Summary The formation and subsequent growth of roots by cuttings of poinsettia, hydrangea, rose and azalea in various propagation media, Jiffy-7, Jiffy-9 and Grodan under different conditions of aeration was investigated. The interrelationships of the effects of air content of the media, temperature and light intensity on the rooting of poinsettia cuttings was also studied.With low air contents (0 cm moisture tension) in the propagation media the formation and growth of roots was strongly inhibited. The rooting performance of rose appeared to be less affected by the poor aeration. Increasing air content improved rooting but best results were obtained at moisture tensions of 4 to 8 cm. Rooting seems to be better correlated with oxygen diffusion rate (ODR) than with air content.For poinsettia cuttings the optimum temperature for rooting was 24 to 28°C. At low temperatures rooting was delayed while at higher temperatures it was almost completely inhibited. Callus formation increased with temperature but decreased with increasing moisture tension. Conditions which induced large callus formation inhibited root formation.High light intensity during rooting reduced overall rooting performance and the inhibition was most pronounced in conjunction with high moisture tensions.Report No. 255.  相似文献

Summary The air content in three types of propagation media, Jiffy-7 and Jiffy-9 which are Sphagnum peat and Grodan which is rockwool, were investigated when they were held at moisture tensions of 0,6 and 12 cm measured from the base of the media. At 0 cm tension the air content (vol. %) was highest in Jiffy-9 and lowest in Jiffy-7. At 12 cm tension the air content was higher in Grodan than in Jiffy-9 and Jiffy-7. Oxygen diffusion coefficients (ODC) and oxygen diffusion rates (ODR) were measured at the different air contents. At air contents below 20 vol. % ODC was about the same for Jiffy-9 and Grodan but at air contents above 20 vol.% it was larger for Jiffy-9 than for Grodan. The oxygen diffusion rate was measured at 0, 4 and 8 cm moisture tension. At all tensions it was approximately 20% higher in Jiffy-9 than in Grodan and Jiffy-7. The ODR in Jiffy-7 and Grodan were affected equally at the same tension, although Grodan contained more air. Report no 253  相似文献

Comparisons between related species with different rooting capacities can provide insights into the mechanisms controlling adventitious root development. The availability of carbohydrates is often considered exclusively as an energetic requirement to drive root development; the major regulatory role in the process is often attributed to phytohormones, particularly auxin. The roles of light quantity (irradiance) and carbohydrate supply available to young aseptic donor-plants on the adventitious rooting response of Eucalyptus globulus (rooting recalcitrant) and Eucalyptus saligna (easy-to-root) were examined. The effects of the type of carbohydrate supply (sucrose or glucose) on the rooting response of cuttings was also evaluated. Light intensity supplied to mother-plants (30 or 60 mol m–2 s–1) had limited influence on the rooting response of both species, whereas dark periods were detrimental, particularly for E. globulus. In E. globulus, rooting was promoted by the absence of sucrose in donor-plant media. Presence of sucrose in donor plant medium promoted root number but did not affect rooting percentage of E. saligna. A positive effect of glucose on cutting rhizogenesis was found if this hexose was supplied during the root induction phase, followed by sucrose in the root formation step, especially for E. globulus. The same effect was not seen with fructose. The beneficial effect of glucose in the induction phase on root number was also evident under suboptimal auxin concentrations.  相似文献

根据高效培养鸡传染性法氏囊病毒(IBDV)时对病毒滴度检测的需要,本文针对BDV的VP4基因的保守序列设计并合成了一对引物,以所构建的重组质粒作为阳性标准品,建立了检测IBDV核酸载量的SYBR Green I荧光定量实时RT-PCR(qRT-PCR)方法,结果表明,其Ct值与标准品模板在4.03×1E1~1E9 拷贝/μL范围内呈良好的线性关系,对IBDV核酸的最低检出量为40拷贝/μL,灵敏度是常规RT-PCR检测方法的1000倍;该方法不与其它禽源病毒发生交叉反应,批内变异系数小于0.05%。应用本方法对DF-1细胞中IBDV的增殖滴度进行了测定,并与经典的TCID50测定方法进行了比较。结果显示两种方法测定的IBDV在微载体悬浮培养和方瓶静态培养条件下DF-1细胞上的增殖曲线都具有一定的平行关系,且qRT-PCR方法比TCID50方法更加快速和敏感,更适合于对IBDV增殖滴度的实时快速测定。  相似文献

As one large class of non-coding RNAs(nc RNAs), long nc RNAs(lncRNAs) have gained considerable attention in recent years. Mutations and dysfunction of lnc RNAs have been implicated in human disorders. Many lnc RNAs exert their effects through interactions with the corresponding RNA-binding proteins. Several computational approaches have been developed, but only few are able to perform the prediction of these interactions from a network-based point of view. Here,we introduce a computational method named lnc RNA–protein bipartite network inference(LPBNI). LPBNI aims to identify potential lnc RNA–interacting proteins, by making full use of the known lnc RNA–protein interactions. Leave-one-out cross validation(LOOCV) test shows that LPBNI significantly outperforms other network-based methods, including random walk(RWR)and protein-based collaborative filtering(Pro CF). Furthermore, a case study was performed to demonstrate the performance of LPBNI using real data in predicting potential lnc RNA–interacting proteins.  相似文献

综述了文冠果 (XanthocerassorbifoliaBunge .)的落花落果、雄性不育、遗传变异、快繁及药用等方面的研究进展。认为造成文冠果“千花一果”的生理原因可能在于树体营养不足 ;导致雄性不育的主要原因可能在于花药难以开裂散粉 ,其次才是花粉败育 ;文冠果花瓣等突变体的发现及体细胞胚培养成功 ,为优选、快繁新品种及研究木本植物的各个器官的形态建成分子机理等提供了条件 ,但文冠果的资源在逐年减少 ,有待尽快加以保护和利用 ;文冠果的药用价值尚有待国内深入系统地研究  相似文献

While a decreasing spectral content of surface electromyography reflects low back muscle fatigue development, reliability of these decreases may be insufficient. Decreasing frequency content is largely determined by decreasing average motor unit action potential conduction velocities (CV), which is considered a more direct measure of muscle fatigue development. However, for the low back muscles it has been proven difficult to identify propagating potentials and consequently estimate the CV. The aim of this study was to estimate the low back muscle CV from high-density multi-channel electromyography by using peak-delay and cross-correlation methods. Fourteen healthy male participants without a history of low-back pain performed a 30 degrees lumbar flexion trial until exhaustion while standing. For 10 out of the 14 participants (118 out of 560 sites) realistic CV estimates were obtained using both methods, the majority likely over the iliocostalis lumborum muscle. Between-method CV differences appeared to be small. Close to the spine a considerable number of sites (79) yielded systematically overestimated low back muscle CV values. Estimating low back muscle CV may allow additional insight into low back muscle fatigue development and potentially improve its monitoring using (high-density) surface electromyography.  相似文献

Cotyledon explants of Korean ginseng (Panax ginseng C. A. Meyer) produced somatic embryos directly on growth regulator-free medium. Somatic embryos developed as either multiple or single-state forms, depending on the degree of maturity of the cotyledons. Cotyledon explants from midmature zygotic embryos formed multiple embryos, while cotyledons from fully mature zygotic embryos formed single embryos. Somatic single embryos regenerated into normal plantlets with both roots and shoots, while multiple embryos did not produce roots but regenerated only into multiple shoots. In full-strength MS basal medium, the root growth of plantlets derived from single embryos was weak compared to that of shoots. Deletion of ammonium nitrate from the MS medium promoted the root growth of the plantlets. The ginseng plants with well-developed shoots and roots regenerated from single embryos were successfully acclimatized in a greenhouse when they were planted in soil. 苦草是水鳖科(Hydrocharitaceae)苦草属(Vallisneria)沉水草本植物,无直立茎,叶基生,线形或带形,常被称为扁担草、蓼萍草、水韭菜、面条草、扁子草和鸭舌条等,广泛分布于世界各地。本属植物有6~10种〔1,2〕,我国的苦草属植物依《中国植物志》第8卷有3种——苦草(V.natansHara)、刺苦草(V.spinulosaYan)和密刺苦草(V.denseserrulataMakino),而没有V.spiralisL.,VasiaticaMiki和V.giganteaGraebner等种〔1〕。本文苦草属植物的命名以《中国植物志》第8卷为准,引用文献一般尊重原作者的意见。下文中,无具体说明,苦草均为苦草属植物的通…  相似文献
