Enumeration of denitrifying microbial populations in turf   总被引:2,自引:0,他引:2  
Summary Denitrifer populations of a silt and silt loam soil under a Kentucky bluegrass turf were enumerated using the most probable number (MPN) procedure. The influence of soil texture, soil depth, soil moisture, and additions of nitrate fertilizer on denitrifier populations were determined. Saturated soil conditions increased denitrifier populations 87-fold in the silt soil and 121-fold in the silt loam soil. Denitrifier populations did not differ significantly between soil depths and additions of fertilizer nitrate did not influence populations.  相似文献

Isozymes and random amplified polymorphic DNA (RAPD) markers were used for precocious identification of non-maternal plants in progenies of the facultative apomict Poa pratensis. Four progenies obtained from controlled crosses that showed different degrees of apomixis on isozyme analysis of phospho-gluco-isomerases, esterases and peroxidases were chosen for RAPD analysis to generate genomic fingerprints using species-specific primers. At an advanced vegetative stage, a morphological analysis was also performed and characteristics related to growth habit and leaf morphology were observed and recorded. On the basis of the isozyme and RAPD electrophoretic pattern and the morphological appearance, each plant was classified as maternal or aberrant. All three classes of genetic markers employed were able to identify plants that exhibited aberrant traits in the four progenies. Overall, the results of RAPD analysis supported those of isozyme and morphology studies. However, in each progeny, some plants which both isozyme and morphological analyses distinguished as of maternal origin were aberrant according to RAPD analysis. Therefore, the RAPD method proved the most precise screening technique. The greater cost of the molecular approach was offset by its higher accuracy. The use of either three isozyme systems or six primers for PCR amplification seems to be sufficient for reliable estimation of the degree of apomixis. Histological analyses were carried out and the aposporic development of the plant material studied.  相似文献

A field experiment encompassing both neighbour- and nutrient-manipulations was conducted in a nutrient-impoverished old-field habitat to investigate how the intensity of plant competition was affected by soil nutrient level. Three perennial grasses were used as target species: Agropyron repens, Poa pratensis and Phleum pratense. Neighbour manipulations involved the removal (through herbicide application) of all neighbouring vegetation within a 20 cm or 40 cm radius around target plants. Target performance was measured under five levels of added nutrients (N-P-K) in both the neighbour-removal plots and in non-removal (control) plots. Both neighbour and nutrient manipulations had a highly significant effect on both biomass and tiller production but the interaction between these treatments was generally insignificant. Below-ground/above-ground biomass quotient was affected only by neighbour manipulations and was greatest in the control plots (with no neighbours removed) for all three species. The suppressive effect of neighbours was not markedly affected by nutrient level. However, yield suppression showed a significant decreasing trend with increasing nutrient level for biomass production in Agropyron and an increasing trend for tiller production in Phleum. For Poa, there was no trend in the intensity of competition across nutrient level. The results suggest that the general intensity of competition within this community neither increases nor decreases with increasing nutrient level. Rather, coexisting species appear to respond individually in terms of the intensity of competition that they experience. These results conflict with predictions from the triangular C-S-R model of plant strategies. However, they are consistent with a recently modified ‘habitat templet’ model for vegetation.  相似文献

With the aim of making clear the boundaries between species in thePoa acroleucahisauchii-nipponica aggregate, chromosomes and morphological features of 746 collections gathered from 125 localities in Japan were examined. For morphological observations, the voucher specimens of 95 collections whose chromosome numbers were reported previously (Tateoka, 1985) were also used. Tetraploids (2n=28) and hexaploids (2n=42), as well as a few pentaploids which were the hybrids between 4× and 6×, were found. By examining morphological features of these collections, two groups were recognized in tetraploids and one in hexaploids. The two tetraploid groups corresponded toPoa acroleuca Steud. andP. hisauchii Honda, and the hexaploid group toP. nipponica Koidz. It was confirmed that the hairiness on the internerve surface of lemma, ligule hairiness and the length ratio of anther/ lemma are the most important features for discriminating between these species; panicle shape, leaf shape and anther length are also helpful for the identification. The ambiguity of the boundaries between species which was hitherto present in the taxonomy of this species aggregate was unrelated to the creation of nature itself but was attributable to the insufficiency of our research work.  相似文献

Seventeen strains of Penicillium spp. have been isolated from Poa huecu Parodi from the Zapala zone, exhibiting toxicity to sheet. The following strains have been identified: P. crustosum, cyclopium, notatum, palitans, puberulum, verrucosum, viridicatum and Penicillium spp. The toxigenic capacity of the strains was studied after growing them under suitable conditions. Toxins produced were analysed by thin layer chromatography (TLC). Penitrem A (PA) and Penitrem B (PB) neurotoxins were identified and quantitated in twelve strains; verruculogen (VERR) and fumitremorgen B (FTB) being present in one of them. The effect of these mycotoxins was studied in mice. Neurological symptoms characteristic of the intoxication by tremorgenic toxins and similar to those observed in sheep suffering from huecu's disease were observed. The possible role of these toxins as causative agents of huecu's disease is discussed.  相似文献

Summary Plant regeneration in Kentucky bluegrass (Poa pratensis L. cv. Touchdown) via culture of seedling tissues was investigated. When coleoptile, leaf, and stem sections of dark-germinated seedlings were cultured on Murashige and Skoog (MS) medium, different types of callus were produced, depending on the expiant source and growth regulator combinations. Only compact-friable callus (type 3) and moderately compact, friable callus (type 2) produced shoots upon subculture. The nonstructured watery callus (type 4) produced roots without shoots. Shoot differentiation from callus tissues was highest when the culture medium contained 0.2 mgL–1 picloram + 0.01 mgL–1 -naphthaleneacetic acid (NAA). Calli grown from coleoptiles had higher shoot regeneration frequency (32%) than that obtained from either stem sections (12%) or young leaf tissues (2%) of the same seedlings. Some organogenic callus lines produced exclusively green plants, while others produced albino shoots or a mixture of green and albino shoots. The green plants were multiplied in a medium containing 0.1 mgL–1 BAP plus either 0.2 mgL–1 picloram or 0.1 mgL–1 indole-3-acetic acid (IAA). Over 90% of the cultures in the shoot proliferation medium produced roots in 4 weeks. This study investigates the nitrogen economy of six altitudinally contrasting Poa species which differ in their relative growth rate (R). Two alpine (Poa fawcettiae and P. costiniana), one sub-alpine (P. alpina)and three temperate lowland species (P. pratensis, P. campressa and P. trivialis) were grown hydroponically under identical conditions in a growth room. The low R exhibited by the alpine species was associated with lower plant organic nitrogen concentration (np) and lower nitrogen productivity (Πp, amount of biomass accumulation per mol organic nitrogen and time). The differences in Πp between the alpine and lowland species did not appear to be due to differences in the carbon concentration or the proportion of total plant organic nitrogen allocated to the leaves, stems or roots. Variations in ΠP were also not due to variations in photosynthetic nitrogen use efficiency (ΨN, the rate of photosynthesis per unit organic leaf nitrogen) or shoot or root respiration rates per unit organic nitrogen (ΛSH and ΛR, respectively) per se. Rather, the lower Λp in the alpine species was probably due to a combination of small variations in several of the parameters (e.g. slightly lower ΨN, slightly higher ΛSH and ΛR, and slightly higher proportions of total plant organic nitrogen allocated to the roots). The alpine species exhibited lower organic acid and mineral concentrations. However, no differences in whole-plant construction costs (grams of glucose needed to synthesize one gram of biomass) were observed between She alpine and lowland Poa species. The lack of sub-stantial differences in ΨN between the alpine and lowland species contrasts with the large differences in ΨN between slow- and fast-growing lowland species that have been reported in the literature. The reasons for the unusually high ΨN values exhibited by the alpine Poa species are discussed.  相似文献

Despite extensive interest in the role of plant size in competition, few formal attempts have been made to quantify the magnitude of asymmetric competition, particularly for interactions between members of different species. This paper introduces the concept of asymmetric interspecific competition at the population livel (i.e. mean plant performance) in mixtures of species. It proposes an index of interspecific competitive asymmetry which allows for a progressively greater asymmetric effect as the average size differences between competing species increase, and allows for such an effect whether individuals of focal species are larger or smaller, on average, than competitors. This index of competitive asymmetry is evaluated in the study of interactions between two widely coexisting annuals of disturbed habitats, Stellaria media and Poa annua. An experiment was conducted in which the density, relative frequency and relative seedling sizes (emergence times) of Poa and Stellaria individuals were varied. The relative growth rate (RGR) for both species was measured over a 22-day period. An inverse linear model was fitted for each species, relating the RGR of the focal species to the initial biomass of each species. Each response model included an asymmetry coefficient () to assess whether the impact of a unit of initial biomass of the associate species changed with the relative sizes of seedlings of the two species. A zero value of implies symmetric competition between the two populations; i.e. the competitive effect of a unit of associate species biomass does not change with its initial seedling size. If is positive the smaller the initial relative size of seedlings of the associate species, the smaller their per unit biomass effect on the response of the focal species. The model fitted our data for Stellaria and Poa well and was validated by an alternative modelling approach. Asymmetry coefficients were estimated as 0.508 (P<0.05) for the effect of Poa in the Stellaria model, and 0.0001 (NS) for the effect of Stellaria in the Poa model; i.e. the effect of Poa on Stellaria was asymmetric while the effect of Stellaria on Poa was symmetric. Differences in interspecific species asymmetric competitive effects are discussed within the context of shoot architecture, and the relative importance of competition for light versus soil resources. Finally, we discuss the relationship of this model to earlier models of competitive asymmetry, and consider the implications of interspecific competitive asymmetry for a number of current theories of plant competition and community organisation.  相似文献
