Summary Electron microscopic examination ofCuscuta odorata, used for transmission trials, revealed mycoplasma-like organisms (MLO) as well as rhabdovirus-like particles, unknown toCuscuta. The virus infection is confined to certain phloem-parenchyma cells and a 1–2 cell thick layer of parenchyma cells with thickened walls surrounding the central cylinder. Virus particles, mostly bacilliform, could be detected mainly in the nucleus but also in the cytoplasm. They reach a length of 350–400 nm and a diameter of approximately 75 nm. Virus assembly takes place exclusively in the nucleus. Virus maturation occurs in membrane bound areas within the nucleus, which have no connection with the perinuclear space. Formation of nucleocapsids is always associated with a nuclear viroplasm. Envelopment of virus particles occurs in these membrane bound areas. Budding into the perinuclear space does not occur. Eight rhabdoviruses from grass and cereal hosts and their antisera were used to examine virus relationships by gel-diffusion serology. Nucleocapsid (Nc) preparations from digitaria striate virus (DSV) and maize sterile stunt virus (MSSV) both contained a major protein of c. 52 OOO daltons, and antisera prepared to these readily discriminated related planthopper-transmitted rhabdoviruses. MSSV showed a moderately close relationship to barley yellow striate mosaic virus (BYSMV) when an antiserum prepared to whole virus was used, but the Nc antiserum showed clearer discrimination. Worthern cereal mosaic virus and DSV showed a distant relationship to BYSMV and MSSV. There was no serological relationship between any of these viruses and cereal chlorotic mottle virus, cynodon chlorotic streak virus, festuca leaf streak virus or maize mosaic virus.  相似文献

利用与N蛋白mRNA3'末端顺序相同的20寡聚核苷酸引物,通过点杂交、限制性内切酶分析从小麦丛矮病毒(WRSV)cDNA文库中筛选到编码N蛋白基因下游顺序的cDNA克隆。序列分析表明,该cDNA片段含有一编码的40kD蛋白的开放读框。将该读框的全长cDNA经PCR扩增后,克隆到pGEX-3X上,在大肠杆菌DE3中用IPTG诱导表达,经蛋白质印迹鉴定,该基因为小麦丛矮病毒NS蛋白基因。  相似文献

从小麦丛矮病毒(WRSV)基因文库含磷蛋白NS基因的质粒中,经亚克隆、测序,得到NS蛋白基因的下游序列,其中含有一读框453nt、编码一分子量约17kD蛋白。用PCR方法获得该读框全长片段并克隆到表达载体pGEX-3X,在大肠杆菌BL21(DE3)菌株中以IPTG诱导表达,产物由谷胱甘肽S-转移酶(GST)亲和层析柱纯化后,用蛋白质印迹分析鉴定,结果表明,该读框为编码病毒基质蛋白M的基因。  相似文献

The rabies virus (RV) phosphoprotein P is a multifunctional protein involved in viral RNA synthesis and in counteracting host innate immune responses. We have previously shown that RV P gene expression levels can be regulated by using picornavirus internal ribosome entry site (IRES) elements. Here we exploited a particular feature of the foot-and-mouth disease virus (FMDV) IRES, namely, preferential initiation at a downstream initiation codon, to address the role of N-terminally truncated RV phosphoproteins usually generated in RV-infected cells through ribosomal leaky scanning. Recombinant RVs in which P synthesis was directed by the poliovirus or FMDV IRES produced full-length P (P1) or a truncated form (P2), as the dominant product, respectively. While the P2 overexpressing virus showed attenuated growth in interferon-incompetent cells, it was superior to the P1 overexpressing virus in preventing expression of host interferon-stimulated genes. This indicates that in RV infected cells the availability of the truncated P2 protein is critical for viral resistance to interferon.  相似文献

牙鲆一株弹状病毒病原的分离与鉴定   总被引:3,自引:0,他引:3  
从患病牙鲆中分离鉴定了一株弹状病毒(Paralichthys olivaceus rhabdovirus,PoRV)。用过滤除菌后的患病牙鲆组织匀浆液,接种不同的鱼类细胞,其中有7种鱼类细胞出现明显的病变在对病毒进行挑斑分离后,测定了PoRV的滴度,显示PoRV在敏感鱼类细胞(Grass Carp Ovary,GCO)中的滴度达到106.5TCID50/mL;绘制了PoRV生长曲线;经蔗糖密度梯度离心提纯PoRV,负染及宿主细胞超薄切片的电镜观察,显示PoRV大小约为60nm×200nm。测定了PoRV的理化性质,显示该病毒对有机溶剂和温度敏感,但对DNA抑制剂阿糖胞苷(Ara-c)不敏感。经SDS-PAGE电泳,对PoRV的蛋白图谱进行了分析,表明该病毒有5种主要蛋白带,其分子量大小分别约为:250kD、67kD、44kD、30kD、23kD。  相似文献

In natural populations of D. melanogaster, a minority of flies are usually infected by the sigma Rhabdovirus, which is not contagious but vertically transmitted. A few parameters have been identified that affect the evolution of the Drosophila — sigma system. Various sets of values of these parameters in males mutually adapted to one another have been established in different populations. This study analyses female characteristics that were previously reputed not to vary much between populations. The newly collected data show that these characteristics (mainly transovarial transmission here) may also be polymorphic in the wild, and may contribute to the diversity of equilibria established between the virus and its host. The observations presented here, obtained from various populations, confirm the complexity of this system of two coevolving organisms: as already noted there seem to be as many different sets of values of parameters as there are populations.  相似文献

Abstract

Initial symptoms of the mottle streak disease on finger millet were observed after only 45 days of sowing. Chlorotic mottling, streaking, striping and yellowing were the common symptoms on leaves and the diseased plants had more nodal branches and unproductive tillers. Among the weather parameters relative humidity and rainfall were positively and wind velocity was negatively correlated with disease incidence and the population of the vector Cicadulina bipunctella. A significant positive correlation existed between the vector population and disease incidence. In the disease management study seed extract of Harpullia cupanioides at 10% was found to reduce the mottle streak incidence by 61.42% with maximum yield of 2.28 t/ha. Acephate 0.01% and neem oil 1% spray were also found to be effective by reducing the disease incidence up to 39.6 and 36.4% with 2.23 and 2.18 t/ha of grain yield respectively. Enhanced activation of defense-related enzymes such as peroxidase (PO), polyphenol oxidase (PPO) and phenylalanine ammonia lyase (PAL) were noticed in AVP sprayed plants compared to control.  相似文献
