苏芸金杆菌液体深层发酵中用营养体接种二级发酵工艺和用二级发酵初期培养物作种子进行三级发酵工艺是可行的。试验结果表明:二级发酵种子罐营养体最佳移种茵龄为9h左右,此时营养体数量多、整齐、染色均匀,显微镜下菌体有折光点存在,三级接种营养体菌龄约为4h左右,菌体数量多,同步率高,少量染色不均匀。发酵液含菌数和发酵产品毒力均达到芽抱接种相同水平,但生产周期明显缩短4-5h,因此相应提高发酵罐生产能力20%。 相似文献

Wildfires are a typical event in many Australian plant communities. Vesicular-arbuscular mycorrhizal (VAM) fungi are important for plant growth in many communities, especially on infertile soils, yet few studies have examined the impact of wildfire on the infectivity of VAM fungi. This study took the opportunity offered by a wildfire to compare the infectivity and abundance of spores of VAM fungi from: (i) pre-fire and post-fire sites, and (ii) post-fire burned and unburned sites. Pre-fire samples had been taken in May 1990 and mid-December 1990 as part of another study. A wildfire of moderate intensity burned the site in late December 1990. Post-fire samples were taken from burned and unburned areas immediately after the fire and 6 months after the fire. A bioassay was used to examine the infectivity of VAM fungi. The post-fire soil produced significantly less VAM infection than the pre-fire soil. However, no difference was observed between colonization of plant roots by VAM fungi in soil taken from post-fire burned and adjacent unburned plots. Soil samples taken 6 months after the fire produced significantly more VAM than corresponding soil samples taken one year earlier. Spore numbers were quantified be wet-sieving and decanting of 100-g, air-dried soil subsamples and microscopic examination. For the most abundant spore type, spore numbers were significantly lower immediately post-fire. However, no significant difference in spore numbers was observed between post-fire burned and unburned plots. Six months after the fire, spore numbers were the same as the corresponding samples taken 1 year earlier. All plants appearing in the burned site resprouted from underground organs. All post-fire plant species recorded to have mycorrhizal associations before the fire had the same associations after the fire, except for species of Conospermum (Proteaceae), which lacked internal vesicles in cortical cells in the post-fire samples. 相似文献

Summary In asexual reproduction of the water mold,Saprolegnia ferax, four distinct and sequentially produced spores are involved in dispersal, two of which are motile and two of which are nonmotile. Composition of cell surface glycoproteins may be important in dispersal strategies for each of these stages. Binding patterns of fluorescently labelled lectins were investigated to identify differences in glycoproteins of asexually produced dispersal stages. The pattern of lectin binding to zoospores was diverse. FITC-Con A bound to surfaces of zoospores and membranes of the water expulsion vacuole system, indicating the prescence of mannosyl and glucosyl residues. In zoospores incubated for more than 30 min in FITC-WGA and FITC-GS II. which bind N-acetyl glucosamine, fluorescence was sometimes localized in peripheral, intracellular patches. In shorter incubations, secondary zoospores bound these lectins along the groove region where K-bodies were located. Surfaces of cystospores typically bound FITC-WGA, but not FITC-GS II. FITC-GS II, however, bound to empty cystospore walls, probably because reactive sugars were available at the inner surface of the wall. Germ tubes emerging from cystospores bound labelled WGA and GS II, but not Con A. The same lectin binding pattern was found along discharge papilla of primary cystospores, indicating that modifications in cystospore walls associated with direct germination and zoospore discharge were similar. Thus, glycoproteins involved in early establishment of the hyphal system differ from those forming the cell surface of cystospores. Differences in the binding pattern of lectins to zoospores and cystospores highlight differences between cell surface carbohydrates of motile and nonmotile asexual stages.Abbreviations BPA

Summary Differential scanning calorimetry data obtained from corn embryos is consistent with the hypothesis of their glassy state. This work extends that hypothesis to explain the speculation about the high heat resistance of bacterial spores. By considering the protoplast to be in a glassy solid-state, it can be assumed that the configurational rearrangements of the key life dependent polymer chain backbones (DNA, etc.) are extremely slow, thereby ceasing thermal motions. It is assumed that at the glass transition temperature, the spore protoplast undergoes a discontinuity in the thermal expansion coefficient, and above this critical temperature, the rate of thermal inactivation of spores is free volume dependent and can be described adequately by the William, Landel and Ferry (WLF) equation. Glass transition temperatures forBacillus stearothermophilus andClostridium botulinum spores, obtained by fitting the inactivation rate data to the WLF equation, indicate a decrease in the inactivation rates with increasing glass-transition temperatures. 相似文献

The formation of vesicular-arbuscular mycorrhizae (VAM) in intact soil profiles from two sites in southeastern Australia were measured at two depths using a bioassay grown in intact soil cores. Intact soil cores were taken from (1) topsoil (0–15 cm) and (2) subsoil (15–30 cm) four times during 1990. Seeds of Acacialinifolia (Vent.) Willd. (Mimosaceae) were sown into the cores and plants harvested 8 and 12 weeks after sowing. For 1990, at both sites and in all seasons, VAM most readily developed in the roots of seedlings of A. linifolia grown in topsoil. Limited VAM occurred in roots grown in subsoil cores. Most colonisation of roots by VAM occurred from cores collected during spring and summer. Spore numbers were quantified for each site and depth by wet-sieving 100-g samples of air-dried soil and counting turgid spores containing oil droplets. Three types of spores were found in the soils. Few spores were extracted from all soils sampled, and for the most abundant of the spore types at least twice as many spores occurred in the topsoil than in the subsoil for all seasons examined. As most of the propagules that initiate VAM infection were observed in the topsoil, disturbances which involve the removal and storage of the top 15 cm will adversely affected these fungi. 相似文献

Palynology, which is the study of pollen and spores in an archaeological or geological context, has become a well-established research tool leading to many significant scientific developments. The term palynomorph includes pollen of spermatophytes, spores of fungi, ferns, and bryophytes, as well as other organic-walled microfossils, such as dinoflagellates and acritarches. Advances in plant genomics have had a high impact on the field of forensic botany. Forensic palynology has also been used and applied more recently to criminal investigation in a meaningful way. However, the use of pollen DNA profiling in forensic investigations has yet to be applied. There were earlier uses of dust traces in some forensic analyses that considered pollen as a type of botanical dust debris. Pollen grains can be studied for comparative morphological data, clues to unexpected aspects relating to breeding systems, pollination biology and hybridization. This can provide a better understanding of the entire biology of the group under investigation. Forensic palynology refers to the use of pollen and other spores when it is used as evidence in legal cases to resolve criminal issues by proving or disproving relationships between people and crime scenes. This overview describes the various contributions and the significance of palynology, its applications, different recent approaches and how it could be further employed in solving criminal investigations. 相似文献

Spore sculpture and wall structure of eight Cyathea (Cyatheaceae) species from southern South America were studied using light microscopy (LM), scanning electron microscopy (SEM) and transmission electron microscopy (TEM) techniques. Two layers, i.e. an inner and an outer layer, were observed in the perispore. The inner layer has two strata: the inner stratum is attached to the exospore and composed of rodlets tangentially oriented to the spore surface and randomly intermixed; the outer stratum consists of a three-dimensional network of rodlets with either free or fused distal edges forming spinules. The outer layer is thin, darkly contrasted and covers the rodlets. In most cases, the exospore has two layers and a pitted surface. In Cyathea atrovirens, the exospore surface is smooth, while in C. delgadii and C. myriotricha it is verrucate. The homogeneity of perispore features within the genus Cyathea is evident, while exospore features are heterogeneous. The exospore has different kinds of surface-structures that are of potential interest for assessing evolutionary trends within the group. 相似文献

Aerobiological studies to evaluate allergenic fungal spores in the atmosphere of Riyadh were conducted. Burkard personal volumetric sampler were operated as volumetric viable spore traps at two different sites (Al-Batha, a more developed area in the south and Al-Ulia, a less developed area in the north) in Riyadh. Twice a week samplings were carried out over a period of 12 month. The seasonal fluctuations of the most frequent fungi were plotted as major components. The dominant species at the two sites were members of the genera Alternaria, Aspergillus, Cladosporium, Penicillium and Ulocladium. Drechslera, Fusarium, Rhizopus and Stachybotrytis species were minor components or sporadic. Seasonal variations of the total colonies were significantly (p < 0.05) different. They showed higher concentrations in the winter season and the lowest in summer. The Al-Batha site was always higher in spore concentrations than the Al-Ulia site. The results provide valuable information for the diagnosis and prophylaxis of allergic diseases due to airborne fungi.This revised version was published online in October 2005 with corrections to the Cover Date. 相似文献

The tolerance of indigenous arbuscular mycorrhizal fungi (AMF) to stressful soil conditions and the relative contribution of spores of these fungi to plant colonization were examined in a Portuguese salt marsh. Glomus geosporum is dominant in this salt marsh. Using tetrazolium as a vital stain, a high proportion of field-collected spores were found to be metabolically active at all sampling dates. Spore germination tests showed that salt marsh spores were not affected by increasing levels of salinity, in contrast to two non-marsh spore isolates, and had a significantly higher ability to germinate under increased levels of salinity (20) than in the absence of or at low salinity (10). Germination of salt marsh spores was not affected by soil water levels above field capacity, in contrast to one of the two non-marsh spore isolates. For the evaluation of infectivity, a bioassay was established with undisturbed soil cores (containing all types of AM fungal propagules) and soil cores containing only spores as AM fungal propagules. Different types of propagules were able to initiate and to expand the root colonization of a native plant species, but spores were slower than mycelium and/or root fragments in colonizing host roots. The AM fungal adaptation shown by this study may explain the maintenance of AMF in salt marshes. 相似文献
