In addition to (+)-, (-)- and (+/-)-heliannuol E, growth-inhibitory activities of five synthetic chromans and four tetrahydrobenzo[b]oxepins were examined against oat and cress. All heliannuol E isomers exhibited similar biological activities against cress, whereas when tested against oat roots, the unnatural optical isomer (+) showed no inhibitory activity. Four brominated chromans and two tetrahydrobenzo[b]oxepin derivatives also showed apparent inhibition against both cress and oat.  相似文献

The use of nuclear analytical techniques for environmental studies in South America is selectively reviewed since the time of earlier works of Lattes with cosmic rays until the recent applications of the PIXE (particle-induced X-ray emission) technique to study air pollution problems in large cities, such as São Paulo and Rio de Janeiro. The studies on natural radioactivity and fallout from nuclear weapons in South America are briefly examined.

AIMS: The screening and initial characterization of bacteriocins produced by lactic acid bacteria (LAB) from raw Tenerife goats' cheese with possible application as biopreservatives or ripening accelerators for Tenerife cheese. METHODS AND RESULTS: One hundred and eighty LAB of the genera Lactobacillus (95), Leuconostoc (64) and Lactococcus (21) isolated from raw Tenerife goats' cheese were screened for the production of antimicrobial substances. Lactobacillus plantarum TF711, which had the broadest spectrum of antimicrobial activity, was selected for further characterization. The antimicrobial compound was determined as a proteinaceous substance, as it was sensitive to proteases. The bacteriocin-like substance, which we called plantaricin TF711, was active against the Gram-positive bacteria Bacillus cereus, Clostridium sporogenes and Staphylococcus aureus; and against the Enterobacteriaceae Shigella sonnei and Klebsiella pneumoniae. It was stable to heat and to treatment with surfactants and organic solvents. Highest antimicrobial activity was found between pH 1 and 9. Plantaricin TF711 exhibited primary metabolite kinetics, a bacteriostatic mode of action and a molecular mass of c. 2.5 kDa as determined by tricine SDS-PAGE. CONCLUSIONS: Lact. plantarum TF711 produces a low molecular mass bacteriocin-like compound with a wide spectrum of activity and interesting technological properties (thermostability, good pH stability and stability against surfactants and organic solvents). SIGNIFICANCE AND IMPACT OF THE STUDY: Plantaricin TF711 was found to have potential for use as a biopreservative in the food industry.  相似文献

The longevity of organisms is intrinsically interesting and can provide useful information on their population structure and dynamics and the dynamics of associated communities. With the exception of perennial Laminariales that have rings in the stipe, the life spans of most perennial macroalgae are unknown or based on anecdotal observations. Using morphological analyses combined with the location and time of the rise in 14C from atmospheric nuclear testing within the thallus, we determined that the growth rate of a specimen of Clathromorphum nereostratum Lebednik from Adak Island was 0.30 mm·yr?1, the 30 bands within the thallus were annual, and the specimen sampled was 61–75 years old. Living crusts of this species from the same geographic region are reported to be up to 20 cm thick. Assuming our growth rate is typical, C. nereostratum can be approximately 700 years old, the oldest known living alga. This longevity and consistent banding within the thallus suggest that smaller scale sampling and additional chemical analyses of this alga could provide a detailed long‐term record of environmental variation at high latitudes in the North Pacific.  相似文献

The purpose of this study was to evaluate the suitability of the Daily QA 3 (Sun Nuclear Corporation, Melbourne, USA) device as a safe quality assurance device for control of machine specific parameters, such as linear accelerator output, beam quality and beam flatness and symmetry. Measurements were performed using three Varian 2300iX linear accelerators. The suitability of Daily QA 3 as a device for quality control of linear accelerator parameters was investigated for both 6 and 10 MV photons and 6, 9, 12, 15 and 18 MeV electrons. Measurements of machine specific using the Daily QA 3 device were compared to corresponding measurements using a simpler constancy meter, Farmer chamber and plane parallel ionisation chamber in a water tank. The Daily QA 3 device showed a linear dose response making it a suitable device for detection of output variations during routine measurements. It was noted that over estimations of variations compared with Farmer chamber readings were seen if the Daily QA 3 wasn’t calibrated for output and sensitivity on a regular eight to ten monthly basis. Temperature-pressure correction factors calculated by Daily QA 3 also contributed towards larger short term variations seen in output measurements. Energy, symmetry and flatness variations detected by Daily QA 3 were consistent with measurements performed in water tank using a parallel plate chamber. It was concluded that the Daily QA 3 device is suitable for routine daily and fortnightly quality assurance of linear accelerator beam parameters however a regular eight-ten monthly dose and detector array calibration will improve error detection capabilities of the device.  相似文献

Most of the steps in, and many of the factors contributing to, glucocorticoid receptor (GR)-regulated gene induction are currently unknown. A competition assay, based on a validated chemical kinetic model of steroid hormone action, is now used to identify two new factors (BRD4 and negative elongation factor (NELF)-E) and to define their sites and mechanisms of action. BRD4 is a kinase involved in numerous initial steps of gene induction. Consistent with its complicated biochemistry, BRD4 is shown to alter both the maximal activity (Amax) and the steroid concentration required for half-maximal induction (EC50) of GR-mediated gene expression by acting at a minimum of three different kinetically defined steps. The action at two of these steps is dependent on BRD4 concentration, whereas the third step requires the association of BRD4 with P-TEFb. BRD4 is also found to bind to NELF-E, a component of the NELF complex. Unexpectedly, NELF-E modifies GR induction in a manner that is independent of the NELF complex. Several of the kinetically defined steps of BRD4 in this study are proposed to be related to its known biochemical actions. However, novel actions of BRD4 and of NELF-E in GR-controlled gene induction have been uncovered. The model-based competition assay is also unique in being able to order, for the first time, the sites of action of the various reaction components: GR < Cdk9 < BRD4 ≤ induced gene < NELF-E. This ability to order factor actions will assist efforts to reduce the side effects of steroid treatments.  相似文献

PurposeThe accuracy of gated irradiation may decrease when treatment is performed with short “beam-on” times. Also, the dose is subject to variation between treatment sessions if the respiratory rate is irregular. We therefore evaluated the impact of the differences between gated and non-gated treatment on doses using a new online quality assurance (QA) system for respiratory-gated radiotherapy.MethodsWe generated dose estimation models to associate dose and pulse information using a 0.6 cc Farmer chamber and our QA system. During gated irradiation with each of seven regular and irregular respiratory patterns, with the Farmer chamber readings as references, we evaluated our QA system’s accuracy. We then used the QA system to assess the impact of respiratory patterns on dose distribution for three lung and three liver radiotherapy plans. Gated and non-gated plans were generated and compared.ResultsThere was agreement within 1.7% between the ionization chamber and our system for several regular and irregular motion patterns. For dose distributions with measured errors, there were larger differences between gated and non-gated treatment for high-dose regions within the planned treatment volume (PTV). Compared with a non-gated plan, PTV D95% for a gated plan decreased by −1.5% to −2.6%. Doses to organs at risk were similar with both plans.ConclusionsOur simple system estimated the radiation dose to the patient using only pulse information from the linac, even during irregular respiration. The quality of gated irradiation for each patient can be verified fraction by fraction.  相似文献

FVE/MSI4, a highly conserved gene through evolution, is considered a classical flowering time gene from the autonomous pathway in Arabidopsis thaliana. Our work provides complementary, yet distinct, evidence that mutations in FVE/MSI4 have highly pleiotropic effects on plant architecture and organ production and growth, indicating an overall role in reprogramming the genome. First, this is illustrated by the fact that fve mutants and transgenic versions of the gene show no morphological aberrations while living, on average, twice as long and producing more biomass and seeds than the wild type. For example, depending on the photoperiod, the vegetative biomass in the mutants is increased three- to eightfold. FVE/MSI4 can therefore be considered a multifactor component of biomass and yield. Second, the gene functions primarily at the shoot apical meristem, acting to calibrate its overall cell proliferation activity and organ initiation. At the same time, FVE/MSI4 regulates cell growth during organ formation mainly by modulating the timing of proliferation—differentiation transition and that of endoreduplication. FVE/MSI4 is an accelerator of ontogenesis. The function of the gene is to scale-down meristem activities, while accelerating developmental transitions and cellular differentiation. Genes of the kind are ideal tools in biotech for engineering biomass and yield according to geographical or climate constraints. The results enhance our understanding on the role epigenetic components of the genome, such as FVE/MSI4, play in adaptation and biomass calibration. They also illustrate the kind of functional characterization biotech applications might require during the first steps of product development. The new technique of tandem accelerator mass spectrometry (TAMS) has improved the sensitivity for measurement of several long-lived radioisotopes and certain stable isotopes by many orders of magnitude. Nuclear physics tandems and new small dedicated accelerators are now able to measure14C,10Be,26Al,32Si,36Cl,41Ca, and129I in natural materials. Sensitivities down to 105 atoms per sample can be achieved in favorable cases. By accelerating ions to MeV energies, one can eliminate molecules and uniquely identify the atomic numbers below 20. Although most applications to date have been in the earth sciences, the opportunity now exists for important new applications in biology and toxicology. Trace elements can be measured at the parts per billion (109) level using a secondary ion mass spectrometry (SIMS) ion source. Radioactive tracer measurements can be made for elements, such as aluminum, for which there are no isotopes with suitable half-lives for conventional decay counting methods. For14C, counting times become much shorter and dose levels can be reduced.  相似文献
