Pseudomonas alcaligenes was detected at a high concentration (10 9 -10 10 cells mL -1 ) in the haemolymph of some dead Oryctes rhinoceros grubs collected from its breeding sites in the three southern districts, viz. Alleppey, Quilon and Kottayam of Kerala State, India. In a laboratory colony maintained for production of Oryctes virus, an important biocontrol agent of this major coconut pest, approximately 52% of the grubs succumbed to septicaemia with similar symptoms. The bacterium was found to be a component of the gut microflora of healthy grubs. Occurrence of the viral infection naturally or when induced in the laboratory in the O. rhinoceros grubs, appeared to be one of the biotic stress factors for P.alcaligenes to become an opportunistic pathogen. A preponderance of this bacterial infection in field populations during the periods when natural viral infection in grubs was above average, agrees with this observation. This finding becomes significant as infection by the opportunistic bacterial pathogen, P. alcaligenes , reduces the production of Oryctes virus inoculum in nature and limits the field-perpetuation of this viral biocontrol agent.  相似文献

岩亮洞是广西扶绥地区最近新发现的早更新世巨猿化石点,经过初步发掘,采集到了丰富的巨猿材料及大量伴生的哺乳动物化石。引人注目的是其中犀类化石居多,包括146颗完整的牙齿及十多件头后骨骼化石,从形态特征判断,这些犀类化石应属于独角犀的一新种Rhinoceros fusuiensis。岩亮洞巨猿动物群中的独角犀以种群丰度最大而不同于其他已知东亚地区的巨猿动物群中的独角犀类材料。本文从犀类牙齿的主尖釉质层、齿质暴露程度、咀嚼面的磨蚀程度等方面对岩亮洞独角犀的年龄结构进行了研究。该死亡群中至少有4头幼年个体、5头青年个体、5头成年个体和1头老年个体。依据牙齿磨蚀程度和牙齿萌出顺序对年龄结构的分析及统计显示,其中青年个体和成年个体最多,幼年个体次之,老年个体最少,其死亡的原因不是通常意义上磨耗型死亡,突发性的灾害可能是造成这种死亡年龄分布的主要原因。  相似文献

The relationships between testosterone concentrations in male African rhinoceros and the presence of conspecific males and females were investigated. Serum testosterone concentrations were measured using enzyme-linked immunoassay (EIA) in 37 male black rhinoceros (Diceros bicornis) and 21 male white rhinoceros (Ceratotherium simum) housed at 37 institutions in the USA. Testosterone concentrations in both black (n = 37) and white (n = 21) rhinoceros males rose with increasing numbers of females present (P < 0.05). Average testosterone concentrations also rose with an increased number of conspecific males (n = 34) in black rhinoceros (P < 0.05). However, no specific pattern was found among male white rhinoceros housed with other males. We inferred that introduction of females to a male may play an important role in stimulating libido and spermatogenesis. The similar response of black rhinoceros and white rhinoceros to increased numbers of females suggested that, at least historically, herd structure for blacks may have been more similar to whites than previously realized, and should be investigated further.  相似文献

Mortality rates are high among captive African black rhinoceroses (Diceros bicornis), due to increased susceptibility to disease. The ability to rescue genetic material from individuals that die unexpectedly represents a practical approach to assist ex situ conservation efforts. The objectives of the present study were to attempt postmortem oocyte recovery from ovaries of African black rhinoceroses (N = 6) and to test the efficacy of equine protocols for rhinoceros oocyte IVM and IVF using cryopreserved rhinoceros sperm. The interval from ovary removal to oocyte recovery was 25.3 ± 13.9 h (mean ± SD). Ovaries were transported at 4 °C or 22 °C and effects of temperature on postmortem oocyte competence was evaluated. Numbers of oocytes collected per female averaged 15.8 ± 6.9. In total, 95 oocytes were recovered. Of these, 85 were inseminated using homologous sperm and 10 were inseminated using heterologous sperm. Overall, substantial numbers of viable oocytes were retrieved from African black rhinoceros ovaries 1 to 2 days postmortem from ovaries stored at ambient temperature. A proportion of these oocytes matured and underwent penetration and fertilization by heterologous or homologous frozen-thawed rhinoceros sperm. The reproductive competence of postmortem oocytes was further demonstrated by development of a single two-cell embryo. Despite the need for further refinements, gamete rescue in the rhinoceros has promise for producing rhinoceros embryos, as well as testing sperm functions in vitro.  相似文献

Vertical enamel prism decussation in the inner-layer enamel of rhinoceroses occurs as the result of vertical translation, in opposite senses, of zones of ameloblasts, which begins very shortly after amelogenesis commences at the enamel-dentine junction. Prisms in the centre of the decussating zones are stacked in the Pattern 3 arrangement. Zone boundary prisms adopt intermediate orientations, are locally nearly perpendicular to the enamel surface, and have a cylindrical, Pattern 1 cross-section. Decussation also continues in the outer-layer enamel, but the prisms all have occlusal-going courses: the occlusal-going zones of the inner enamel continue as the more occlusally oriented zones of the outer layer. Abrasion resistance to diamond polishing and soft abrasive projectile erosion (air-polishing with NaHCOs) and resistance to ion beam erosion is greater with distance from the nearest prism boundary discontinuity. Polished surface areas containing longitudinally sectioned prisms are more prone to 'air-polishing' and 'airbrading' erosion than areas with transversely sectioned prisms. These observed relationships fully explain the relief developed at natural wear surfaces.  相似文献

The objective of these studies was to investigate the practicality of flow cytometric sex-sorting for spermatozoa from the white and the black rhinoceros (Ceratotherium simum, Diceros bicornis). In Experiment 1, four semen extenders were tested regarding their suitability for liquid preservation of spermatozoa before sorting. Dilution in MES-HEPES-based semen extender followed by incubation generated best sperm quality parameters (motility, viability, and acrosome integrity). In Experiment 2, the effect of staining method (15 °C for 4 to 6 h during transport or 37 °C for 1 to 1.5 h) on sort efficiency and sperm quality was investigated. Staining at 15 °C during transport resulted in a higher percentage of sperm samples showing a resolution of X- and Y-chromosome-bearing populations (60%) compared with that for staining at 37 °C after transport (33%) and resulted in superior sperm integrity after staining (43.8 ± 11.3% vs. 19.6 ± 12.1%). Sort rate was 300 to 700 cells/sec and sort purity, determined for one sorted sample, was 94% for X-chromosome-bearing spermatozoa. In Experiment 3, the highly viscous component of rhinoceros seminal plasma, which complicates the process of sperm sorting, was examined by gel electrophoresis and mass spectrometry. Results suggested a 250-kDa glycoprotein (most likely originating from the bulbourethral gland) to be responsible for the characteristic viscosity of ejaculates. In Experiment 4, viscosity of seminal plasma, as measured by electron spin resonance spectroscopy, was significantly decreased after addition of α-amylase or collagenase (0.5 and 3 IU per 100 μL seminal plasma, respectively) by 28% and 21%, respectively, with no negative effect on sperm characteristics. The results of this study demonstrate for the first time that rhinoceros spermatozoa can be successfully sorted into high-purity X- and Y-chromosome-bearing populations. Furthermore, the successful liquefaction of viscous ejaculates provides the means to greatly improve sort-efficiency in this species.  相似文献

Skulls of identified individual Asian rhinos from zoos were compared with wild-shot skulls of the same species to detect any captivity-induced changes. Rhinoceros unicornis and Rhinoceros sondaicus are typically stunted by captivity, while Dicerorhinus sumatrensis is not and may even be affected in the opposite manner, becoming giant. Rhinos are the only modern perissodactyls that possess cranial weapons similar to the horns, antlers and ossicones of modern ruminants. Yet, unlike ruminants, there is no clear relationship between sexual dimorphism and sociality. It is possible to extend the study of the coevolution of sociality and sexual dimorphism into extinct rhinos by examining the demographic patterns in large fossil assemblages. An assemblage of the North American early Miocene (∼22 million years ago) rhino, Menoceras arikarense, from Agate Springs National Monument, Nebraska, exhibits dimorphism in incisor size and nasal bone size, but there is no detectible dimorphism in body size. The degree of dimorphism of the nasal horn is greater than the degree of sexual dimorphism of any living rhino and more like that of modern horned ruminants. The greater degree of sexual dimorphism in Menoceras horns may relate to its relatively small body size and suggests that the horn had a more sex-specific function. It could be hypothesized that Menoceras evolved a more gregarious type of sociality in which a fewer number of males were capable of monopolizing a larger number of females. Demographic patterns in the Menoceras assemblage indicate that males suffered from a localized risk of elevated mortality at an age equivalent to the years of early adulthood. This mortality pattern is typical of living rhinos and indicates that young males were susceptible to the aggressive behaviors of dominant individuals in areas conducive to fossilization (e.g., ponds, lakes, rivers). Menoceras mortality patterns do not suggest a type of sociality different from modern rhinos although a group forming type of sociality remains possible. Among both living and extinct rhinos, the severity of socially mediated mortality seems unrelated to the degree of sexual dimorphism. Thus, sexual dimorphism in rhinos is not consistent with traditional theories about the co-evolution of sexual dimorphism and sociality.  相似文献

Eleven novel polymorphic microsatellite loci are presented for the highly endangered Indian (or greater one horned) rhinoceros Rhinoceros unicornis (Mammalia: Rhinocerotidae). These will be used to analyse the genetic variability within and between the two remaining large populations of the Indian rhinoceros and to manage captive breeding.  相似文献
