Non-obstructive azoospermia (NOA) is the most severe form of male infertility, defined by lack of spermatozoa in the ejaculate caused by impaired spermatogenesis. The chance of biological fatherhood of these men has been improved since the introduction of microdissection testicular sperm extraction (MD-TESE) combined with intracytoplasmic sperm injection. A thorough patient evaluation preoperatively is essential to recognize any underlying conditions, and to assist in patient counseling on the sperm recovery rate and pregnancy results. This review article summarizes the present data on MD-TESE to reach optimal results is treating men with NOA.  相似文献

白念珠菌(Candida albicans)是侵袭性真菌感染的主要致病性病原体。抗菌肽AMP-17有较强的抗念珠菌活性,经AMP-17作用白念珠菌后的蛋白组学结果显示,细胞壁(XOG1)和氧化应激(SRR1)蛋白基因表达差异显著,提示AMP-17可能通过影响XOG1和SRR1基因发挥其抗白念珠菌作用。为进一步探究XOG1和SRR1基因是否为AMP-17的作用靶点,本研究利用规律成簇间隔短回文重复序列相关蛋白9(clustered regulatory interspaced short palindromic repeats-associated protein 9,CRISPR/Cas9)系统构建了白念珠菌xog1Δ/Δ和srr1Δ/Δ缺失菌;表型观察结果发现除XOG1基因缺失可影响白念珠菌的体外应激和菌丝形成,2个基因缺失对白念珠菌生长繁殖和生物膜形成无明显影响;药敏实验分析显示AMP-17对xog1Δ/Δ和srr1Δ/Δ缺失菌的MIC80值从野生菌的8μg/mL增至16μg/mL,而对srr1Δ/Δ::srr1回补菌的MIC80值降至野...  相似文献

Protein supra-domains are defined as recurring arrangements of two or three domains present adjacent to each other along a polypeptide chain. Such combinations have novel functions beyond those of the individual partner domains that compose them, which can exist in isolation. Here, we describe a new type of large supra-domain (∼ 360 residues) in which one of the component partners (∼ 200 residues) appears to be incapable of existing in a context other than immediately adjacent to the C-terminus of the well-characterized Hsp90-like ATPase domain. We found that this supra-domain has a broad phylogenetic distribution, with examples in Archaea, Bacteria, and Eukarya. There is strong selective pressure for this arrangement to occur as part of repeated regions of unprecedented length. We identified multiple strategies of convergent evolution to attain such configurations. In humans, this supra-domain is present in triplicate at the N-terminus of the protein sacsin (4579 residues), mutated in the neurodegenerative disorder known as spastic ataxia of Charlevoix-Saguenay, and thus, we termed it “sacsin repeating region” (SRR). Biochemical characterization demonstrated that SRRs possess ATPase activity, which appears to be a requirement for sacsin function, as a disease-causing mutation leads to an alternate conformation completely incapable of hydrolyzing ATP. We also found evidence of a convergent evolutionary strategy to place SRRs in proteins containing C-terminal J domains, which we demonstrated here to be capable of stimulating the intrinsic ATPase activity of Hsp70. Our sequence and biochemical analyses indicate that SRRs necessitate nucleotide hydrolysis for their function, provided by the common Hsp90 ATPase domain, which, when coupled to the unique adjacent sequence, may give rise to a novel activity related to protein quality control.  相似文献

A dual-band microstrip antenna operating at GSM 900 and GSM 1800 MHz is designed initially. Then a single split ring resonator (SRR) structure is used as a superstrate for this dual-band antenna. A circular current is induced in the SRR due to the perpendicular plane wave excitation, which in turn leads to an electric excitation coupled to the magnetic resonance. It also exhibits higher order excitations at 0.9 and 1.8 GHz which ultimately resulted in specific absorption rate (SAR) reduction of human head at both the designed frequencies of the antenna. The antenna and the SRR superstrate are printed on a 1.6 mm thick FR-4 substrate of dimension 59.6 × 49.6 mm2. Analysis of the SRR using the classic waveguide theory approach is discussed. Radiation pattern of the antenna in the presence of SRR superstrate and human head is also discussed. Prototype of the antenna along with the SRR superstrate is fabricated and measured for return loss and radiation pattern. Measurement results fairly agree with the simulated results. A human head phantom is utilized in the calculation of SAR.  相似文献

The chromatographic behavior of seven 16-oximino derivatives of 3beta-hydropxy-5-androstene have been investigated using the normal-phase (NP) HPTLC chromatographic mode of the type silica-non-polar diluent (benzene)-polar modifier (acetonitrile, ethyl acetate, or dioxane). The linear relationship between the retention constants (R(M)) and the logarithm of the organic modifier content in the mobile phase allowed for the calculation of R(M)0 values. The influence of substituent in the molecule on extrapolated retention data is discussed. To better understand the retention mechanism in the separation of androstene compounds, the functional group contributions (tauX) were compared with Hansch substituent constants (pi). An attempt to quantitate the lipophilicity of the investigated compounds using normal phase thin-layer chromatographic R(M)0 value was made. Also, the relative lipophilicity values determined previously by RPC as well as activity were compared with NPC data.  相似文献

Abstract

Associated microorganisms have been described in numerous marine sponges. Their metabolic activity, however, has not yet been investigated in situ. We quantified for the first time microbial processes in a living sponge. Sulfate reduction rates of up to 1200 nmol cm?3d?1 were measured in the cold-water bacteriosponge Geodia barretti . Oxygen profiles and chemical analysis of sponge tissue and canal water revealed steep oxygen gradients and a rapid turnover of oxygen and sulfide, dependent on the pumping activity of the sponge. Identification of the microbial community with fluorescently labelled oligonucleotide probes (FISH) indicates the presence of sulfate-reducing bacteria belonging to the Desulfoarculus/Desulfomonile/Syntrophus -cluster in the choanosome of this sponge. Analysis of lipid biomarkers indicates biomass transfer from associated sulfate-reducing bacteria or other anaerobic microbes to sponge cells. These results show the presence of an anoxic micro-ecosystem in the sponge G. barretti, and imply mutualistic interactions between sponge cells and anaerobic microbes. Understanding the importance of anaerobic processes within the sponge/microbe system may help to answer unsolved questions in sponge ecology and biotechnology.  相似文献
