Summary Oxidative and peroxidative activities were localized at the ultrastructural level in suspension cells of an anthocyanin-producing strain of carrot after treatment with dihydroxyphenylalanine (DOPA) and diaminobenzidine (DAB). In DOPA-treated cells a reaction ascribed to polyphenoloxidase (PPO) occurred in the thylakoids of plastids. After DAB treatment at pH 9.0 reactions occurred in microbodies and plastid thylakoids; after treatment at pH 6.8 additional reactions occurred in the mitochondrial cristae and cytoplasmic ground substance. No reaction occurred in the cell walls at either pH. A reaction could not be unequivocally detected in the vacuoles because of the natural occurrence of osmiophilic material. Application of peroxidase and PPO inhibitors indicated that four distinct systems were involved in the DAB reactions: catalase was correlated with the reaction in the microbodies, peroxidase with the reaction in the cytoplasmic ground substance, cytochromes with the mitochondrial reaction, and PPO with the reaction in the thylakoids of the plastids.Contribution No. 1964 of the Rhode Island Agricultural Experiment Station.  相似文献

3-hydroxykynurenine as a substrate/activator for mushroom tyrosinase   总被引:1,自引:0,他引:1  
3-Hydroxykynurenine is a tryptophan metabolite with an o-aminophenol structure. It is both a tyrosinase activator and a substrate, reducing the lag phase, stimulating the monophenolase activity, and being oxidized to xanthommatin. In the early stage of monophenol hydroxylation, catechol accumulation takes place, whereas 3-hydroxykynurenine is substantially unchanged and no significant amounts of the o-quinone are produced. These results suggest an activating action of 3-hydroxykynurenine toward o-hydroxylation of monophenols. 3-Hydroxykynurenine could therefore well act as a physiological device to control phenolics metabolism to catechols and quinonoids.  相似文献

The seedless grapes BRS Clara and BRS Morena, developed in Brazil, are currently growing in popularity due to their premium texture and taste. However, there are no reports on the polyphenoloxidase (PPO) from these cultivars. In this paper, active and latent PPO from BRS Clara and BRS Morena seedless grapes were extracted using the non-ionic detergents Triton-X-100 (active) and Triton-X-114 (latent), and their catecholase activities were characterized. The PPO extracted using Triton-X-110 exhibited maximum activities at pH 6.0 and at 25 °C. Above 30 °C, a gradual decline in activities was noted, with complete inactivation at 60 °C. The PPO from grapes extracted with Triton-X-114 was activated with 0.2% of the ionic detergent sodium dodecyl sulfate (SDS), and exhibited maximum activities at pH 5.5 and at 30 °C. It was stable until the temperature reached 60 °C.  相似文献

In plants, PPO has been related to defense mechanism against pathogens and insects and this role was investigated in coffee trees regarding resistance against a leaf miner and coffee leaf rust disease. PPO activity was evaluated in different genotypes and in relation to methyl-jasmonate (Meja) treatment and mechanical damage. Evaluations were also performed using compatible and incompatible interactions of coffee with the fungus Hemileia vastatrix (causal agent of the leaf orange rust disease) and the insect Leucoptera coffeella (coffee leaf miner). The constitutive level of PPO activity observed for the 15 genotypes ranged from 3.8 to 88 units of activity/mg protein. However, no direct relationship was found with resistance of coffee to the fungus or insect. Chlorogenic acid (5-caffeoylquinic acid), the best substrate for coffee leaf PPO, was not related to resistance, suggesting that oxidation of other phenolics by PPO might play a role, as indicated by HPLC profiles. Mechanical damage, Meja treatment, H. vastatrix fungus inoculation and L. coffeella infestation caused different responses in PPO activity. These results suggest that coffee resistance may be related to the oxidative potential of the tissue regarding the phenolic composition rather than simply to a higher PPO activity.  相似文献

对光叶楮扦插生根过程中吲哚乙酸氧化酶(IAAO)、多酚氧化酶(PPO)、过氧化物酶(POD)3种酶进行了动态跟踪分析。结果表明:IAAO活性在扦插初期逐渐上升,第10d上升到高峰,之后下降再上升,第30d达到新高峰,然后迅速下降;前25d POD活性变化规律与IAAO相似,但30d以后活性一直上升;PPO活性在扦插前期缓慢上升,第20d上升到了最高点,此后变化不大。还研究了IAAO、PPO、POD与不定根的发生和发展关系,认为光叶楮扦插生根可分为愈伤组织形成期、根诱导期和根的伸长期3个阶段,愈伤组织形成期3种酶活性都呈上升趋势,根诱导期IAAO和POD的活性达到高峰;而根伸长期IAAO和POD活性下降,PPO活性上升。  相似文献

Induced resistance of the mountain birch against its main defoliator Epirrita autumnata is a well-characterized phenomenon. The causal mechanism for this induced deterioration, however, has not been unequivocally explained, and no individual compound or group of traditional defensive compounds has been shown to explain the phenomenon. Phenolic compounds are the main secondary metabolites in mountain birch leaves, and the biological activity of phenolics usually depends on their oxidation. In this study, we found that the activity of polyphenoloxidases (PPOs), enzymes that oxidize o-diphenols to o-diquinones, was induced in trees with introduced larvae, and bioassays showed that both growth and consumption rates of larvae were reduced in damaged trees. PPO activity was negatively associated with both larval growth and consumption rates in trees with bagged larvae, but not in control trees. Our results suggest that the oxidation of phenolics by PPOs may be a causal explanation for the rapidly induced resistance of mountain birch against E. autumnata. This finding also helps to explain why correlations between insect performance and phenolics (without measuring indices explaining their oxidation) may not produce consistent results.  相似文献

Abstract

A search for plant activators capable of inducing systemic resistance in sugarcane showed that plants pre-treated with synthetic signal inducers confer a high degree of resistance to Colletotrichum falcatum – the red rot pathogen. Among the various treatments, Acibenzolar S- methyl (ASM) was found to be very effective in restricting the pathogen colonization inside the inoculated cane stalk tissues. The induction of resistance was accompanied by a significant increase in peroxidases and polyphenoloxidases activities. A considerable decrease of pathogen titre in the pre-treated tissues as determined by ELISA, clearly demonstrated the restriction of pathogen colonization and proliferation in the sensitized cane stalks. Specific induction of new isoforms of peroxidases and polyphenoloxidases in C. falcatum elicitor treatment indicates the pathogen elicitor induced specific cellular response of sugarcane suspension-cultured cells.  相似文献

凤眼莲及其根际细菌的相互关系明显地受营养条件的影响,生长在含糖培养基中的凤眼莲根分泌物不改变其根际细菌的生长动态,但在含酚和无机营养条件下生长的凤眼莲根分泌物分别使其根际细菌出现2次和3次增长;而且在含酚环境中,根际细菌对凤眼莲表现出更为强烈的根际效应.细菌的代谢产物能提高凤眼莲体内的多酚氧化酶的活性,却抑制过氧化物酶的活性和减少其体内酚的富集量;然而接菌后的凤眼莲,其体内多酚氧化酶和过氧化物酶同时得到激活,且酚富集量增加.  相似文献
