Eight novel microsatellite primer pairs are presented for Penstemon rostriflorus, representing the first microsatellite markers available for this genus. Loci were characterized for 20 individuals from two populations in the Great Basin, USA. All loci are polymorphic within P. rostriflorus (seven to 13 alleles per locus; observed heterozygosity between 0.40 and 0.95), and therefore useful for population genetic studies within the species. Cross‐species transferability was tested on 40 additional species of Penstemon, and results indicate that these primers pairs will likely be useful for population genetic studies on many Penstemon species.  相似文献

Echinacoside is an important bioactive compound extracted from Cistanche tubulosa which was endangered by overexploitation. It is imperative to find an alternative source. Echinacoside was isolated from Penstemon barbatus (Can.) Roth for the first time. The peak contents of echinacoside are 9.09 ± 0.32 mg/g and 7.25 ± 0.36 mg/g respectively in the leaves and roots annually. The methanolic extracts from 20 g of dried powder of the roots of P. barbatus were pre-purified by AB-8 resin and the fraction containing echinacoside was further purified by conventional high-speed counter-current chromatography (HSCCC) and recycling HSCCC with the solvent system n-butanol–water (1:1, v/v). Totally 42.0 mg echinacoside with a purity of 96.3% was recovered. The recovery rate of echinacoside by recycling HSCCC reached 91.0%. The structure of our echinacoside confirmed by IR, 1H NMR and 13C NMR is identical to the standard sample. This indicates that P. barbatus might be ideal source for preparation of large scale of echinacoside.  相似文献

By setting out arrays of potted plants of Penstemon strictus,I tested whether freely foraging bumblebee (Bombus spp. ) workerswould establish regular foraging routes that reflected the geometryof the array. They did, passing through an asymmetrical arrayin a pattern that minimized interplant flight distances. Afterthe array was changed to a symmetrical pattern, however, theexperienced bees continued to show their previous asymmetricalflight patterns. New bees without experience on the asymmetricalarray showed no asymmetry on the symmetrical array. I term thispersistence of flight-path geometry "trapline holdover, " anddiscuss its implications for the study of animals' learningand foraging behavior.  相似文献

Employing a capillary-scale NMR probe enables the miniaturisation of structure determination and de-replication of purified natural products from plants using only 5-100 microg of material. Approximately 5 microg are required to perform one-dimensional proton and two-dimensional homonuclear (COSY and NOESY) NMR experiments; some 30 microg are needed to acquire HMQC- or HSQC-NMR spectra; ca. 75-100 microg are necessary to measure HMBC-NMR spectra; and around 200 microg of a compound are needed to perform 13C- and DEPT-NMR experiments. In order to illustrate the integration of the outputs from high-throughput natural product chemistry methods with the capabilities of the state-of-the-art CapNMR technology, the preparation of a natural product library from the extract of Penstemon centranthifolius, and the subsequent isolation, purification and structure determination of six known iridoid glycosides with 25-300 microg of material are presented.  相似文献

Trapline foraging—repeated sequential visits to a seriesof feeding locations—presents interesting problems seldomtreated in foraging models. Work on traplining is hampered bythe lack of statistical, operational approaches for detectingits existence and measuring its strength. We propose severalstatistical procedures, illustrating them with records of interplantflight sequences by bumble bees visiting penstemon flowers.An asymmetry test detects deviations from binomial expectationin the directionality of visits between pairs of plants. Severaltests compare data from one bee to another frequencies of visitsto plants and frequencies of departures to particular destinationsare compared using contingency tables; similarities of repeatedsequences within bees are compared to those between bees bymeans of sequence alignment and Mantel tests. We also comparedobserved movement patterns to those generated by null modelsdesigned to represent realistic foraging by non-traplining bees,examining: temporal patterns of the bee's spatial displacementfrom its starting point using spectral analysis; the varianceof return times to particular plants; and the sequence alignmentof repeated cycles within sequences. We analyzed the temporal structure of visitation by bumble beeworkers to a single Penstemon strictus plant growing in an arrayof conspecifics. When tested against a null distribution usinga randomization model, the observed pattern of arrivals forthe whole group of bees was random, but departures were clusteredin time. Certain individuals visited the plant repeatedly andfrequently throughout the day. These showed significantly regulararrival and departure schedules, which were likely producedby traplining. We explored whether these more frequent and regularforagers gained a higher reward than random or incidental plantvisitors. Using an analytical model, Possingham predicted thata dominating forager that visited a simple, renewing resourcein a regular pattern would garner higher and less variable rewardsthan random visitors. Inspired by these results, but interestedin plant-level visitation, we constructed a simulation modelof resource dynamics for a multiflowered plant with high visitation.The model incorporates the observed visitation schedules ofall bees and independent reward dynamics for each flower onthe plant. We calculated the rewards that observed bees wouldhave collected given a range of resource-renewal parameters.More frequent visitors did not return to the plant when whole-plantresource levels were higher, but these visitors did get greaterrewards. Their increased reward resulted from greater foragingefficiency, primarily through selecting (on average) more rewardingflowers than those selected by less frequent, random visitors.  相似文献

Floral phenotypes may be as much the result of selection for avoidance of some animal visitors as selection for improving the interaction with better pollinators. When specializing on hummingbird-pollination, Penstemon flowers may have evolved to improve the morphological fit between bird and flower, or to exclude less-efficient bees, or both. We hypothesized how such selection might work on four floral characters that affect the mechanics of pollen transfer: anther/stigma exsertion, presence of a lower corolla lip, width of the corolla tube, and angle of flower inclination. We surgically modified bee-pollinated P. strictus flowers changing one trait at a time to make them resemble hummingbird-pollinated P. barbatus flowers, and measured pollen transfer by bumblebees and hummingbirds. Results suggest that, apart from 'pro-bird' adaptations, specific 'anti-bee' adaptations have been important in shaping hummingbird-flowers. Moreover, some trait changes may have been selected for only if changing in concert with other traits.  相似文献

In the clade of Penstemon and segregate genera, pollination syndromes are well defined among the 284 species. Most display combinations of floral characters associated with pollination by Hymenoptera, the ancestral mode of pollination for this clade. Forty-one species present characters associated with hummingbird pollination, although some of these ornithophiles are also visited by insects. The ornithophiles are scattered throughout the traditional taxonomy and across phylogenies estimated from nuclear (internal transcribed spacer (ITS)) and chloroplast DNA (trnCD/TL) sequence data. Here, the number of separate origins of ornithophily is estimated, using bootstrap phylogenies and constrained parsimony searches. Analyses suggest 21 separate origins, with overwhelming support for 10 of these. Because species sampling was incomplete, this is probably an underestimate. Penstemons therefore show great evolutionary lability with respect to acquiring hummingbird pollination; this syndrome acts as an attractor to which species with large sympetalous nectar-rich flowers have frequently been drawn. By contrast, penstemons have not undergone evolutionary shifts backwards or to other pollination syndromes. Thus, they are an example of both striking evolutionary lability and constrained evolution.  相似文献

Natural selection is thought to have shaped the evolution of floral scent; however, unlike other floral characters, we have a rudimentary knowledge of how phenotypic selection acts on scent. We found that floral scent was under stronger selection than corolla traits such as flower size and flower color in weakly scented Penstemon digitalis. Our results suggest that to understand evolution in floral phenotypes, including scent in floral selection, studies are crucial. For P. digitalis, linalool was the direct target of selection in the scent bouquet. Therefore, we determined the enantiomeric configuration of linalool because interacting insects may perceive the enantiomers differentially. We found that P. digitalis produces only (S)-(+)-linalool and, more interestingly, it is also taken up into the nectar. Because the nectar is scented and flavored with (S)-(+)-linalool, it may be an important cue for pollinators visiting P. digitalis flowers.  相似文献
