棒头草幼穗在含2,4-D的MS培养基上诱导出了胚性、非胚性和中间型愈伤组织。根据形态、淀粉粒等指标可将组成这些愈伤组织的细胞分为三类。改变培养基中2,4-D的浓度,能诱导三类愈伤组织相互转变。从胚性愈伤组织中诱导形成了大量体细胞胚;体细胞胚是从单个原胚细胞直接发育而来,它们能正常萌发、再生小植株。这种再生能力现已保持了34个月。小植株移植在土壤中可以正常生长、分蘖、开花和结实。  相似文献

Summary The concentration, uptake and element use efficiency of N, P and K in one C3 annual (Polypogon monspeliensis) and two C4 (Echinochloa colonum, an annual, andDichathium annulatum, a perennial) grasses were determined during winter and summer seasons in monocultures raised in field plots at three moisture levels,viz. full, half and one-fourth of field capacity. At each moisture regime the plants were clipped thrice at moderate and severe levels corresponding to 40 and 80% of live green. The concentration of these elements was characteristic of the growth habit of these plants;e.g. the build up of concentration was maximum in leaf of the annuals while it was comparable in crown and leaf of Dichanthium. The N level was maximum in Polypogon. The nutrient use effiency was comparable in the two annuals and maximum K and N use were obtained in Polypogon and Dichanthium, respectively.  相似文献

Gene flow is the most frequently expressed public concern related to the deregulation of transgenic events ( Snow 2002 ; Ellstrand 2003 ). However, assessing the potential for transgene escape is complex because it depends on the opportunities for unintended gene flow, and establishment and persistence of the transgene in the environment ( Warwick et al. 2008 ). Creeping bentgrass (Agrostis stolonifera L.), a turfgrass species widely used on golf courses, has been genetically engineered to be resistant to glyphosate, a nonselective herbicide. Outcrossing species, such as creeping bentgrass (CB), which have several compatible species, have greater chances for gene escape and spontaneous hybridization (i.e. natural, unassisted sexual reproduction between taxa in the field), which challenges transgene containment. Several authors have emphasized the need for evidence of spontaneous hybridization to infer the potential for gene flow ( Armstrong et al. 2005 ). Here we report that a transgenic intergeneric hybrid has been produced as result of spontaneous hybridization of a feral‐regulated transgenic pollen receptor (CB) and a nontransgenic pollen donor (rabbitfoot grass, RF, Polypogon monspeliensis (L.) Desf.). We identified an off‐type transgenic seedling and confirmed it to be CB × RF intergeneric hybrid. This first report of a transgenic intergeneric hybrid produced in situ with a regulated transgenic event demonstrates the importance of considering all possible avenues for transgene spread at the landscape level before planting a regulated transgenic crop in the field. Spontaneous hybridization adds a level of complexity to transgene monitoring, containment, mitigation and remediation programmes.  相似文献

研究了谷子原生质体看护培养中的一些问题。结果表明:不同种植物愈伤组织做看护细胞对谷子原生质体培养植板率有不同的影响。用光头种草愈伤组织对谷子胚性、非胚性或中间型意伤组织的原生质体进行看护培养,以对胚性意伤组织原生质体的效果最好。看护培养主要是作用于原生质体形成完整细胞后的生长发育。试验没有观察到明显的看护细胞数量效应。  相似文献

The role of rhizosphere bacteria in facilitating Se and Hg accumulation in two wetland plants, saltmarsh bulrush (Scirpus robustus Pursh) and rabbitfoot grass (Polypogon monspeliensis (L.) Desf.), was studied. Ampicillin-amended plants (i.e., with inhibited rhizosphere bacteria) supplied with Na2SeO4 or HgCl2 had significantly lower concentrations of Se and Hg, respectively, in roots than plants without ampicillin. These results were confirmed by inoculating axenic saltmarsh bulrush plants with bacteria isolated from the rhizosphere of plants collected from the field; these plants accumulated significantly more Se and Hg compared to axenic controls. Therefore, rhizosphere bacteria can increase the efficiency of Se and Hg phytoremediation by promoting the accumulation of Se and Hg in tissues of wetland plants. The question of whether annual weeds are allelopathic under natural conditions still remains to be critically answered. Investigations were carried out to understand the involvement and mode of operation of allelopathy in an annual weed, Polypogon monspeliensis. Comparative studies of soils associated with and without the weed under field conditions revealed that there was no significant difference in toxicity of the two soils, and thus the possibility of its allelopathic effect on crops grown in the same season could be ruled out. However, soil amended with weed straw had significantly higher total phenolics including higher relative concentrations of phenolic fractions that were not detected in unamended soil. Phenolic fractions significantly affected the seedling growth of radish and cluster bean. It is likely that P. monspeliensis did not interfere chemically with the crops cultivated during the same season, but interfered with the following season crop through incorporated straw. These results indicate how a monocarpic annual such as P. monspeliensis can be allelopathic under field conditions and allelopathic potential can be managed. We suggest that before detailed investigations on allelopathy are performed as per earlier recommended protocols, data on weed life cycle pattern and agricultural practices should be collected.  相似文献

Abstract

Mercury has been extracted in Queretaro, Mexico since the 1960s. The mining wastes were open-air disposal and these mercury wastes have polluted the zone. The aim of this research was to evaluate mercury's fate in lab scale terrestrial reactors considering the following mercury species: soluble, interchangeable, strongly bound, organic, and residual ones. Soils were sampled in two former mines of Pinal de Amoles, Queretaro, Mexico (N 20° 58′ to 21° 21′ and West 99° 26′ to 99° 43′) with initial mercury concentrations were 424?±?29 and 433?±?12?mg?kg?1 for “La Lorena” and “San Jose” former mines, respectively. Two vegetal species Polypogon monspeliensis and Cyperus odoratus were used and 20 reactors were constructed for the lab process. Total mercury was removed to 49–79% from both soils. Mercury elemental, exchangeable, and organic species had the most removal or exchange in the process. Metal uptake, by the plants, was of 5–6% for P. monspeliensis and 5–15% for C. odoratus. Also, mercury fate was estimated to the atmosphere to be 3.3–4.5?mg m?2 h?1 for both soils.  相似文献
