Cyanobacteria possess thylakoid membranes that differ in their protein composition from the cytoplasmic membrane. To study possible pathways of protein targeting to these membranes, we have investigated whether or not cyanobacteria have a homologue or homologues of the signal recognition particle-like chaperone Ffh. We have amplified a fragment of ffh by polymerase chain reaction and established that ffh is present as a single copy in the genomes of three cyanobacterial species. We have cloned and sequenced ffh from Synechococcus sp. PCC7942 and predict that Ffh functions as a ribonucleoprotein in cyanobacteria and chloroplasts.  相似文献

Cells of the non-N2-fixing cyanobacteriumPhormidium laminosum were immobilized in polyurethane (PU) foams either by absorption or by entrapment in the PU prepolymer followed by polymerisation and by adsorption onto polyvinyl (PV) foams. Although entrapment caused toxicity problems which lead to rapid death of the immobilized cells, they were immobilized successfully by adsorption onto PU or PV foams and maintained their photosynthetic electron transport activities (PS I, II, I + II) for at least 7 weeks. Changes in the morphology resulting from immobilization, as revealed by scanning electron microscopy (SEM) and low temperature-SEM, were investigated. Batch cultures and a continuous-flow packed bed photobioreactor were used to study nitrate removal from water. The effects of light intensity and CO2 concentration on bioreactor performance were studied with respect to the nitrate uptake efficiency of the system. The redox state of plastoquinone was measured in vivo in the blue-green alga, Phormidium uncinatum by means of a double beam UV-spectrophotometer. The difference in absorbance of the oxidized and the reduced forms of plastoquinone was amplified, and stored and averaged in a computer. The redox state was changed by two alternating actinic light beams. When one actinic wavelength was kept constant at 700 nm (PSI) variation of the other yielded an action spectrum representing photosystem II. Abstract Pigment decomposition, oxygen evolution and CO2 fixation were measured in the cyanobacterium Phormidium uncinatum after infection with cyanophage LPP-1, under light and dark conditions. A gradual decrease in para benzoquinone supported O2 evolution, chlorophyll a and phycocyanin level were noticed after 6 h of infection. These results demonstrated decreased photosynthetic activity of the host P. uncinatum prior to the start of LPP-1 multiplication. Metabolic inhibitor investigations confirmed that the cyanophage LPP-1 multiplication was independent of host photosynthesis.  相似文献

Abstract Screening of fatty acid composition in 150 strains of marine microalgae, cyanobacteria and green algae was carried out, and 20 strains showed relatively high contents of palmitoleic acid. Among them, two cyanobacteria, Phormidium sp. NKBG 041105 and Oscillatoria sp. NKBG 091600, showed an unusually high cis -palmitoleic acid content (54.5% and 54.4% of total fatty acid, respectively). Phormidium sp. NKBG 041105 had the highest cis -palmitoleic acid content per biomass (46.3 mg (g dry cell weight)−1), and cis -palrnitoleic acid composition was found to be constant with varying temperature. These results indicate that this cyanobacterium could be considered as a new source for palmitoleic acid.  相似文献

The chemical composition of organisms in relation to their environmental resource availability is an area of intense research activity. We studied the changes in cell composition of the cyanobacterium Phormidium autumnale in response to prolonged darkness. Cells allocated their internal resources in a homeostatic manner, oxidizing all the three major cellular constituents in a proportional way. This resulted in constant C/N and carbohydrates, lipids and proteins ratios that remained unaltered throughout the whole incubation period. We propose the maintenance of balanced cell composition (homeostasis) as an evolutionary strategy related to environmental transitory changes.  相似文献

Field studies on three perennially ice-covered Antarctic oasis lakes with little or no outflow disclosed a unique biological phenomenon. Benthic algal mats dominated by the prokaryotic Phormidium frigidum Fritsch and several pennate diatoms growing in shallower, more brightly illuminated areas beneath 4 to 5.5 m of ice accumulate and entrap bubbles of photosynthetically produced oxygen. Clumps of this gas-filled mat tear loose from the gravelly substrate, lift off and float to the bottom of the lake ice. Some of these floating mat pieces become frozen into newly forming ice with the onset of winter. Through the combination of ablation of ice from the upper surface and the formation of new ice from below, algal mat pieces reach the upper lake surface in 5–10 years. Here, they are lyophilized by polar winds and dispersed in at least a partially viable state. The process of mat lift-off and escape is important in removing nutrients and salts from these lakes and helps to perpetuate their oligotrophic state. Neutron activation and X-ray dispersive analyses of elements in the algal mats along with other analyses, field observations, and calculations suggest that significant quantities of organic matter, select minerals and salts are lost from the lakes annually through this process whose magnitude has not been recognized previously.  相似文献

In an attempt to clarify the interactions between the available nitrogen source and the photosystems in cyanobacteria, O2 exchange and fluorescence emission were monitored in spheroplasts and intact cells of the non N2-fixing cyanobacterium Phormidium laminosum (strain OH-1-p.Cl1) growing on different nitrogen sources or in the absence of nitrogen. Short-term (time scale of seconds to minutes), NH+4 addition to NO3-growing or N-starved cells and, to a minor extent, NO3addition to N-starved cells, induced state 2 transitions both in light and dark. Long term (time scale of days), the fluorescence yield of PSII relative to that of PSII at 77 K was higher in NO3- than in NH+4 growing cells, and even higher in N-starved cells. In the dark, the plastoquinone pool was more reduced in NH+4- than in NO3-growing cells. Both PSII and PSI activities and the degree of linking between both photosystems were affected in the long term, so that non-cyclic electron transport decreased in parallel to the ferredoxin requirement to assimilate each nitrogen source. Results indicate that nitrogen metabolism exerts short- and long-term control over the photosynthetic apparatus, which acclimates to the energy requirement of the available nitrogen source.  相似文献
