The modulation effects of d-amphetamine and procaine on the spontaneously generated action potentials were studied on the RP1 central neuron of giant African snails (Achatina fulica Ferussac). Extra-cellular application of d-amphetamine or procaine reversibly elicited bursts of potential (BoP). Prazosin, propranolol, atropine or d-tubocurarine did not alter the BoP elicited by either d-amphetamine or procaine. KT-5720 or H89 (protein kinase A inhibitors) blocked d-amphetamine-elicited BoP, whereas they did not block the procaine-elicited BoP. U73122, neomycin (phospholipase C inhibitors) blocked the procaine-elicited BoP, whereas they did not block the d-amphetamine-elicited BoP in the same neuron. These results suggest that BoP elicited by d-amphetamine or procaine were associated with protein kinase A and phospholipase C activity in the neuron.  相似文献

旨在探讨盐酸普鲁卡因(procaine,PCA)对人结肠癌HT-29细胞Syk基因甲基化及表达的影响。应用巢式双重甲基化特异性聚合酶链反应(Methylation-specific PCR,MSP)检测盐酸普鲁卡因处理前后HT-29细胞中Syk基因启动子的甲基化水平。逆转录-聚合酶链反应(RT-PCR)和蛋白印迹技术观察HT-29细胞内Syk基因表达情况。MSP检测发现人结肠癌HT-29细胞中Syk基因存在甲基化,经盐酸普鲁卡因处理能够使Syk基因甲基化水平下降。RT-PCR和蛋白印迹分析结果显示,人结肠癌HT-29细胞经盐酸普鲁卡因处理后Syk基因表达上调。人结肠癌HT-29细胞中Syk基因启动子甲基化导致基因表达沉默,盐酸普鲁卡因能逆转Syk基因启动子区域CpG岛甲基化,使Syk基因活化并表达上调,提示盐酸普鲁卡因具有治疗结肠癌的潜在应用价值。  相似文献

A rapid and sensitive method for the extraction and quantification of penicillin-G and procaine in horse urine and plasma samples has been successfully developed. The method involves the use of solid-phase extraction (SPE) for penicillin-G, liquid–liquid extraction (LLE) for procaine, and high-performance liquid chromatography (HPLC) for the quantification of penicillin-G and procaine. The new method described here has been successfully applied in the pharmacokinetic studies of procaine, penicillin-G and procaine–penicillin-G administrations in the horse.  相似文献

We have made density functional calculations and molecular dynamics (MD) simulations to investigate the structure and pharmacological action of local anesthetics: tetracaine, procaine and lidocaine. The MD simulations were made in a NPT ensemble, in a 0.9% NaCl solution, on both unprotonated and protonated forms of the molecules. The radial distribution function was used to study solvent effects in different regions of the molecules. Although all three anesthetics have different degrees of hydrophobicity, the amino-terminals were the mostly affected by the protonation yielding hydrophilic regions. The charged amino-esters present hydrophilicity on the ester as well as amine terminals. Cl from the solvent solution forms hydrogen bonds via protonated hydrogen attached to nitrogen, yielding neutral molecules, which could, in principle, penetrate the membranes and loose Cl to act in the protonated form. Density functional theory calculations indicated a change in the electrostatic potential and showed that Cl weakly binds to the amine hydrogen, what suggests it is a favorable interaction and supports the existence of the hydrochloric forms of these local anesthetics.  相似文献

A photoaffinity probe, procaine azide, was employed to determine the sites of interaction of procaine in normal and sickle cell erythrocytes. Studies show that the number of binding sites and affinity of procaine to membranes derived from normal and sickled cell erythrocytes were similar, although procaine retards the in vitro formation of irreversibly sickled cells from cells. The results show that procaine azide, a photoaffinity analogue of procaine, is covalently incorporated into both protein (60–70%) and lipid (40–30%) components of the membrane. Sodium dodecyl sulfate-gel electrophoresis of the labeled ghosts show that procaine binds specifically to band 3 and periodic acid-Schiff staining bands in membranes derived from labeled erythrocytes. Binding of procaine or covalent incorporation of procaine azide into membrane proteins does not affect the phosphate transport. Moreover, pre-treatment of intact erythrocytes with 4,4′-diisothiocyano-2,2′-stilbene disulfonate, an anion transport inhibitor, did not affect either the binding or covalent incorporation of procaine azide into erythrocytes. These results indicate that the binding of procaine azide to Band 3 protein occurs at a locus different than that involved in anion translocation process.  相似文献

X-ray diffraction studies were made on lecithin/cholesterol multilayers with very high water content and containing the local anaesthetic procaine. Narrow-angle diffraction experiments show that the procaine molecules are located with the uncharged aromatic amine group approx. 10 Å from the centre of the bilayer. The polar tertiary amine group of these molecules is almost certainly located in the polar head-group region of the membrane. Wide-angle diffraction experiments show that the incorporation of procaine molecules into such lipid membranes produces an approx. 30% increase in the spread of acyl chain separation, although the average spacing between the chains is slightly reduced.  相似文献

We examined the effects of local anesthetics lidocaine and procaine on glutamatergic spontaneous excitatory transmission in substantia gelatinosa (SG) neurons in adult rat spinal cord slices with whole-cell patch-clamp techniques. Bath-applied lidocaine (1-5 mM) dose-dependently and reversibly increased the frequency but not the amplitude of spontaneous excitatory postsynaptic current (sEPSC) in SG neurons. Lidocaine activity was unaffected by the Na+-channel blocker, tetrodotoxin, and the TRPV1 antagonist, capsazepine, but was inhibited by the TRP antagonist, ruthenium red. In the same neuron, the TRPA1 agonist, allyl isothiocyanate, and lidocaine both increased sEPSC frequency. In contrast, procaine did not produce presynaptic enhancement. These results indicate that lidocaine activates TRPA1 in nerve terminals presynaptic to SG neurons to increase the spontaneous release of l-glutamate.  相似文献

The present study describes the simultaneous determination of seven different kinds of local anesthetics and one metabolite by GC–MS with solid-state extraction: Mepivacaine, propitocaine, lidocaine, procaine (an ester-type local anesthetics), cocaine, tetracaine (an ester-type local anesthetics), dibucaine (Dib) and monoethylglycinexylidide (a metabolite of lidocaine) were clearly separated from each other and simultaneously determined by GC–MS using a DB-1 open tubular column. Their recoveries ranged from 73–95% at the target concentrations of 1.00, 10.0 and 100 μg/ml in plasma, urine and water. Coefficients of variation of the recoveries ranged from 2.3–13.1% at these concentrations. The quantitation limits of the method were approximately 100 ng/ml for monoethylglycinexylidide, propitocaine, procaine, cocaine, tetracaine and dibucaine, and 50 ng/ml for lidocaine and mepivacaine. This method was applied to specimens of patients who had been treated with drip infusion of lidocaine, and revealed that simultaneous determination of lidocaine and monoethylglycinexylidide in the blood and urine was possible.  相似文献

Procaine or tetrodotoxin microinjected into the midbrain increased temperatures of the interscapular brown adipose tissue (BAT) and rectum. l-Glutamate microinjected into the same area decreased these temperatures. Neurons in the inferior olive (IO) expressed c-fos following body temperature increase with midbrain procaine. Electrical stimulation of and glutamate injection into the IO increased body temperature, and lO lesions blocked the midbrain procaine-induced body temperature increase. Since decerebration of the hypothalamus had no effect on the temperature increase, it was concluded that neurons in the midbrain tonically inhibit BAT thermogenesis via IO neurons independently of the hypothalamus.  相似文献
