Anoplophora glabripennis (Motschulsky) (Coleoptera: Cerambycidae: Lamiini) is an invasive wood‐boring beetle with an unusually broad host range and a proven ability to increase its host range as it colonizes new areas and encounters new tree species. The beetle is native to eastern Asia and has become an invasive pest in North America and Europe, stimulating interest in delineating host and non‐host tree species more clearly. When offered a choice among four species of living trees in a greenhouse, adult A. glabripennis fed more on golden‐rain tree (Koelreuteria paniculata Laxmann) and river birch (Betula nigra L.) than on London planetree (Platanus × acerifolia (Aiton) Willdenow) or callery pear (Pyrus calleryana Decaisne). Oviposition rate was highest in golden‐rain tree, but larval mortality was also high and larval growth was slowest in this tree species. Oviposition rate was lowest in callery pear, and larvae failed to survive in this tree species, whether they eclosed from eggs laid in the trees or were manually inserted into the trees. Adult beetles feeding on callery pear had a reduced longevity and females feeding only on callery pear failed to develop any eggs. The resistance of golden‐rain tree against the larvae appears to operate primarily through the physical mechanism of abundant sap flow. The resistance of callery pear against both larvae and adults appears to operate through the chemical composition of the tree, which may include compounds that are toxic or which otherwise interfere with normal growth and development of the beetle. Unlike river birch or London planetree, both golden‐rain tree and callery pear are present in the native range of A. glabripennis and may therefore have developed resistance to the beetle by virtue of exposure to attack during their evolutionary history.  相似文献

The effect of different extraction methods on the composition of samples of soluble cuticular lipids (SCL) of Citrus aurantium L. was investigated. The variation of extraction yields, when whole leaves were immersed in solvent, was studied as a function of solvent type and duration of immersion. Cuticular waxes were also quantitatively extracted from isolated cuticular membranes of C. aurantium and their composition was compared to that of samples obtained by the immersion method. Significant differences were observed. Higher carbon number homologues of the aliphatic constituent classes were discriminated against when whole C. aurantium leaves were extracted by immersion. The alkyl ester fraction was almost entirely lacking in extracts from whole leaves. The dependence on carbon chain length of the saturation concentrations in chloroform of major aliphatic SCL constituents was determined. The results are discussed in terms of the major physico-chemical processes involved in the extraction of SCL.  相似文献

Summary Dynamics of F-actin organization during activation and germination ofPyrus communis (pear) pollen was examined using rhodaminephalloidin. Prior to activation, the rhodamine-phalloidin labelling pattern appeared as circular profiles in the peripheral cytoplasm of the vegetative cell and as coarse granules around the vegetative nucleus. In activated pollen, parallel arrays of cortical F-actin were aligned circumferentially, along the polar axis in non-apertural areas of the pollen grain, and at 45° to 90° to the polar axis beneath the apertures. Some pollen also showed fluorescent granules or fusiform bodies dispersed throughout the cytoplasm, but as the number of such pollen diminished with prolonged incubation, these are being considered as intermediate patterns. In later stages, the filaments became organized as interapertural bundles traversing the three apertures. However, prior to emergence of the pollen tube, labelling became confined to a single aperture. In germinated pollen grains, actin microfilaments are aligned more or less axially with respect to the axis of the developing pollen tube.The granular labelling pattern seen around the vegetative nucleus prior to pollen activation also became clearly filamentous with pollen activation; this filamentous pattern persisted until germination when it was replaced by cables that aligned longitudinally with respect to the emerging tube axis.The results demonstrate that the organization of actin undergoes considerable changes in the period preceding pollen germination and that microfilament polarization is achieved before pollen germination.  相似文献

Storage of apple fruits ( Pyrus malus L. cv. Golden Delicious) at different temperatures (0, 12 and 35°C) markedly altered the pattern of water freezing in the seeds. Higher temperatures of storage brought about a shift from the sequential to the discontinuous type of freezing in seeds, the low temperature exotherm (LTE) being much more pronounced than the high temperature one (HTE). The occurrence of LTE was highly correlated with embryo death. Fruit storage at higher temperatures also caused a decrease in the threshold super cooling temperature of seeds and of naked embryos, removed from the seed coat and endosperm. The decrease was less pronounced in naked embryos than in intact seeds. The results presented show that the frost resistance of apple seeds relies mainly on the supercooling ability of the embryos and is increased by the presence of seed coat and endosperm. The broad peak of the LTE indicates that, in contrast with other seeds and many super cooling organs, massive ice nucleation in apple seeds occurs within the embryo tissues and that extra organ freezing seems to be of less importance. Therefore, the increased super cooling ability of apple seeds, isolated from fruits stored at higher temperatures, seems to rely on those seed properties that protect embryo cells against heterogeneous ice nucleation.  相似文献

The contribution of the alternative pathway to the respiration of suspension-cultured pear ( Pyrus communis cv. Passa Crasanne) cells was enhanced, often severalfold, within 2 to 4 days following the addition of cycloheximide, actinomycin D, or 2-(4-methyl-2,6-dinitroanalino)- N -methyl propionamide (D-MDMP). Concomitant inhibition of cellular protein synthesis by cycloheximide and actinomycin D was transient and incomplete. However, inhibition by D-MDMP was virtually complete (>97%) and persisted over several days. [35S]-labelling and polyacrylamide gel separation indicated that cycloheximide precluded the appearance of discernable new proteins in mitochondria. Probes with monoclonal antibodies revealed a conservation of alternative oxidase protein levels in the mitochondria of inhibitor-treated cells. The data, appraised within the complexities of cell-culture dynamics, lead to the conclusion that the observed increases in capacity for cyanide-resistant respiration are the consequence, likely indirect, of inhibited protein synthesis with resultant retention and activation of constitutive alternative oxidase.  相似文献

PCR detection of MLOs in quick decline-affected pear trees in Italy   总被引:1,自引:0,他引:1  
Polymerase chain reaction (PCR) amplification, using primers derived from the 16S rRNA gene, followed by restriction fragment length polymorphism (RFLP) analysis with Alu I restriction endonuclease was used to detect myc-oplasma-like organisms (MLOs) associated with pear decline. MLOs were consistently detected in pear trees that suddenly wilted and died within a few days during summer, as well as in pears of the same orchards with symptoms similar to the slow form of pear decline. In both cases the same RFLP pattern was obtained. Declining pear trees were 5 to 8-yr-old cvs Williams, Kaiser and Max Red Bartlett grafted on to Pyrus communis seedling rootstocks. All the orchards affected by quick decline had severe attacks of pear psyllid (Cacopsylla pyri) during the year this study was performed and during the previous year. The results showed the suitability of DNA amplification by the polymerase chain reaction for the detection of pear decline MLOs and established that MLOs can be detected in infected tissues of dead trees.  相似文献

The effects of ionizing radiation, used in post-harvest treatment of fruit and vegetables. were investigated on cultured apple cells ( Pyrus malus L. cv. Royal Red) on a short-term period. Irradiation (2 kGy) induced an increase of passive ion effluxes from cells and a decrease of cell capacity to regulate external pH. These alterations are likely due to effects on plasma membrane structure and function and were further investigated by studying the effects of irradiation on plasma membrane H+-ATPase activity. Plasma membrane-enriched vesicles were prepared and the H+-ATPase activity was characterized. Irradiation of the vesicles induced a dose dependent inhibition of H+-ATPase activity. The loss of enzyme activity was immediate, even at low doses (0.5 kGy), and was not reversed by the addition of 2m M dithiothreitol. This inhibition may be the result of an irreversible oxidation of enzyme sulfhydryl moieties and/or the result of changes induced within the lipid bilayer affecting the membrane-enzyme interactions. Further analysis of the H+-ATPase activity was carried out on vesicles obtained from irradiated cells confirming the previous results. In vivo recovery of activity was not observed within 5 h following the treatment, thus explaining the decrease of cell capacity to regulate external pH.
This rapid irreversible inhibition of the plasma membrane H+-ATPase must be considered as one of the most important primary biochemical events occurring in irradiated plant material.  相似文献

Anthocoris nemorum L. and Anthocoris nemoralis Fabricius (Heteroptera: Anthocoridae) are important predators of insect pests in pome fruit. Females insert their eggs in leaf tissue. The females’ choice of oviposition site is important for the subsequent distribution of nymphs on host plants. Oviposition preference for apple and pear leaves was tested in the laboratory in four experiments (experiments 1–4). In three experiments it was tested whether simulated insect damage to leaves (experiments 5 and 6) or the presence of prey (experiment 7) influenced oviposition preference. The effect of the presence of prey was only tested for A. nemorum on apple leaves. There was a highly significant anthocorid species × plant interaction for the number of eggs laid on apple and pear leaves. Anthocoris nemorum laid more eggs on apple than on pear leaves, while A. nemoralis preferred pear. Anthocoris nemorum's preference for apple increased over the 6‐week period in which experiments 1–4 were performed, from 66% to 91% eggs laid on apple leaves. No change over time in preference was found for A. nemoralis. Across experiments 1–4, the majority of A. nemorum eggs were laid near leaf margins, whereas eggs of A. nemoralis were more commonly found in the leaf centre, 5 mm or more from the margin, with a highly significant leaf region × species interaction. There was no significant difference in preference for leaf side between A. nemorum and A. nemoralis, but there was a highly significant plant × leaf side × experiment interaction. Thus, more eggs were laid on the ventral than on the dorsal side of pear leaves in experiment 4, while significantly more eggs were laid on the dorsal side of apple leaves in experiments 3 and 4. Choice tests between damaged and healthy leaves showed that A. nemorum laid significantly more eggs on the damaged leaves, while A. nemoralis preferred healthy leaves. Anthocoris nemorum showed a near‐significant preference for ovipositing on leaves with eggs of Operophtera brumata (Lepidoptera: Geometridae). The oviposition preferences found correspond to the natural distribution of these predators in apple and pear orchards. The preference of A. nemorum for leaf margins, and of A. nemoralis for the leaf centre as an oviposition site, supports earlier observations. A preference for leaf side for oviposition site has not been reported earlier. Preference for damaged leaves could help A. nemorum to locate prey in a field situation.  相似文献

The phenological growth stages of nashi tree were firstly described here using the BBCH scale. Based on this general scale, nashi phenology showed 8 of the 10 principal stages (0–9): bud, leaf and shoot development, inflorescence emergence, flowering, fruit development, fruit maturity and senescence. A schematic representation of the chronological progression of principal growth stages of nashi is also shown. The codification of the different growth stages is important for correct timing of general orchard management, particularly for disease and pest management. Besides, it will help farmers to efficiently schedule and manage nashi cultivation, as well as to improve knowledge dissemination among scientists around the World.  相似文献

利用SRAP标记分析贵州砂梨资源遗传多样性   总被引:1,自引:0,他引:1  
采用SRAP标记技术,对贵州59份砂梨种质的遗传多样性进行分析。结果表明:(1)筛选的15对SRAP引物共扩增出151个位点,其中120个是多态性位点,多态百分率为79.47%。(2)POPGENE分析表明,贵州砂梨资源在物种水平上有效等位基因数(Ne)为1.355 9,Nei’s基因多样性指数(H)为0.216 9,Shannon’s信息指数(I)为0.336 2;在区域品种群水平Ne=1.261 1,H=0.155 5,I=0.235 2,以黔西南地区资源的遗传多样性最高,六盘水市最低。(3)聚类分析显示,在遗传相似系数0.75处,可将59份砂梨资源分为6个组,聚类分析结果具有一定的地理区域特性。研究认为,贵州砂梨种质具有较为丰富的遗传多样性,并表现出明显的地理区域特性;SRAP标记与ISSR标记在揭示贵州砂梨种质遗传多样性上具有高度的一致性,但SRAP标记能在更小的变异范围内揭示更多的遗传信息。  相似文献
