稀有鮈鲫──一种新的鱼类毒性试验材料   总被引:7,自引:0,他引:7  
本文研究了稀有鲫(Gobiocyprisrarus)作为毒性试验材料的可行性。采用换水式试验,在硬度为200mg/L(以CaCO3计)、pH7.8±0.2、温度24-25℃条件下研究了铬、铜、锌和五氯酚(PCP)对稀有鲫的急性毒性。重铬酸钾对2日龄稀有鲫的24h和96h和LC50控制范围分别263.6-334.7和1153-178.5mg/L(n=8)。铬、铜、锌和五氯酚对2日龄稀有鲫的急性毒性值(96hLC50)范围,从铜的52.2μg/L到铬的52000μg/L,毒性大小的顺序是铜>五氯酚>锌>铬。研究结果表明,稀有鲫有可能发展成为一种较为理想的毒性试验材料。  相似文献

Expression of hsromega stress gene in the third-instar larvae of 951-lacZ2 (hsromega-lacZ having-844pb sequence) and 498-lacZ1 (hrsomega-lacZ having -498bp sequence) strains of Drosophila melanogaster at LC(50) and lower dietary concentrations of hexachlorocyclohexane (HCH) pentachlorophenol (PCP), and endosulfan was examined in relation to larval mortality by beta galactosidase activity, vital dye staining, and salivary gland polytene chromosome puffing. Our results showed that both HCH and PCP at lower concentrations evoked strong hsromega stress gene expression in the larval tissues while endosulfan did not. On the other hand, puffing data revealed that endosulfan at lower doses, induced well-developed puff at the resident site (93D) of the hsromega gene but the transgenic sites (30B in 951-lacZ2 and 44B in 498-lacZ1 strain) did not show any well-developed puff. Regression in hsromega stress gene expression in 951-lacZ2 strain at LC(50) concentrations of HCH and PCP after 48 h was concurrent with extensive tissue damage as evident by trypan blue staining. Similarly, strong hsromega expression was accompanied by insignificant trypan blue staining in the larval tissues of this strain after shorter duration of exposure (2-12 h) to these toxicants. Although endosulfan under similar experimental condition did not induce hsromega, strong trypan blue staining indicated extensive tissue damage after 48 h of exposure. The present study suggests that all the three toxicants pose cytotoxic potential to Drosophila. While protective role of this stress gene was evident at the initial stages of exposure, extensive tissue damage in the later stages of intoxication accompanied by autorepression of hsromega led to larval mortality. The study further suggests that -844bp upstream sequence of the gene is adequate for hsromega inducibility against HCH and PCP but not for endosulfan for which responsive elements may be searched further upstream.  相似文献

Researches on the polymerization of aqueous pentachlorophenol (PCP) by the catalysis of horseradish peroxidase (HRP) with the existence of hydrogen peroxide (H2O2) were conducted. Factors, such as acidity, temperature, enzyme activity, and initial concentration of PCP and H2O2 that could influence the degradation were studied. Results showed that the optimum pH value for free enzyme was 5–6; relative higher temperature could accelerate the reaction greatly; PCP removal increased with an increase of enzyme concentration, and PCP (initial concentration 12.6 mg/L) removal percentage could reach nearly 70% under the highest enzyme concentration (about 0.05 u/ml) adopted in the experiment; removal percentage increased slightly with an increase of initial concentration of PCP, and when initial PCP concentrations were 13.0 and 0.7 mg/L, the removal percentages were about 73.7% and 35.7%, respectively; the molar ratio of the reaction between PCP and H2O2 was about 1:2.Based on the above results, researches on the removal of PCP by the immobilized HRP were conducted. The free HRP was immobilized on the polyacrylamide gel prepared by gamma-ray radiation method; then the immobilized HRP was filled into a column, and PCP was successfully removed by the immobilized HRP column. The results were compared with results using free HRP enzyme, which showed that the optimum pH value for the immobilized HRP is similar to that for the free HRP, and when pH=5.15, the immobilized HRP could reduce PCP with initial concentration 13.4 mg/L to the concentration of 4.9 mg/L within 1 h, and the immobilized HRP column could be used to repeatedly.  相似文献

Pentachlorophenol (PCP) was more rapidly degraded in acetate and glucose-fed microbial fuel cells (MFCs) than in open circuit controls, with removal rates of 0.12 ± 0.01 mg/Lh (14.8 ± 1.0 mg/g-VSS-h) in acetate-fed, and 0.08 ± 0.01 mg/L h (6.9 ± 0.8 mg/g-VSS-h) in glucose-fed MFCs, at an initial PCP concentration of 15 mg/L. A PCP of 15 mg/L had no effect on power generation from acetate but power production was decreased with glucose. Coulombic balances indicate the predominant product was electricity (16.1 ± 0.3%) in PCP-acetate MFCs, and lactate (19.8 ± 3.3%) in PCP-glucose MFCs. Current generation accelerated the removal of PCP and co-substrates, as well as the degradation products in both PCP-acetate and PCP-glucose reactors. While 2,3,4,5-tetrachlorophenol was present in both reactors, tetrachlorohydroquinone was only found in PCP-acetate MFCs. These results demonstrate PCP degradation and power production were affected by current generation and the type of electron donor provided.  相似文献

To investigate the feasibility of using biochar to control organic pollutants in sediments, we extracted biochar from rice-straw combustion residues (RBC) and studied its adsorption ability and effect on seed germination ecotoxicity of pentachlorophenol (PCP). The results showed that the Freundlich and dual-mode models could describe all the sorption isotherm data well, and the log KOC values increased with increasing RBC content. With 50 mg kg−1 PCP in the sediment, a significant seed growth inhibition (P < 0.01) was observed. The addition of 2.0% RBC lowered the PCP concentration in the extraction liquid from 4.53 to 0.17 mg L−1 and increased the germination rate and root length significantly. Furthermore, it was found that the addition of RBC had no toxic but stimulative effect on root elongation. Consequently, RBC could serve as a potential supersorbent for the remediation of organic pollution in situ.  相似文献

Chlorophenols, widespread soil and water contaminants and often degradation products of some pesticides, are a potential stress factor for survival of environmental bacteria. The effect of pentachlorophenol (PCP) and 2,4-chlorophenol (2,4-CP) on the growth, amount of lipid, and fatty acid composition in the membrane lipids was examined in a strain of the bacterium Kocuria varians, able to degrade chlorophenols. The index of fatty acid unsaturation in two main membrane lipids, phosphatidylcholine (PC) and phosphatidylethanolamine (PE) decreased in the presence of chlorophenols. Transformation of stearic acid into oleic acid was significantly increased by PCP addition only in PE, but conversion of oleic acid into linoleic acid was blocked by PCP and 2,4-CP in both PC and PE. This observation may indicate that while Δ9 desaturase was sensitive mainly to 2,4-CP, activity of Δ12 desaturase was inhibited by both PCP and 2,4-CP.  相似文献

研究了污染沉积物泥浆液、固两相五氯酚(PCP)厌氧生物降解.结果表明,投加10g·kg-1厌氧颗粒污泥,经31d处理泥浆液、固两相PCP降解率达98.9%,平均降解速率达到80mg·kg-1·d-1,对照处理平均降解速率仅为4.4mg·kg-1·d-1,颗粒污泥生物强化作用明显.作为泥浆修复过程的调控因子,有机溶剂、共基质和表面活性剂对PCP降解效应不同,投加乙醇,可提高PCP解吸和降解速率,4d内两相PCP降解速率达到54.3mg·kg-1·d-1;而投加共基质和非离子表面活性剂乙二醇丁醚后,液、固两相PCP降解均出现迟滞,两者均不同程度地抑制PCP降解.  相似文献

A bacterium degrading pentachlorophenol (PCP) as the only source of carbon and energy was grown in a nutristat, i.e., a continuous culture with on-line measurement and control of the substrate concentration. We improved the PCP nutristat by incorporation of a personal computer with a proportional integral derivative (PID) algorithm for controlling the medium feed pump. The controlled value deviated from the average (set-point) value by 1% maximally. In the PCP nutristat (30°C), the steadystate dilution rate, and hence, specific growth rate, showed a maximum value of 0.142±0.004 h-1 at set-point PCP concentrations between 37 and 168 M. At PCP concentrations above 168 M, the steady-state growth rate decreased because of inhibition. The growth yield coefficient was not seriously affected by the PCP concentration, suggesting that uncoupling was not the inhibitory mechanism. It was concluded that the PCP nutristat is very useful for establishing steady-state conditions that maintain growth-inhibitory PCP concentrations and high cell concentrations, conditions for which the chemostat is not suitable.Abbreviations MCA Metabolic control analysis - NTA Nitrilotriacetic acid - PCP Pentachlorophenol - PID Proportional integral derivative  相似文献

The sublethal biochemical effects of pentachlorophenol (PCP) were investigated in live, intact red abalones (Haliotis rufescens), using a flow-through exposure system, by in vivo 31P NMR spectroscopy. Based on rangefinding tests (6-hr LC50 = 1.6 mg/L; 6-hr no-observable-effect-level (NOEL) = 0.8 mg/L), three abalones were separately exposed to a sublethal concentration (1.2 mg/L) for 5 hr, followed by a 13 hr recovery period. Effects in foot muscle included both a decrease in phosphoarginine and an increase in inorganic monophosphate concentrations ([PA] and [Pi], respectively); both foot muscle concentrations of adenosine triphosphate [ATP] and intracellular pH (pHi) also declined. Parallel in vitro experiments revealed that concentrations of glycerol 3-phosphate, lactate, citrate, succinate, malate, and alanine (Ala) all increased, while those of glyceraldehyde 3-phosphate and glutamine (Gln) remained stable. Also, these effects were not evident until 2 hr into exposure, possibly the time required for PCP to attain an effective concentration in foot muscle. During recovery, while Pi declined to pre-exposure levels, [PA] completely recovered in only one individual. Also, realkalinization of pHi was similar to recovery of [Pi], and ATP returned to near-initial levels, as did glycerol 3-phosphate, lactate, succinate, malate, and Ala; glyceraldehyde 3-phosphate, citrate, and Gln levels declined. Recovery responses corresponded to the time for PCP clearance from foot muscle. The effects of PCP were similar to those of hypoxia, fatigue, hypersalinity, and arginine kinase inhibitors, and so sublethal PCP concentrations may also inhibit electron transport and arginine kinase as well as uncouple mitochondrial oxidative phosphorylation in intact molluscs. Thus, the effects of pollutants on key biochemical processes may now be measured in intact aquatic organisms as they occur, improving our ability to accurately assess the environmental effects of pollutants in the laboratory.  相似文献

The toxicokinetics and biotransformation of pentachlorophenol (PCP) were determined in the topsmelt (Atherinops affinis) In a static system, topsmelt (n = 9) were exposed to 50 μg/L of [U-14C]PCP for 24 hours to determine the absorption rate constant (Ka), the whole-body bio-concentration (at steady-state conditions), the elimination rate constant UQ, and the elimination half-life (t1/2). Kinetics were determined by direct quantitation of radioactivity in the exposure water. Following exposure, fish were placed in a flow-through metabolism chamber for 24 hours to allow depuration of retained residues, which were collected on XAD-4 resin. Excreted residues were identified and quantified by high-pressure liquid co-chromatography, fraction collection, and liquid scintillation counting. The Ka and Ke, calculated using a simplified model, were 0.012 M-1 0.005/h and 0.014±;0.003/h, respectively, while the 24 hour total concentration factor was 278.0×182.0 and the t1/2 was 52.7±;11.2. During 24 hours of exposure to dean seawater, topsmelt depurated 32.9% of retained residues, and while PCP was primarily excreted unchanged (64.9%), significant amounts of both pentachlorophenylsulfate (18.9%) and pentachloro-β-D-glucuronide (16.2%) were also formed.  相似文献
