Ceramides are sphingolipids that greatly stabilize ordered membrane domains (lipid rafts), and displace cholesterol from them. Ceramide-rich rafts have been implicated in diverse biological processes. Because ceramide analogues have been useful for probing the biological function of ceramide, and may have biomedical applications, it is important to characterize how ceramide structure affects membrane properties, including lipid raft stability and composition. In this report, fluorescence quenching assays were used to evaluate the effect of analogues of ceramide with different N-acyl chains or different sphingoid backbones on raft stability and sterol content. The effect of replacing 18 mol% of sphingomyelin (SM) with ceramide in vesicles composed of a 1:1 (mol:mol) mixture of SM and dioleoylphosphatidylcholine (DOPC), with or without 25 mol% sterol, was examined. In the absence of sterol, the thermal stability of the SM-rich ordered domains increased with ceramide N-acyl chain length in the order C2:0 ∼ C6:0 ∼ C8:0 < no ceramide < C12:0 < C16:0. In vesicles containing 25 mol% cholesterol (1:1:0.66 sphingolipid:DOPC:cholesterol), the dependence of raft stability on ceramide N-acyl chain length increased in the order C8:0 ∼ C6:0 < C2:0 < C12:0 ∼ no ceramide < C16:0. We also studied the stability of lipid rafts in the presence of N-lauroyl- and N-palmitoylsphingosine analogues containing altered structures in or near the polar portion of the sphingoid base. In almost all cases, the analogues stabilized rafts to about the same degree as a normal ceramide containing the same acyl chain. The only exception was N-palmitoyl-4D-ribophytosphingosine, which was very strongly raft-stabilizing. We conclude that variations in sphingoid base structure induce only insignificant changes in raft properties. N-Lauroyl and N-palmitoylsphingosine and their analogues displaced sterol from rafts to a significant degree. Both C12:0 and C16:0 analogues of ceramide may be good mimics of natural ceramide, and useful for cellular studies in which maintenance of the normal physical properties of ceramide are important.  相似文献

Rhythmic grouping and discrimination is fundamental to music. When compared to the perception of pitch, rhythmic abilities in animals have received scant attention until recently. In this experiment, four pigeons were tested with three types of auditory rhythmic discriminations to investigate their processing of this aspect of sound and music. Two experiments examined a meter discrimination in which successively presented idiophonic sounds were repeated in meters of different lengths in a go/no-go discrimination task. With difficulty, the birds eventually learned to discriminate between 8/4 and 3/4 meters constructed from cymbal and tom drum sounds at 180 beats per minute. This discrimination subsequently transferred to faster tempos, but not to different drum sounds or their combination. Experiment 3 tested rhythmic and arrhythmic patterns of sounds. After 40 sessions of training, these same pigeons showed no discrimination. Experiment 4 tested repetitions of a piano sound at fast and slow tempos. This discrimination was readily learned and showed transfer to novel tempos. The pattern of results suggests that pigeons can time periodic auditory events, but their capacity to understand generalized rhythmic groupings appears limited.  相似文献

Interaction of filipin and amphotericin B with sterols in phosphatidylcholine membranes has been studied using various spin probes; epiandrosterone, cholestanone, phosphatidylcholine with 12-nitroxide or 5-nitroxide stearate attached to 2 position and also with tempocholine at the head group. Filipin caused increase in the fluidity of cholesterol-containing phosphatidylcholine membranes near the center, while it rather decreased the fluidity near the polar surface. On the other hand, amphotericin B did not apparently affect the fluidity. In the electron spin resonance spectrum of steroid spin probes in the antibiotic-containing membranes, both bound and free signals were observed and the association constant was calculated from the siganal intensity. In the binding of steroids with filipin, both 3 and 17 positions were involved, while the 17 position was less involved in the binding with amphotericin B. Phase change in the host membrane markedly affected the interaction of filipin with epiandrosterone probe. The bound fraction jumped from 0.4 to 0.8 on going to the crystalline state and increased further with decrease in temperature. The overall splitting of the bound signal also increased on lowering the temperature below phase transition. This change was attributed to aggregate formation of filipin-steroid complexes in the crystalline state. On the other hand, effect of phase transition was much smaller on the interaction of amphotericin B with the steroid probe.  相似文献

本文以1979年和1985年两次全国体质调研的结果为基础材料,对我国汉族儿童在身高加速现象方面的城乡差别以及躯干比例在身高加速生长中的变动进行了分析,初步得出如下结论:(1) 6年间城乡汉族儿童的身高加速生长、发育提前;(2) 乡村儿童身高增加和身高及坐高发育高峰年龄提前的幅度大于同性别城市儿童;(3) 青春期儿童的身高和躯干比例变化较大;(4) 儿童身高和躯干比例的变化主要是成熟节律提前造成的。  相似文献

The fast–slow continuum hypothesis has been proposed to explain the diversity of life-history patterns exhibited by biological populations, but the quantification and population-dynamic consequences of the continuum has remained unclear. I used the ratio of fertility rate to age at first reproduction (F/α ratio) to quantify the tempo of life-history of 138 populations of mammals, and investigated the life-history and population-dynamic consequences of being “fast” or “slow”. “Fast” mammals (F/α>0.60) were characterized by early maturity, short lifespans, low survival rates, and high fertility and projected population growth rate (λ) compared to “slow” (F/α<0.15) mammals. In “fast” populations, λ was overwhelmingly most sensitive to changes in reproductive parameters (age at first reproduction and fertility rates) and relatively insensitive to changes in survival rates. In “slow” populations, λ was very sensitive to changes in juvenile or adult survival rates, and relatively insensitive to changes in reproductive parameters. The pattern of relationships between the F/α ratio and life-history variables, λ, and elasticity of λ to changes in life-history variables persisted even after the effects of body size and phylogeny were statistically removed. These results suggest that fast–slow continuum in mammalian life-history is independent of body size or phylogeny, that the F/α ratio adequately quantifies the position of a population along a fast–slow continuum, and that the tempo of life- histories has substantial population-dynamic consequences.

Zusammenfassung

Die r-K-Kontinuum-Hypothese wurde aufgestellt, um die Diversität von ,,life-history“-Mustern biologischer Populationen zu erklären, aber die Quantifizierung und die Kosnsequenzen für die Populationsdynamik des Kontinuums blieben unklar. Ich benutze das verhältnis der Fortpflanzungsrate zum Fortpflanzungsalter (F/α-Verhältnis) um die Geschwindigkeit der ,,life-history“ von 138 Populationen von Säugetieren zu quantifizieren und untersuchte die Konsequenzen fur die Lebensweise sowie die Populationsdynamik des,,schnell“oder,,langsam “-Seins. ,,Schnelle“Säugetiere (F/α>0.60) waren durch eine frühe Reife, kurze Lebenszeiten, geringe Überlebensraten sowie durch eine große Fertilität und hochgerechnete Populationswachstumsrate (λ) im Vergleich zu ,,langsamen“(F/α<0.15) Säugetieren charakterisiert. In ,,schnellen“ Population reagierte (λ) überwältigend sensibel auf Änderungen in den Fortpflanzungsparametern (Fortpflanzunsalter und Fertilitätsrate) und relativ gering auf Veräanderungen in der Überlebensrate. In ,,langsamen“ Populationen reagierte (λ) sehr sensibel auf Veräanderungen in den reproduktiven Parametern. Das Muster der Beziehung zwischen dem (F/α-Verhältnis) und den Variablen der ,,life-history“,λ, und die Elastizität von λ gegenüber Veränderungen in den variablen der Lebensweise bliev sogar bestehen, nachdem die Effekte von Körpergröße und Phylogenese statistisch eliminiert wurden. Diese Ergebnisse lassen vermuten, dass das r-K-Kontinuum in der ,,life-history“der Säugetiere unabhängig von der Körpergröße und Phylogenie ist, dass das F/α-Verhältnis die Position einer Population im r-K-Kontinuum quantifiziert und dass die Geschwindigkeit der,,life-history“beachtliche konsequenzen fur die Populationsdynamik hat.  相似文献

With application of EPR and 1H NMR techniques genistein interaction with liposomes formed with egg yolk lecithin and with erythrocyte membranes was assessed. The present study addressed the problem of genistein localization and its effects on lipid membrane fluidity and protein conformation. The range of microscopic techniques was employed to study genistein effects on HeLa cells and human erythrocytes. Moreover, DPPH bioassay, superoxide anion radical test and enzymatic measurements were performed in HeLa cells subjected to genistein. The gathered results from both EPR and NMR techniques indicated strong ordering effect of genistein on the motional freedom of lipids in the head group region and the adjacent hydrophobic zone in liposomal as well as in red blood cell membranes. EPR study of human ghost showed also the changes in the erythrocyte membrane protein conformation. The membrane effects of genistein were correlated with the changes in internal membranes arrangement of HeLa cells as it was noticed using transmission electron microscopic and fluorescent techniques. Scanning electron and light microscopy methods showed that one of the aftermaths of genistein incorporation into membranes was creation of echinocytic form of the red blood cells with reduced diameter. Genistein improved redox status of HeLa cells treated with H2O2 by lowering radicals' level.  相似文献

High levels of reactive oxygen species (ROS), which may be related to reduced semen quality, are detected during semen cryopreservation in some species. The objectives of this study were to measure the oxidative stress during ram semen cryopreservation and to evaluate the effect of adding 2 antioxidant mimics of superoxide dismutase (Tempo and Tempol) during the cooling process on sperm motility, viability, acrosomal integrity, capacitation status, ROS levels, and lipid peroxidation in frozen and/or thawed ram spermatozoa. Measuring of ROS levels during the cooling process at 35, 25, 15, and 5 °C and after freezing and/or thawing showed a directly proportional increase (P < 0.05) when temperatures were lowering. Adding antioxidants at 10 °C confered a higher motility and sperm viability after cryopreservation in comparison with adding at 35 °C or at 35 °C/5 °C. After freezing and/or thawing, sperm motility was significantly higher (P < 0.05) in Tempo and Tempol 1 mM than that in control group. Percentage of capacitated spermatozoa was lower (P < 0.05) in Tempo and Tempol 1 mM in comparison with that in control group. In addition, ROS levels and lipid peroxidation in group Tempo 1 mM were lower (P < 0.05) than those in control group. These results demonstrate that ram spermatozoa are exposed to oxidative stress during the cooling process, specifically when maintained at 5 °C and that lipid peroxidation induced by high levels of ROS decreases sperm motility and induces premature sperm capacitation. In contrast, the addition of Tempo or Tempol at 0.5 to 1 mM during the cooling process (10 °C) protects ram spermatozoa from oxidative stress.  相似文献

A novel biopolymer-based antioxidant, chitosan conjugated with gallic acid (chitosan galloylate, chitosan-GA), is proposed. Electron paramagnetic resonance (EPR) demonstrates a wide range of antioxidant activity for chitosan-GA as evidenced from its reactions with oxidizing free radicals, that is, 1,1-diphenyl-2-picryl-hydrazyl (DPPH), horseradish peroxidase (HRP)-H2O2, carbon-centered alkyl radicals, and hydroxyl radicals. The EPR spectrum of the radical formed on chitosan-GA was attributed to the semiquinone radical of the gallate moiety. The stoichiometry and effective concentration (EC50) of the DPPH free radical with chitosan-GA show that the radical scavenging capacity is maintained even after thermal treatment at 100 °C for an hour. Although the degree of substitution of GA on chitosan was about 15%, its antioxidant capacity, that is, the reaction with carbon-centered and hydroxyl radicals, is comparable to that of GA.  相似文献

Kelvin considered it unlikely that sufficient time had elapsed on the earth for life to have reached its present level of complexity. In the warm surroundings in which life first appeared, however, elevated temperatures would have reduced the kinetic barriers to reaction. Recent experiments disclose the profound extent to which very slow reactions are accelerated by elevated temperatures, collapsing the time that would have been required for early events in primordial chemistry before the advent of enzymes. If a primitive enzyme, like model catalysts and most modern enzymes, accelerated a reaction by lowering its enthalpy of activation, then the rate enhancement that it produced would have increased automatically as the environment cooled, quite apart from any improvements in catalytic activity that arose from mutation and natural selection. The chemical events responsible for spontaneous mutation are also highly sensitive to temperature, furnishing an independent mechanism for accelerating evolution.  相似文献

Synopsis The interrelationships of 31 actinistian species (including Latimeria chalumnae) are analyzed based on a cladistic analysis of 75 osteological characters. Inference of evolutionary trends (e.g., modification of body shape and skull morphology) from the phylogenetic patterns demonstrates that the morphology of actinistians is less conservative than has been proposed previously. This empirical cladistic approach supports two distinct tempos of evolution during an evolutionary history of 380 million years. Along a phylogenetic pathway originating with a Devonian stem-species and ending with the living Latimeria chalumnae (including 101 morphological changes and 18 cladogenetic events), the first tempo occurred during the Devonian — Permian periods as a decreasing rate of morphological changes, which was followed by a stabilizing tempo during the Permian — Recent periods. The decreasing tempo is characterized by a sequence of gradual versus quantum temporal changes and low versus faster rates, whereas the stabilizing tempo primarily is gradual and low. In contrast to a common assumption, no significant correlation was found between the rates of morphological evolution and the temporal diversity of species.  相似文献
