Summary The sequence of the synthesis and transport of the organic matrix of spicules has been elucidated in the gorgonian Leptogorgia virgulata by use of 3H-aspartic acid as the tracer in electron-microscopic autoradiography. The entire process of matrix synthesis and transport takes approximately 2 h. It seems that the protein moiety of the organic matrix is synthesized in the RER prior to 5 min following the initial 10 min incubation in the tracer. At the 5 min chase the label is moving from the RER to the Golgi complexes where the carbohydrate moiety of the matrix is presumed to be synthesized. At the 5 to 15 min chases the label is transported out of the Golgi complexes via Golgi vesicles. This phase continues for 30 min. From 60 to 120 min the 3H-aspartic acid moves to the spicules. After 120 min the majority of the label has moved into the spicules. Silver grain counts over both multivesicular and electron-dense bodies remain at relatively low and constant levels over 4 h indicating that neither organelle is involved in the synthesis and transport of the organic matrix.Contribution No 512; Belle W. Biomass production and plant species diversity in grassland in southern England was monitored before and after a change from conventional to organic farming. Our 18-year study, part of the UK's Environmental Change Network long-term monitoring programme, showed that the cessation of artificial fertiliser use on grassland after conversion to organic farming resulted in a decrease in biomass production and an increase in plant species richness. Grassland productivity decreased immediately after fertiliser application ceased, and after two years the annual total biomass production had fallen by over 50%. In the subsequent decade, total annual grassland productivity did not change significantly, and yields reached 31–66% of the levels recorded pre-management change. Plant species richness that had remained stable during the first 5 years of our study under conventional farming, increased by 300% over the following 13 years under organic farm management. We suggest that the change in productivity is due to the altered composition of species within the plots. In the first few years after the change in farming practice, high yielding, nitrogen-loving plants were outcompeted by lower yielding grasses and forbs, and these species remained in the plots in the following years. This study shows that grassland can be converted from an environment lacking in plant species diversity to a relatively species-rich pasture within 10–15 years, simply by stopping or suspending nitrogen additions. We demonstrate that the trade-off for increasing species richness is a decrease in productivity. Grassland in the UK is often not only managed from a conservation perspective, but to also produce a profitable yield. By considering the species composition and encouraging specific beneficial species such as legumes, it may be possible to improve biomass productivity and reduce the trade-off.  相似文献

Proteolysis of the hydroxylase component of soluble methane monooxygenase (MMO) with trypsin yielded a protein which retained 50% activity in a standard MMO assay. In an H2O2-driven assay, in which H2O2 replaced two of the protein components, NADH and O2 used in the standard assay, the proteolysed hydroxylase retained full activity for ethane, propane and propene, but had a 2–3 fold increase with methane as substrate. Several crosslinking reagents have been tested for their ability to stabilise the proteolysed form of the hydroxylase. Using polyoxyethylene bis(imidazolyl carbonyl) (Mr 3350) as the crosslinking agent, increased thermostability of the hydroxylase was observed. Activated methoxypolyethylene glycol (Mr 5000) was used to modify the hydroxylase which was now soluble in organic solvents as well as water and could be activated by H2O2. The glycol-modified hydroxylase functioned well in organic solvents in the catalysis of propene oxidation.  相似文献

Two experiments were conducted in central southern England between September 1994 and August 1996 to identify the critical weed-free period in organically grown winter wheat (Triticum aestivum, cv. Mercia). In competition with a mixed weed infestation of predominately Alopecurus myosuroides and Tripleurospermum inodorum it was found that wheat yield decreased as the duration of the weed-infested period increased and that the crop needed to be kept free of weeds from sowing in order to completely avoid any yield loss. Also, weeds emerging in the wheat crop (predominately T. inodorum) during the growing season had a significant and detrimental effect on yield. The existence of the critical period, therefore, depends on the imposition of an acceptable yield loss. If a 5% yield loss gives a marginal benefit compared with the cost of weed control, the critical period will begin at 506°C days after sowing (November) and end at 1023°C days after sowing (February). This information could be used by farmers to target mechanical weeding operations to control weeds at a time that will have maximum benefit to the crop.  相似文献

Summary The rhizosphere microflora of arecanut palm under continuous application of organic manures and inorganic fertilizers was studied. The nutrients applied are 100 g N, 40 g P2O5 and 140 g K2O/palm/year in the form of organics and inorganics. The application of organic manure increased the microbial population. The increase in microbial population was observed between the rhizosphere samples collected at 0–30cm and 30–60 cm depths. Summary Soil temperature, moisture, and CO2 were monitored at four sites along an elevation transect in the eastern Mojave Desert from January to October, 1987. Climate appeared to be the major factor controlling CO2 partial pressures, primarily through its influence of rates of biological reactions, vegetation densities, and organic matter production. With increasing elevation, and increasing actual evapotranspiration, the organic C, plant density, and the CO2 content of the soils increased. Between January and May, soil CO2 concentrations at a given site were closely related to variations in soil temperature. In July and October, temperatures had little effect on CO2, presumably due to low soil moisture levels. Up to 75% of litter placed in the field in March was lost by October whereas, for the 3 lower elevations, less than 10% of the litter placed in the field in April was lost through decomposition processes.  相似文献

The use of phytoplankton as test organisms in bioassays has recently gained momentum due to their simplicity, availability, sensitivity, rapidity of analysis, and cost-effectiveness. Increasing emphasis is currently being given to field and in situ experiments using indigenous populations, particularly ultraplankton/picoplankton (2–20 m) which play a key role in the microbial loop and food chain dynamics. Impact evaluation can be determined at the structural, ultra-structural, and functional level. An array of techniques is available for toxicity testing including the use of either algal cultures or natural assemblages in laboratory or in situ experiments, the selection of which depends on the objectives, precision required, and project budget of the particular study. An overview is presented of the various procedures using algae in toxicity testing with a focus on the Great Lakes and an emphasis on field techniques. The effective use and application of such sensitive technology has tremendous potential for early warning detection of ecosystem perturbations in concert with a multi-trophic battery of tests.  相似文献

Various components of the Microbial loop such as bacteria, heterotrophic nanoflagellates and autotrophic picoplankton were analyzed, for the first time across the Great Lakes, during a cruise in the summer of 1988. In addition, the size fractionated primary productivity using carbon-14 techniques was also determined. The statistical analysis indicated that bacteria, autotrophic picoplankton and ultraplankton/picoplankton productivity were significantly higher in Lakes Ontario and Erie than Lakes Huron and Michigan. The autotrophic picoplankton and ultraplankton/picoplankton productivity was higher in Lake Erie compared to Lake Ontario.The autotrophic picoplankton showed sensitivity to nutrients and contaminants in various types of environments. A dramatic decrease of autotrophic picoplankton in eutrophic-contaminated areas, such as Ashbridges Bay, Hamilton Harbour and western Lake Erie was observed. Conversely, in Saginaw Bay, another eutrophic environment, the autotrophic picoplankton were significantly higher than in Lake Huron. The sensitivity of autotrophic picoplankton to nutrients/contaminants might have implications to trophic interactions. Our results suggest that structural and functional characteristics of the microbial loop may be operating differently in stressed versus unstressed ecosystems. The possibility of using autotrophic picoplankton as an early warning indicator of environmental perturbation is proposed.  相似文献

Summary Relationships between fine root growth, rates of litter decomposition and nutrient release were analysed in a mixed forest on Tierra Firme, a Tall Amazon Caatinga and a Low Bana on podsolized sands near San Carlos de Rio Negro. Fine root growth in the upper soil layers (root mat+10 cm upper soil) was considerably higher in the Tierra Firme forest (1117 g m-2 yr-1) than in tall Cattinga (120) and Bana (235). Fine root growth on top of the root mat was stimulated significantly by added N in Tall Caatinga and Low Bana forests, by P in Tierra Firme and Bana forests, and by Ca only in the Tierra Firme forest. Rate of fine root growth in Tierra Firme forest on fresh litter is strongly correlated with the Mg and Ca content of litter. Rate of litter decomposition was inversely related to % lignin and the lignin/N ratio of litter. Litter contact with the dense root mat of the Tierra Firme increased rates of disappearance for biomass, Ca and Mg as compared with litter permanently separated or lifted weekly from the root mat to avoid root attachment. Nitrogen concentration of decomposing litter increased in all forests, net N released being observed only in Caryocar glabrum and Aspidosperma megalocarpum of the Tierra Firme forest after one year of exposure. Results emphasize the differences in limiting nutrients in amazonian forest ecosystems on contrasting soil types: Tierra Firme forests are particularly limited by Ca and Mg, while Caatinga and Bana forests are limited mainly by N availability.  相似文献

Milk samples from the stomachs of harp seal pups were analysed for Cu, Zn, Se, Cd and Hg, as were liver, kidney, and muscle from mother-pup pairs. Tissues were also analysed for MeHg. Milk contained, in addition to essential trace metals, Cd and Hg (57 ng/g and 6.5 ng/g respectively).
Pups had mercury in all three tissues. The percent methyl mercury in liver of pups was higher than in liver of mothers. Mercury in muscle was mostly methyl mercury in both mothers and pups. Total mercury in liver of mothers but not pups was correlated positively with selenium. Estimates of ingested mercury by pups indicated they had acquired most of their mercury during gestation.
Although mothers had cadmium in liver and kidney, it was not detected in tissues of pups. Cadmium did not transfer across the placenta, while mercury did. Tissue concentrations of Cu and Zn were higher in pups than mothers. The presence of metallothionein in pup tissues was postulated.
A strong positive correlation of copper and selenium between mothers and pups indicated transfer of these elements from mother to pup in direct proportion to their concentrations in maternal liver and kidney.  相似文献
