Methods for production of containerized seedlings ofAlnus species were developed which permit nitrogen-fixing nodules to form on the root systems prior to outplanting, in order to provide an early nitrogen input during seedling establishment. The methods are based on procedures for inoculating root systems with suspensions ofFrankia (Actinomycetales), applied either directly in the container cell as a soil drench at the time of seeding, or as a root dip for seedlings transplanted into the containers. Germination of dried, stored seed was enhanced by light and by presoaking for 16 h in water. Pretreatments to overcome seed dormancy or to eliminate fungal pathogens did not further enhance germination. Some loss of seedlings was recorded in the early stages of growth shortly after germination, which is a factor in calculating projected seedling yield. Nodulation and seedling growth were evaluated in terms of growth media characteristics. Seedlings performed well in peat-vermiculite, at soil pH between 5.5 and 8.0.  相似文献

A 3-deoxy-2-heptulosaric acid (DHA), very probably with the lyxo-configuration, was identified in the R-core region of lipopolysaccharides from nodulating strains of Rhizobium leguminosarum, Rhizobium meliloti and from all three biovars of the phytopathogenic Agrobacterium tumefaciens. Its structure could be deduced from the fragmentation pattern of the corresponding alditol acetates obtained after reduction of the 2-keto and the 1.7-carboxy groups by sodium borohydride or sodium borodeuteride. DHA in lipopolysaccharide was not destroyed by periodate and is therefore not in a terminal position. Two DHA-containing oligosaccharides, namely glucosyl (1----4)-3-deoxy-2-heptulosaric acid and rhamnosyl-rhamnosyl-(1----5)-3-deoxy-2-heptulosaric acid could be tentatively identified by mass spectrometric methods amongst the products of mild acidic hydrolysis of lipopolysaccharides of Rhizobium leguminosarum strain 24. The two types of non-nodulating mutants of Rhizobium leguminosarum included in this study did not contain 3-deoxy-2-heptulosaric acid.  相似文献

Summary Two methods have been developed in order to discriminate between lateral roots, nodules and root-derived structures which exhibit both root and nodule histological features and which can develop on legumes inoculated with certainRhizobium mutants. The first method, known as the clearing method, allows the observation by light microscopy of cleared undissected root-structures. The second, known as the slicing method, is a complementary technique which provides a greater degree of structural information concerning such structures. The two methods have proved invaluable in defining unequivocally the nature of the interaction between a rhizobial strain and a legume host.  相似文献

To date, the sequences of 45 Bradyrhizobium japonicum genes are known. This provides sufficient information to determine their codon usage and G+C content. Surprisingly, B. japonicum nodulation and NifA-regulated genes were found to have a less biased codon usage and a lower G+C content than genes not belonging to these two groups. Thus, the coding regions of nodulation genes and NifA-regulated genes could hardly be identified in codon preference plots whereas this was not difficult with other genes. The codon frequency table of the highly biased genes was used in a codon preference plot to analyze the RSRj9 sequence which is an insertion sequence (IS)-like element. The plot helped identify a new open reading frame (ORF355) that escaped previous detection because of two sequencing errors. These were now corrected. The deduced gene product of ORF355 in RSRj9 showed extensive similarity to a putative protein encoded by an ORF in the T-DNA of Agrobacterium rhizogenes. The DNA sequences bordering both ORFs showed inverted repeats and potential target site duplications which supported the assumption that they were IS-like elements.  相似文献

Summary Six carbamate pesticides namely 1-naphthol, sevin, dimetilan, trematan, NaDDC and dymid were studied to see their effect on nodulation and nitrogen fixation inPisum sativum andVigna sinensis. Low concentrations of the pesticides have little effect on nodulation and nitrogen fixation, whereas higher concentrations adversely effect these processes. The results also indicate that then sensitivity depends upon the species of the Rhizobium and also the type of the pesticide. Pesticides belonging to the carbamate group differ in their capacity to affect nodulation and nitroge fixation.  相似文献

Summary Bean (Phaseolus vulgaris L. Cv. Burpees Stringless) seeds were subjected to two cycles of presowing soaking and drying treatments with sodium molybdate and cobalt nitrite at 1 and 5 ppm concentrations used separately and also in combination. Sodium molybdate 2 ppm and cobalt nitrite 1 ppm used singly proved better than the remaining treatments with respect to nodulation, dry matter, nitrogen and yield. Combined treatment with sodium molybdate and cobalt nitrite did not produce additive effect on any parameter studied compared to their usage alone.  相似文献

Multiple endonuclease digestion of template DNA or amplification products can increase significantly the detection of polymorphic DNA in fingerprints generated by multiple arbitrary amplicon profiling (MAAP). This coupling of endonuclease cleavage and amplification of arbitrary stretches of DNA, directed by short oligonucleotide primers, readily allowed distinction of closely related fungal and bacterial isolates and plant cultivars. MAAP analysis of cleaved template DNA enabled the identification of molecular markers linked to a developmental locus of soybean (Glycine max L. Merrill). Ethyl methane sulfonate (EMS)-induced supernodulating, near-isogenic lines altered in the nts locus, which controls nodule formation, could be distinguished from each other and from the parent cultivar by amplification of template pre-digested with 2–3 restriction enzymes. A total of 42 DNA polymorphisms were detected using only 19 octamer primers. In the absence of digestion, 25 primers failed to differentiate these soybean genotypes. Several polymorphic products co-segregated tightly with the nts locus in F2 families from crosses between the allelic mutants nts382 and nts1007 and the ancestral G. soja Sieb. & Succ. PI468.397. Our results suggest that EMS is capable of inducing extensive DNA alterations, probably around discrete mutational hot-spots. EMS-induced DNA polymorphisms may constitute sequence-tagged markers diagnostic of specific genomic regions.  相似文献

Summary Most soybean varieties fail to nodulate effectively in tropical soils unless inoculated with a competitive strain ofRhizobium japonicum. Developing countries in the tropics, with few exceptions, lack inoculant industries to produce and distribute viable inoculants to small farmers and extension programs to teach them to use inoculant. Several soybean genotypes have been identified that nodulate effectively with many strains of the cowpea inoculation group which is ubiquitous in tropical soils of Africa. Soybean genotypes that nodulate and grow well without inoculant application are called promiscuous. Summary The effect of exogenous applications of gibberellins (GAs) or the growth retardant -chloroethyltrimethylammonium chloride (CCC) on root nodule formation and activity (C2H2-reduction) in soya was studied. Daily foliar application of GA3 (2.89×10–6 M) delayed the formation of nodule initials and reduced the numbers mass nodule–1 and specific activity of nodules by 43%, 31% and 47% respectively, without affecting plant growth. Similar effects on nodulation were produced by foliar application of GA4 (3.01×10–5 M) or GA7 (3.03×10–5 M), or by the addition of GA3 (2.89×10–6 M) to the rooting medium. GA effectiveness in reducing nodule numbers was decreased by delaying its application until after the initial infection process had occurred, but the nodules formed were smaller and less active than those of the untreated control plants. The GA effect on nodulation and nodule activity was not associated with alterations in root exudate or due to a direct inhibitory effect of the hormone on the nitrogenase system. When the endogenous root content of GA-like substances was reduced (86% decrease) by foliar application of CCC (6.30×10–5 M), nodule numbers were increased by 56%, but nodule size and total nodule activity were similar to those of control plants. The GA and CCC treatments had no effect on rhizobial growth in liquid culture nor on root colonisation by rhizobia.The results suggest that the endogenous content of root GA may have a regulatory role in both the infection process and in subsequent nodule morphogenesis, thus controlling both the number and effectiveness of the root nodules formed.  相似文献

Summary Greenhouse experiments were done with two purposes: (1) to identify strains of rhizobia effective and acid-tolerant in symbiosis withLablab purpureus, and (2) to determine whether soil acidity or the symbiotic condition increased the phosphate requirement for growth.Five rhizobial strains were tested in one neutral soil, two acid soils, and the two acid soils limed to pH 6.6. In the neutral and limed soils, three of the strains were effective (CB1024, CB756, TAL169), but only two strains (CB756, TAL169) remained effective in acid soil.Strain CB756 and plus-N treatments were further compared in a factorial trial involving combinations of five levels of P with lime, no lime and CaCl2 treatments, applied to an acid soil. Some of the treatments were also applied to plants inoculated with CB1024. Between the N-fertilized and CB756 treatments there was no clear difference in growth response to applied P, and the critical internal concentration of P for 95% of maximal growth was the same (0.22% shoot dry weight). Increasing P beyond levels needed for maximal growth increased nodulation and N concentration in plants inoculated with CB756. It lowered N concentration in N-fertilized plants. There was evidence suggesting that the P requirement of symbiotic plants increased if the soil was acid, or if CB756 were replaced by CB1024 as microsymbiont; but the critical statistical interactions were not significant.  相似文献
