Abstract: Binding of 1-[1-(2-[3H]thienyl)cyclohexyl]piperidine ([3H]TCP) to mouse brain and spinal cord membranes was studied using compounds selective for the NMDA-coupled 1-(1-phenylcyclohexyl)piperidine (PCP) and/or σ recognition sites. In both tissues, [3H]TCP labeled two populations of binding sites. Density of the low-affinity sites was approximately the same in both tissues, but the population of the high-affinity [3H]TCP sites was three times bigger in the brain than in the spinal cord. Self- and cross-displacement studies showed that the high-affinity [3H]TCP binding sites could be identical with NMDA receptor-coupled PCP sites, whereas the low-affinity [3H]TCP sites may be associated with σ binding sites in both tissues. The NMDA-coupled PCP sites labeled in the presence of 6.25 n M [3H]TCP constituted a much higher percentage of the total binding in the brain (75%) than in the spinal cord (44%). Consistent with this, reintroduction of glycine and glutamate significantly increased, but DA antagonists significantly inhibited [3H]TCP binding in the brain but not in the spinal cord. Together, these data suggest that a large component of [3H]TCP-labeled binding sites in the spinal cord may be associated with σ but not the NMDA receptor-coupled PCP sites.  相似文献

The first structurally characterised oxomolybdenum(V) complexes with thienyl carboxylate ligands were prepared by the reaction of [Mo2O3(C5H7O2)4] or (NH4)2[MoOCl5] with the corresponding acid (2-thiophenecarboxylic, 5-methyl-2-thiophenecarboxylic or 3-(3-thienyl)acrylic acid). Complexes [Mo2O3(μ-OC2H5)(μ-O2CR)(C5H7O2)2](R = -C4H3S (1), -C4H2S(CH3) (2) or -CHCHC4H3S (3)) were obtained upon substitution of two acetylacetonate ligands from [Mo2O3(C5H7O2)4] with RCOO in dry ethanol. Reactions of (NH4)2[MoOCl5] with the corresponding thienyl carboxylic acid in the presence of γ-picoline (C6H7N) yielded complexes (C6H7NH)[Mo2O4(μ-O2CR)Cl2(C6H7N)2] (R = -C4H3S (4), -C4H2S(CH3) (5) or -CHCHC4H3S (6)). All of the six new complexes were characterised as dinuclear. The molecular structures of 1, 3, 4·0.5CH3CN and 5 were determined by the single crystal X-ray diffraction method. In the complexes the two molybdenum atoms are doubly bridged either by one oxygen and one ethoxy-oxygen, or alternatively by two oxo-oxygens, and are additionally bridged by the thienyl carboxylate ion in a didentate bridging manner. All complexes were further characterised by means of chemical analysis, IR spectroscopy, TG and in some cases by the one and two-dimensional NMR method.  相似文献

A NMR study of the reaction mixture of the square planar [Rh((C4H3S)COCHCOR)(CO)(PPh3)] complex and CH3I, where R = CF3, C6H5 or C4H3S, revealed that two types of alkyl and one (R = CF3) or two (R = C6H5 or C4H3S) types of acyl species exist in the system. Two isomers of each species with an unsymmetrical β-diketonato ligand were observed. 1H-1H NOESY NMR unambiguously showed that the PPh3 group is in the apical position in the more stable RhIII-alkyl product. Theoretical computations of the equilibrium geometry of the possible reaction products, consistent with experimental observations, revealed that the first alkyl product results from trans addition to RhI and that the second thermodynamic alkyl product adopts an octahedral geometry with the PPh3 group and the iodide above and below the square planar plane. Theoretical computations also revealed that the thermodynamic acyl product adopts a square-pyramidal geometry with the COCH3 group in the apical position.  相似文献
