans, an oomycete, in a gnotobiotic system. This was accomplished by light microscopy using 3,3′‐diaminobenzidine, guaiacol and H2O2 and starch potassium iodide (KI) staining procedures, which facilitated the observation of the reaction in vivo and in situ, without physically damaging the tissues. All three staining methods promoted staining at the rhizoid regions. Although most of the cells were destroyed when gametophytes were exposed to S. rolfsii and S. sclerotiorum, there was staining where mycelial growth was confluent with cell walls. A qualitative test confirmed that the colour change in starch KI agar medium, as well as in the histochemical test with starch KI, was because of H2O2 secreted by S. rolfsii or S. sclerotiorum and not because of oxalic acid. When gametophytes were exposed to P. infestans, no infection occurred, but localisation of H2O2 and peroxidase was detected irrespective of staining methods tested. Based on the observation on gametophytes grown in presence of P. infestans, it is possible that the peroxidase in plants coupled with H2O2 may prevent the invasion of nonpathogens by functioning as a barrier. This fern–pathogen model system has potential for application as a tool to study the host–parasite interaction in a gnotobiotic system.  相似文献

Organochlorine (OC) pesticides and polychlorinated biphenyls (PCBs) have been detected in a variety of marine mammal species at levels associated with adverse health effects. Little is known about OC levels and impacts on health in pinnipeds with different life histories. We determined the health and levels of 18 OC pesticides and 16 PCB congeners in blubber samples from 20 Steller sea lions and 39 Pacific harbor seals stranded from Oregon and Southern Washington. The most commonly detected OC at the highest concentration was p,p′- dichlorodiphenyldichloroethylene (DDE). PCBs were detected in all samples as well. Hypothesis testing indicated that diseased Steller sea lions (males and females combined) had higher contaminant concentrations than healthy Steller sea lions, and diseased Pacific harbor seals had higher concentrations of total OCs than healthy animals. Differences were also noted between diseased and healthy animals when looking at individual sexes of each species. Diseased Steller sea lions had higher mean contaminant levels than diseased harbor seals and healthy Steller sea lions had higher mean contaminant concentrations than healthy Pacific harbor seals. These results show that species differences exist in both contaminant loads and sensitivity to contaminants, which may be due to differences in life histories and physiology.  相似文献

In this study, we observed the behavior of two age groups of harbor seal (Phoca vitulina richardii), i.e., yearlings, which often become bycaught and individuals 2 yr or older, during their stays at salmon set nets at Cape Erimo, Japan. From July to November 2011–2015, acoustic receivers that track tagging seals were attached to three set nets located near the haul‐out site. Our observations showed that the mean visit interval and the mean stay of seals at the set nets during fishing operations were significantly higher than before operations. A cluster analysis, where Cluster 1 consisted of seals that spent a long time at the set nets and Cluster 2 consisted of the other seals, showed that seals in Cluster 1 visited the set nets every day, while Cluster 2 seals visited once every 3–5 d. In addition, age 2 + seals in Cluster 1 stayed longer at regular time frames (at night), suggesting that individuals of this age group in Cluster 1 are highly dependent on the salmon set nets as foraging sites at night. It is especially clear that the set nets near the haul‐out site have influenced the foraging ecology of some seals.  相似文献

The stl1 and stl2 mutations confer low and high levels of NaCl tolerance to gametophytes of the fern Ceratopteris richardii, respectively. As an initial characterization of these mutations, the levels of various organic solutes, tissue ion content and water relations were examined in the wild-type and mutant strains in the absence and presence of 60 mol m-3 NaCl stress (a level which results in a 20, 15 and 0% reduction in gametophyte growth in the wild-type, stl1 mutant and stl2 mutant, respectively). All strains exhibited major changes in organic and inorganic solute levels and water relations in response to 60 mol m-3 NaCl stress. Differences in organic solute levels and water relations between the wild-type and mutant strains in the absence and in response to 60 mol m-3 NaCl stress were minimal. Analysis of tissue ion content showed that stl1 was associated with a slight reduction in Na+ accumulation during 60 mol m-3 NaCl stress. stl2 was associated with (1) higher constitutive levels of K+ and (2) continued selective accumulation of K+ and reduced accumulation of Na+ during 60 mol m-3 NaCl stress. A K+/Na+ ratio close to 1 was observed in the wild-type during 60 mol m-3 NaCl stress, while higher ratios were detected in stl1 and stl2 (1·7 and 4·0, respectively). The findings of this study suggest that the tolerance imparted by stl1 and stl2 is associated with altered ion accumulation during NaCl stress, rather than an enhanced ability to accumulate organic solutes to be used for osmotic adjustment of the cytoplasm.  相似文献

The roles of K+ uptake and loss in the salinity response of the wild type and the salt-tolerant mutant stl2 of Ceratopteris richardii were studied by measuring Rb+ influx and loss and the effects of Na+, Mg2+, Ca2+ and K+-transport inhibitors. In addition, electrophysiological responses were measured for both K+ and Rb+ and for the effects of Na+ and NH4+ on subsequent K+-induced depolarizations. stl2 had a 26–40% higher uptake rate for Rb+ than the wild type at 0.5–10 mol m?3 RbCl. Similarly, membrane depolarizations induced by both RbCl and KCl were consistently greater in stl2. In the presence of 0–180 mol m?3 NaCl, stl2 maintained a consistently greater Rb+ influx than the wild type. stl2 retained a greater capacity for subsequent KCl-induced depolarization following exposure to NaCl. Five mol m?3 Mg2+ decreased Rb+ uptake in stl2; however, additional Mg2+ up to 40 mol m?3 did not affect Rb+ uptake further. Ca2+ supplementation resulted in a very minor decrease of Rb+ uptake that was similar in the two genotypes. Tetraethylammonium chloride and CsCl gave similar inhibition of Rb+ uptake in both genotypes, but NH4Cl gave substantially greater inhibition in the wild type than in stl2. NH4Cl resulted in a greater membrane depolarization in the wild type and the capacity for subsequent depolarization by KCl was markedly reduced. stl2 exhibited a higher Independent loss of Rb+ than the wild type, but, in the absence of external K+, loss of Rb+ was equivalent in the two genotypes. Since constitutive K+ contents are nearly identical, we conclude that high K+ influx and loss exact a metabolic cost that is reflected in the inhibition of gametophytic growth. Growth inhibition can be alleviated by reduced supplemental K+ or by treatments that slightly reduce K+ influx, such as moderate concentrations of Na+ or Mg2+. We propose that high throughput of K+ allows maintenance of cytosolic K+ under salt stress and that a high uptake rate for K+ results in a reduced capacity for the entrance and accumulation of alternative cations such as Na+ in the cytosol.  相似文献
