ABSTRACT. A non-dispersive infrared gas analyser equipped with a Luft-type sonic detector and flow-through reference cell was automated to monitor the total volume of carbon dioxide (CO2) respired by single insects or groups of insects. The infrared analyser was interfaced with an integrator for quantification, a microprocessor to control intermittent air flow through the insect respiration chambers, and a microcomputer for data storage and reduction. This technique has been used to monitor the CO2 Output of diapausing and non-diapausing mature fifth instar larvae and of developing pupae of the codling moth, Cydia pomonella (L.) (Lepidoptera: Tortricidae). The resulting data were accurate, quantitative and reproducible.  相似文献

Origin of bombesin-like peptides in human fetal lung   总被引:2,自引:0,他引:2  
Four different forms of bombesin-like immunoreactive peaks were detected in extracts of human fetal lung by the use of reversed-phase high performance liquid chromatography (HPLC). Peaks I, II, III and IV, (increasing retention time), were eluted using a 14-38% of acetonitrile gradient containing 0.1% trifluoroacetic acid (TFA). Peak II was the major material found in the extract of human fetal lung obtained at 16-20 weeks gestation. None of the four compounds contained in the eluted peaks had the same retention time as amphibian bombesin or porcine gastrin releasing peptide (GRP). On reversed-phase HPLC using two different solvent systems TFA or heptafluorobutyric acid (HFBA) as a hydrophobic counter ion, and in gel filtration chromatography, the chromatographic behavior of the main peak (peak II) was the same as that of the carboxyl terminal fragments of GRP, GRP18-27 or GRP19-27. This suggested that the peptide(s) in peak II resembled in composition the carboxy terminal 9 or 10 amino acids of porcine GRP. Following tryptic digestion the material in peak IV was converted to the more polar compound present in peak II. Two other peptide peaks were eluted close to peak II and these were presumed to be a modification of this main peak. One of the possible biosynthetic steps in the formation of bombesin-like peptides in human fetal lung could be a tryptic conversion of a less polar peptide to a more polar form (peak IV to II).  相似文献

Experimental evaluation of the mass of food withdrawn by larvae of the Siberian moth during their development on larch, cedar pine, fir, spruce, and common pine was carried out. The obtained dependences between the masses of larvae and food withdrawn by them can be directly used to determine the withdrawn food rate of Siberian moth on each of the Siberian conifer species.  相似文献

Male moths locate conspecific females by pheromone‐induced upwind flight maintained by detecting a visual flow, termed optomotor anemotaxis. Their behavioural pattern is characterized by an upwind surge in response to a pheromone stimulus and crosswind casting after odour loss, which is considered to be reset and restarted on receipt of another pheromone pulse. However, pheromone‐stimulated males of the potato tuberworm moth Phthorimaea operculella exhibit a series of short and straight intermittent flights, or hops, when moving upwind. It is unclear whether they navigate by employing the same behavioural pattern and wind detection mechanism as that used by flying moths. To analyze odour‐modulated anemotaxis in male potato tuberworm moths, a flat wind tunnel is constructed to give regular odour stimuli to an insect regardless of its location. Moths are subjected to pheromone pulses of different frequencies to test whether they show a behavioural pattern that is reset and restarted by a pheromone pulse. Moths on the ground are also subjected to crosswind shear to examine their detection of wind direction. Path analyses reveal that males surge upwind when they receive a pheromone pulse and exhibit casting by successive hops when they lose odour. This behavioural pattern appears to be similar to that of flying moths. When the direction of the airflow is switched orthogonally, males adjust their course angle accordingly when they are on the ground. It is suggested that, instead of optomotor anemotaxis, this ‘aim‐then‐shoot’ system aids the detection of wind direction, possibly by mechanosensory means.  相似文献

Melanic polymorphism in B. betularia has been extensively studied. Correlations between high melanic frequency and high levels of air pollution have been demonstrated. Kettlewell and others have shown that differential bird predation has an important effect on the maintenance of the polymorphism, and coefficients of visual selection have been obtained on the assumption that the moth habitually rests on tree trunks. Computer models based on these selective coefficients show that they are not sufficient accurately to explain observed melanic frequencies. Other non-visual selective factors and weak frequency-dependent selection have been invoked to improve fits. Analysis of the resting positions of moths recorded in the wild demonstrates that B. betularia does not usually rest in exposed positions on tree trunks, but rather rests on the underside of branches, on trunks in shaded positions just below major branch joints or on foliate twigs. The results of a pilot selection experiment, while agreeing qualitatively with Kettlewell's results, suggest that fitness estimates that assume trunk-resting are quantitively incorrect. The error is greatest for melanic moths in rural areas. It is suggested that visual selective coefficients based on a true assessment of the resting behaviour of the moths may considerably improve the fit between computer predictions and observed phenotype frequency distributions.  相似文献

Artificial diets were reviewed and two tested. The highest level of survival to the adult stage (72%) was obtained on a modified diet of Odell & Rollinson (1966). Rearing required 61 days for males and 70 days for females. Wing deformation occurred in 16% of the adults. The sex ratio ( : ) was 0.80. Average pupal weight was 281 mg for males and 368 mg for females. Females averaged 102 eggs (range 80–125 eggs). Neonates from field-collected egg masses started hatching on 6 December; frost was not necessary for hatching. Hatching could be postponed until at least 10 November of the following year by storing egg masses at-2°C. L. monacha can be maintained continuously in the laboratory.
Zusammenfassung Eine Massenvermehrung der Nonne, Lymantria monacha (L.), trat in 1984 auf 500 ha Pinus sylvestris bei Budelerbergen (S.O. der Niederlande) auf und betraf 1985 eine Fläche von 2800 ha. Obwohl die Vermehrung durch eine Luftapplikation mit Dimilin 25 bekämpft werden konnte, wurden Untersuchungen begonnen zur Entwicklung von für die Umwelt sicheren Bekämpfungsmethoden, besonders die Anwendung von Virosen. Das Ziel bestand in der Virusproduktion in künstlichen Raupenzuchten. Die Nonne wurde bisher mit verschiedenen für andere Insektenarten oder speziell für diesen Zweck entwickelten künstlichen Nährböden gezüchtet. Da die Zusammensetzung dieser Nährböden und die Zuchtmethoden nicht veröffentlicht waren und da wenige Details über die Raupenentwicklung vorlagen, wurden modifizierten Nährböden von McMorran sowie von Odell & Rollinson für die Massenzucht der Nonne geprüft.

The cabbage root fly, Delia radicum (L.), was deterred from laying eggs on cauliflower plants that had been sprayed with a suspension of the frass of caterpillars of the garden pebble moth, Evergestis forficalis (L.). Polar extracts of the frass deterred oviposition irrespective of the cruciferous plant species on which the caterpillars had been feeding. Non-polar extracts of the frass had no effect. Spraying plants with macerates from Brassica leaves stimulated fly oviposition whereas spraying plants with macerates from garlic mustard leaves deterred fly oviposition. Macerates from the leaves of all other plants tested had no effect. In field experiments the deterrent effect persisted 2–3 days after leaves were sprayed with frass extracts. Plants infested with feeding caterpillars and contaminated with only a few discrete frass pellets were as deterrent to the fly as those sprayed with frass suspensions.

Les extraits polaires de ces excréments empêchent la ponte quelle que soit l'espère de crucifère sur laquelle les chenilles se sont alimentées. Les extraits non polares sont sans effet. Dans la nature, l'action dissuadante persiste 2 à 3 jours après la pulvérisation des feuilles avec les extraits d'excréments. La pulvérisation tous les deux jours a maintenu l'action dissuadante. Des plantes contaminées par des chenilles en train de s'alimenter et souillées par quelques crottes sont aussi dissuadantes pour la mouche que celles pulvérisées avec une suspension d'excréments.

Summary Aberrations of photoreceptor ultrastructure resulting from carotenoid/retinoid (vitamin A) deprivation were studied in the retina of Manduca sexta. The syndrome of chromophore deficiency included hypertrophy of smooth endoplasmic reticulum, variable dilation of rhabdomeric microvilli, the insertion of endomembrane fingers into such enlarged microvilli, and the formation of rhabdomeric vacuoles, intracellular compartments containing microvilli similar to those of the rhabdomere. Retinas were processed either with conventional procedures employing preliminary aldehyde fixation followed by heavy metal postfixation, or by fixation and incubation in unbuffered OsO4. The latter method deposits osmium throughout the endomembrane system, within the rhabdomeric vacuoles, and in the extracellular space of the rhabdom. However, the intravillous fingers were rarely impregnated with osmium, despite their continuity with densely stained cisternae of the smooth endoplasmic reticulum. We suggest that the insertion of endomembrane fingers into dilated microvilli results from a cytoskeleton-mediated link between cisternae of the smooth endoplasmic reticulum and the rhabdomeric membrane, an association that may be important in the turnover of photoreceptor membrane. We interpret endomembrane hypertrophy and development of rhabdomeric vacuoles as symptoms of disturbance in the pathway leading to the assembly of the rhabdomere resulting from reduced synthesis of visual pigment.  相似文献

Temporal events of gypsy moth vitellogenesis and ovarian development   总被引:1,自引:0,他引:1  
Abstract The vitellogenic period of gypsy moth ovarian development starts on day 3 of the pupal stage and continues through adulthood. During this period, rapid increases occur in follicle size, protein content, and wet weight of the ovary. Patency is observed on day 3 of the pupal stage.
Pre-vitellogenic follicles are formed in the last larval stadium. Newly formed follicles detach from the germarium on day 4, and increase rapidly to 140 per ovariole at the end of the last larval stadium. The pre-vitellogenic follicles are uniformly around 50 um in diameter. No vitellogenin is incorporated into the oocytes until the pupal stage.
Polyacrylamide gel electrophosesis (PAGE) in the presence of sodium dodecylsulphate (SDS) analysis of male and female haemolymph samples and vitellogenic ovaries demonstrates the presence of two female-specific subunits of vitellogenin of 180 kD and 160 kD. These proteins are detected only in haemolymph and ovarian extracts of vitellogenic females. The molecular weight of the native protein determined by size exclusion chromatography is approximately 400–420 kD.
A highly sensitive double antibody sandwich enzyme-linked immunosorbent assay (ELISA) was developed to monitor the temporal changes in vitellogenin titre in haemolymph. Vitellogenin production starts on day 2 of the last larval stadium, reaching a maximum level by day 6 of the last larval stadium, and decreasing in the late pupal stage as vitellogenin was internalized into the oocytes. This is the first report of vitellogenin production occurring in the larval stage of a holometabolous insect. The fact that vitellogenin production and uptake occur during different stages of development in the gypsy moth, opens up some interesting questions concerning the underlying regulatory mechanisms controlling each process.  相似文献

A spring-fed, headwater stream in central Rhode Island was examined during the period from June to October, 1979 to 1982. In the first two summers, a dense riparian canopy reduced the light penetration at the stream surface to a range of 5 to 18% of incident radiation. The lotic macroalgal community during this period was limited to 1 to 4 species covering < 1 to 35% of the stream bottom. However, in June and July, 1981, the surrounding leaf canopy was removed by a massive gypsy moth larval outbreak. Light penetration to the stream during this summer increased to 73% by early July, thereby resulting in a rise in water temperatures by 3.7°C. Even though there was a partial regrowth of leaves in late July and August of 1981, macroalgal cover values continued to rise to an early August peak of 80%. During the third summer, 88% of the macroalgal abundance could be attributed to illumination and water temperature. The filamentous diatom Funotia pectinalis ( O.F. Müll.) Rabh. was the predominant species in the midsummer of all four years, accounting for at least 60% of the total cover. In 1981. an important taxon was the desmid Hyalotheca dissiliens (S. Smith) Bréb., a species which was not seen in other years. A less severe gypsy moth defoliation occurred in 1982 but did not produce significant differences in light, temperature or macroalgal cover from 1979 and 1980. The results indicate that light and temperature can be limiting during the summer in spring-fed, headwater streams and that seed populations of some species are present in undetect-able levels during these periods of suboptimal growth conditions. In addition, it appears that stream macroalgal communities can be quite resilient, recovering rapidly following a major perturbation .  相似文献
