ABSTRACT. Sequences of the small subunit rRNA genes of Amblyospora californica and an Amblyospora sp. from Culex salinarius were determined. These sequences were compared phylogenetically with 16 other microsporidia. The results suggest Amblyospora forms a sister taxon to the rest of the microsporidia examined. The basal position of Amblyospora is discussed with respect to the evolution of microsporidian life cycles. These sequences represent the longest microsporidian small subunit rRNA genes sequenced to date, 1,359 and 1,358 bp, respectively. Structural features and GC content (49% for both) are comparable to those of other microsporidia which have been sequenced.  相似文献

SYNOPSIS. Nosema parkeri sp. n. is described from nymphs and adults of the argasid tick, Ornithodoros parkeri Cooley, from a laboratory colony. Schizogonic and sporogonic stages are described from various tick tissues. Spores are binucleate, measuring 3.2 (3–4) × 1.9 (1.8–2.5) μm. Transmission is transovarial and transstadial. The parasite does not appear to affect adversely the development or reproduction of the tick. Dermacentor andersoni Stiles was experimentally infected. Attempts to infect Swiss mice by tick feeding or by injection of infected tick suspensions were unsuccessful. The microsporidan differs in structure from Encephalitozoon ixodis Weiser) and Nosema slovaca Weiser & Reháček, the only other microsporidans known from ticks.  相似文献

RESUME. La Microsporidie Nosema bombycis, Protozoaire parasite agent de la pébrine du ver à soie, a étéétudiée cytochimiquement à la fois en microscopie photonique et électronique. Les examens ont porté sur la détection et la localisation des acides nucléiques (ADN et ARN), des polysaccharides, de la phosphatase acide, au cours des différents stades du développement dans les cellules de I'hôte (du schizonte à la spore). Les principaux résultats concernent les observations relatives aux polysaccharides et à la phosphatase qui ne sont détectés qu'au stade de la spore et ne sont pas observés au stade du schizonte. Les polysaccharides sont présents au niveau du sac polaire, du filament polaire et sur la membrane cytoplasmique; la phosphatase acide est localisée au niveau du sac polaire, du filament polaire et dans la vacuole postérieure. SYNOPSIS. Nosema bombycis, agent of pebrine disease of silkworm, was studied cytochemically, using both light and electron microscopy. Presence of nucleic acids (DNA and RNA), polysaccharides, and acid phosphatases was demonstrated and localization of these substances was determined in various stages of the parasite (from the schizont to the spore). DNA and RNA were detected in all these stages. Polysaccharides and acid phosphatase were found in the spore but not in the schizogonic stages. Polysaccharides were detected in the polar cap, the polar filament, and the limiting membrane of the cytoplasm of the spore. Acid phosphatase was found in the polar cap, the polar filament, and the posterior vacuole.  相似文献

A new species of microsporidia from Drosophila melanogaster was investigated by light and electron microscopy and by ribosomal RNA (rRNA) sequencing. This microsporidium and the previously described Nosema kingi and Nosema acridophagus have been transferred to the new genus Tubulinosema gen. nov. with the following characters: nuclei are in diplokaryotic arrangement during the life cycle. All stages are in direct contact with the host cell cytoplasm, slightly anisofilar polar tube with the last coils being smaller in diameter arranged in one or two rows on both sides of the diplokaryon and small tubuli on the surface of late meronts. Spores are oval or slightly pyriform. Thick endospore wall, thinner over anchoring disc. This new genus and the genus Brachiola have been placed in a new family Tubulinosematidae fam. nov. Phylogenetic analysis of small subunit rRNA sequences by different methods placed Tubulinosema spp. in one clade with the genus Brachiola forming its sister clade, which is distant from the clade containing the true Nosema spp. including Nosema bombycis.  相似文献

The microsporidian Nosema antheraeae is a pathogen that infects the Chinese oak silkworm, Antheraea pernyi. We sequenced the complete small subunit (SSU) rRNA gene and the internal transcribed spacer (ITS) of N. antheraeae, and compared the SSU rRNA sequences in other microsporidia. The results indicated that Nosema species, including N. antheraeae, formed two distinct clades, consistent with previous observations. Furthermore, N. antheraeae is clustered with N. bombycis with high bootstrap support. The organization of the rRNA gene of N. antheraeae is LSU-ITS1-SSU-ITS2-5S, also following a pattern similar to the Nosema type species, N. bombycis. Thus, N. antheraeae is a Nosema species and has a close relationship to N. bombycis.  相似文献

A local variety of sorghum (Serena) was planted in a screenhouse and infested with first‐instar Chilo partellus larvae. Sorghum rows were then inoculated at random with an aqueous suspension of Nosema marucae containing 1.6 ×108 spores ml‐1 at 2‐day intervals from 1 to 16 days after infestation (DAI). Foliar damage was low in plants inoculated with the pathogen at 1 and 3 DAI, where the highest score was 0.2 on a scale of 0–9. However, leaf damage increased rapidly as the interval between infestation and spraying of N. marucae became longer. At 19 DAI the foliar damage score on plants that had been inoculated at day 16 was 8.4. Similarly, the proportion of plants with either fully or partially formed heads decreased from 97.9% in plants sprayed at 3 DAI, to 10.8% at 11 DAI and 0% at 16 DAI. The timing of application of N. marucae is therefore important in the utilization of the microsporidian for the biological control of stem‐boring lepidopterans. This study suggests that a single, well timed application of Nosema spores can effectively control C. partellus for up to 19 days following oviposition.  相似文献

家蚕微孢子虫孢壁蛋白与其发芽的相关性   总被引:3,自引:0,他引:3  
为了研究孢壁蛋白与家蚕微孢子虫发芽(孢原质弹出)的相关性,我们采用碳酸钾诱导微孢子虫体外发芽结合密度梯度离心的方法(简称GDGC法),收集纯化发芽后的孢子空壳(简称孢壳),对发芽液、纯化的孢壳及成熟孢子的孢壁蛋白组分进行了分析。结果表明:GDGC法可以获得高纯度孢壳,计算出其密度为1.130g/cm^3;与发芽前成熟孢子提取的孢壁蛋白相比,空孢壳可以提取到主要孢壁蛋白SWP32、SWP30、SWP25,同时发现SWP32、SWP25丰度有所降低;结合碳酸钾发芽液的蛋白电泳分析,发现孢壳上丰度降低的SWP32在发芽液蛋白样品中存在,LC—MS/MS数据分析也发现SWP32、SWP30、SWP25在碳酸钾处理液中都有存在;而用碳酸钾溶液处理冷冻孢子时,未观察到发芽现象,电泳结果显示此时K2CO,溶液中只有SWP30条带,说明在碳酸钾溶液诱导的发芽过程中SWP32和SWP25从孢壳上脱落可能与发芽相关而不是被碱性的碳酸钾溶解下来的[动物学报54(6):1068—1074,2008]。  相似文献

【目的】本研究旨在对前期鉴定到的nce-miR-34537进行表达和序列验证,预测nce-miR-34537的靶基因并明确其分子特性,进而检测nce-miR-34537及其靶基因在东方蜜蜂微孢子虫(Nosema ceranae)侵染意大利蜜蜂(Apis mellifera ligustica)工蜂过程的表达谱,为进一步探究nce-miR-34537调控东方蜜蜂微孢子虫侵染的功能和作用机制提供基础。【方法】通过Stem-loop-RT-PCR和Sanger测序验证nce-miR-34537的表达和序列。通过生物信息学软件预测nce-miR-34537的靶基因PIP5KI(I型磷脂酰肌醇4-磷酸-5-激酶基因)的理化性质等分子特性和保守基序,并构建基于氨基酸序列的系统进化树。采用RT-qPCR检测nce-miR-34537及其靶基因的表达谱。【结果】nce-miR-34537在东方蜜蜂微孢子虫孢子中真实存在和表达。nce-miR-34537共靶向PIP5KI等151个基因。PIP5KI蛋白的分子式为C882H1 364N226  相似文献

1. Bumblebees are important pollinators in North America and are attacked by a range of parasites that impact their fitness; however, few studies have investigated the extent or causes of parasitism in North America. 2. This study used a 2‐year multi‐site survey of bumblebee parasitism to ask: (i) how common are parasitoid conopid flies and the internal parasites Crithidia bombi and Nosema bombi in Massachusetts; and (ii) what factors are correlated with parasitism? 3. Infection rates by all three parasites were higher in this study than previously documented in North America. Overall, conopids infected 0–73% of bees in each sample, C. bombi infected 0–82% of bees, and N. bombi infected 0–32%. 4. Conopid flies infected female bees more than males and intermediate‐sized bees more than large or small bees. Crithidia bombi infection rates were higher in certain bee species and sites, and exhibited a unimodal pattern of prevalence over time. Nosema bombi parasitism was higher in male than female bees. 5. Infection by N. bombi in two rare bumblebee species was higher than expected based on parasitism rates of common bee species but C. bombi infection was lower. If high prevalence of N. bombi in these bumblebee species is common, parasitism may be a potential cause of their decline. 6. Given the documented effects of these parasites, the high levels of infection may affect bee populations in Massachusetts and threaten the stability of their valuable ecosystem services.  相似文献
