The acaricidal activity of Eugenia caryophyllata essential oil was evaluated in vitro and in vivo on Psoroptes cuniculi, a mange mite. In vitro, different concentrations of the oil were tested and the observed mites mortality was compared with that observed in untreated and treated (Acacerulen R®) controls. In vivo, six P. cuniculi infected rabbits were topically treated with the oil diluted at 2.5% and compared with untreated and treated control groups of six rabbits each. In vitro, up to the concentration of 0.10% the oil gave highly significant (P < 0.01) percentages of mite mortality respect to the untreated controls, but only up to 0.16% it showed the same efficacy of Acacerulen R®. In vivo, the treatment with the essential oil cured all infested rabbits and no statistical differences were observed respect to the treated control group. Numerically equal groups of between five and ten young Merino and Dorper sheep were artificially infested with sheep scab
mites, Psoroptes ovis. Separate groups of sheep were infested during the early winter of two successive years as well as during the early summer
of the second year. Lesion growth was measured at two-weekly intervals for a period of six weeks during the first winter and
for eight weeks during the second winter and in the summer. The rate of lesion growth and mean lesion size on Merino sheep
was significantly greater than that on Dorper sheep at each two-weekly recording during the winter of both years. The rate
of growth of the lesions did not differ significantly between the two sheep breeds during summer, although, with the exception
of eight weeks post-infestation, the mean lesion size on Merino sheep was significantly larger than that on Dorper sheep at
each assessment.

Three in vitro assays for the evaluation of the hatchability of the eggs of the mite Psoroptes ovis (Hering) (Acari: Psoroptidae) are described. Preliminary trials showed that hatching occurs at very high rates when eggs are incubated under conditions of high humidity, on a liquid medium and in agarose dishes. These three protocols were compared, taking into account the ease of preparation, follow-up and accuracy of counting. The best protocol was found to be the use of agarose dishes. It was accurate, easy to carry out and reproducible for further evaluation of existing or potentially new compounds against both adults and eggs of Psoroptes spp. The acaricidal properties of phoxim and amitraz were then evaluated against eggs and adults using the three protocols. Results showed that for both drugs, in vitro adulticidal activity was complete, whereas the in vitro ovicidal activity was only partial. Nevertheless, efficacy of amitraz against both adults and eggs was shown to be higher than that of phoxim. 相似文献

为了探讨水牛痒螨株和兔痒螨株的分类地位,采用 PCR 技术扩增了四川水牛痒螨株和四川兔痒螨株的第二内部转录间隔区(ITS-2)基因,并与 GenBank 中注册的 5 个国外痒螨株的同源基因进行了比较。序列分析发现:兔痒螨株和水牛痒螨株的序列长度分别为 277 bp 和 281 bp,两序列间存在多处转换、颠换和缺失。四川水牛痒螨株同四川兔痒螨株间及国外痒螨分离株间的 ITS-2 基因同源性较低(87.1%~88.0%); 而四川兔痒螨株与国外痒螨分离株的同源性较高(95.5%~100.0%)。用痒螨 ITS-2 基因构建的 MP,NJ,ME 及 UPGM 树中,兔痒螨株和水牛痒螨株在不同的系统树中其位置比较固定,且两者相距均较远。根据痒螨 ITS-2 基因同源性分析和系统树构建结果以及其他已报道的相关证据,作者认为:兔痒螨株和水牛痒螨株可能为痒螨属 Psoroptes 中两个不同的种,兔痒螨分离株为马痒螨 P. equi ;而水牛痒螨株与来自兔、山羊、绵羊和黄牛等痒螨株亲缘关系较远,可能为痒螨属中的另一个独立有效种。 相似文献

A series of cinnamic acid derivatives and its heteroaromatic ring analogues were synthesized and evaluated for acaricidal activity in vitro against Psoroptes cuniculi, a mange mite. Among them, eight compounds showed the higher activity with median lethal concentrations (LC50) of 0.36–1.07 mM (60.4–192.1 µg/mL) and great potential for the development of novel acaricidal agent. Compound 40 showed both the lowest LC50 value of 0.36 mM (60.4 µg/mL) and the smallest median lethal time (LT50) of 2.6 h at 4.5 mM, comparable with ivermectin [LC50 = 0.28 mM (247.4 µg/mL), LT50 = 8.9 h], an acaricidal drug standard. SAR analysis showed that the carbonyl group is crucial for the activity. The type and chain length of the alkoxy in the ester moiety and the steric hindrance near the ester group significantly influence the activity. The esters were more active than the corresponding thiol esters, amides, ketones or acids. Replacement of the phenyl group of cinnamic esters with α-pyridyl or α-furanyl significantly increase the activity. Thus, a series of cinnamic esters and its heteroaromatic ring analogues with excellent acaricidal activity emerged. 相似文献

Mites are an important group of arthropod pests affecting crops, animals and humans. Despite this, detailed physiological studies on these organisms remain sparse due largely to their small size. Unifying models are required to draw together the diverse information from studies on different groups and species. This paper describes a model for digestion in the parasitic mite, Psoroptes ovis, the causative agent of psoroptic mange or sheep scab disease. The limited information about this species is supplemented with data from other acarines, especially house dust mites and ticks. We review the range of enzymes and allergens found in mites and consider their possible roles in digestion in mites, generally and in particular, P. ovis. Histological studies, enzyme biochemistry and molecular biology and experimental evidence suggest that P. ovis utilises a digestive system reliant upon acid peptidases functioning in a largely intracellular environment. The actions of the digestive enzymes are supplemented by the involvement of bacteria as potential direct and indirect sources of nutrition. It is possible that some extra-corporeal digestion also takes place. The interaction of bacteria and digestive enzymes on the skin surface of the sheep may be responsible for the excessive pathological reactions evident in clinical sheep scab. 相似文献

Feeding and maintaining sheep scab mites off-host is an important step towards achieving a safer, environmentally-friendly
means of controlling sheep scab disease. The test arena developed by Thind and Muggleton (Exp Appl Acarol 22:543–552, 1998)
has been adapted for feeding and maintaining Psoroptes ovis off-host. The test arenas are simple to construct and use, and escape-proof to all stages of mites. The performance of the
test arenas was assessed by determining the survival of the scab mites at 33 and 36°C and >95% r.h. on different diets. With
this test arena the best reported mean survival of 10.3 days was achieved with both females and nymphs at a temperature of
33°C on a diet of sheep serum; the mean maximum survival of females at these conditions was 18.8 days, which is at least a
threefold improvement on previous reports. With further development it may be possible to adapt the test arena for assessing
both slow and fast acting agents for controlling sheep scab.

Arcaricidal properties of decoctions, infusions and macerates of dried flower heads of camomile, Matricaria chamomilla L. (Asterales: Asteraceae) were tested in vitro against the mite Psoroptes cuniculi Delafond (Parasitiformes: Psoroptidae). This mite species is responsible for otoacariasis in domestic animals. Mites were exposed to the extracts for 24, 48 or 72 h. All the extracts tested showed highly significant acaricidal activity when compared with controls. Among them, a decoction of 10% was the only formulation which gave 100% activity at all the three observations times. 相似文献

Serum from successful vaccine trials against the sheep scab mite, Psoroptes ovis, was used to immunoscreen a cDNA library constructed from mixed-stage and gender P. ovis to identify potential recombinant vaccine candidates. Immunodominant recombinant proteins recognised by IgG in these sera were selected for further analysis. Two candidates were identified in this way; a catchin-like protein (CLP) and a novel mu class glutathione S-transferase (GST). Both candidates were expressed in bacteria as recombinant proteins, the GST as an active enzyme, and combined with four other recombinant allergens in a multi-component recombinant vaccine. Strong serum IgG responses were induced in sheep against each of the components of the recombinant vaccine, however, the protective efficacy of the vaccine could not be determined because of variability in the establishment of a challenge infection. 相似文献
