Summary. In the present study we describe a simple and fast method to measure the concentration of total free amino acids in very
small amounts of biological tissues. The procedure described here is based on the reaction of free amino acids with o-phthaldialdehyde (OPA) in the presence of a reducing agent, β-mercaptoethanol (MET), to give a complex which can be measured by fluorescence. It is a very rapid process and has the same
reliability as the conventional ninhydrin method of Moore and Stein but is about 500 times more sensitive. The sensitivity
of the new protocol is such to permit the determination with high reliability of very small amounts of free amino acids at
picomole levels, either in a standard amino acid mixture or in biological tissues, without chromatographic separation of the
amino acids. It is particularly useful when the amount of the sample is very low, e.g. on a single pituitary or pineal gland
of small animals or on single cells, such as oocytes or eggs, as well as single ganglions or axons of marine invertebrates.

Summary. Identification of amino acids is extremely important for the evaluation of protein structure. Thin layer chromatography is
an important tool for detecting amino acids by variety of spray reagents. Among these ninhydrin is the most popular due to
its high sensitivity. However, ninhydrin produces the same purple/violet color with most amino acids. A spray reagent with
high sensitivity for easy and rapid identification of amino acids on thin-layer plates has been introduced.

Until now quantification of proteins in gel-based methods relies on the amount of protein loaded onto the gel. This does not, however, represent the amount of proteins in the gel and therefore determination of proteins within the gel is mandatory. A method to quantify proteins, both hydrophilic and hydrophobic, using in-gel digestion with proteases, subsequent acid hydrolysis and the use of the ninhydrin reaction is herein presented. 相似文献

谷氨酸和茚三酮可以发生特有的显色反应 ,在 pH 5~ 6时产物蓝紫色最明显。其颜色深浅与谷氨酸浓度大小有关。用分光光度计可以在其最大光吸收波长处测定其OD值。分别以OD值和相应的一系列浓度的L谷氨酸纯品浓度值为纵坐标和横坐标 ,绘制出 1条标准曲线。谷氨酸发酵液中的谷氨酸含量值即可通过分光光度计测得它与茚三酮在最适 pH处反应得到的蓝紫色产物的OD值 ,然后从上述标准曲线上查到。误差率在 1 0 %左右。这为谷氨酸定量测定提供了一种简单实用的方法。 相似文献

Contents of the funduses and ducts of the postacetabular glands of Schistosoma mansoni cercariae, the secreted deposits, and the surface film were compared by their histochemical reactions. Techniques for carbohydrate-containing substances, neutral and acid mucosubstance, proteins and amino acids, and enzymes were used. The secretion reacted differently before (within the glands) and after (in secreted deposits) emission.Before emission, the postacetabular gland contents reacted as a neutral mucosubstance containing periodate-engendered and periodate-reactive aldehydes rich in vic-glycols or their substituted amines, probably hexoses other than glucose, such as fucose or galactose. No reactions of significance were observed for acid groups or for glycogen or lipids. In this state, the secretion is termed mucigen.After emission, the secretion stained not only as mucigen, but also as acid mucosubstance, apparently sialomucin. After emission, it is termed mucin.It is probable that acid radicals were present in mucigen but were masked stearically by the presence of adjacent neutral radicals or basic proteins. The surface film reacted as both a neutral and an acid mucosubstance. Evidence suggested that the film itself was neutral and that the reaction for acid mucosubstance was from an overlay of mucin secreted from the postacetabular glands.Proteins and amino acids, especially arginine, and some tyrosine and tryptophan were indicated in mucigen and in mucin by the histochemical tests. There was no histochemical evidence of enzymes. Secretion of the postacetabular glands is concluded from histochemical reactions, as from earlier chromatographic data (Stirewalt and Evans 1960), to be a carbohydrate-protein-lipid complex. 相似文献

Sealed reticulocyte ghosts were treated with reagents that modify a variety of amino acid residues. Only ninhydrin and phenylglyoxal, both modifiers of arginyl residues, produced inhibition of the initial rate of 59Fe2+ uptake. The inhibition (i) was dependent on the concentration of ninhydrin or phenylglyoxal, (ii) increased from pH 7 to 9, a feature of the modification of arginine by ninhydrin or phenylglyoxal, and (iii) was blocked when Fe2+ was present during the modification step. A23187, an effective membrane Fe2+ transporter, diminished the inhibitory effect of ninhydrin and phenylglyoxal, indicative that the transport of iron through the membrane, and not a secondary process, was selectively inhibited. We conclude that the iron transporter from the plasma membrane of erythroid cells has one or more arginyl residues in a segment accessible to ninhydrin or phenylglyoxal, and that this residue is involved in the transmembrane transport of iron.This work was supported by grant 1080-91 from FONDECYT, Chile. 相似文献

A violet coloured complex was developed when cobalt metal reacts with ninhydrin at pH 8.2, using sodium acetate buffer solution. Absorbance of the complex was measured at 395 nm. Various factors, such as volume of the ligand used, solution pH, stability of the complex with time and interference of other metals, which effect the complex formation have been studied in detail. Present developed method can be used for the spectrophotometric estimation of cobalt with ninhydrin complex. The method is simple, selective and cheap for the determination of cobalt in very less time. 相似文献

Energies corresponding to optimum geometries of ninhydrin, some of its analogs, the corresponding Ruhemann’s Purple analogs
and some of the intermediates of the reaction between ninhydrin analogs and amino acids are calculated at Hartree–Fock/6-31G**
level of theory. Such a study is significant from a forensic science point of view because of the strong interest in the forensic
chemistry and law enforcement communities in developing alternatives to the current generation of ninhydrin-like chemicals
for the detection and development of latent fingerprints. In examining our new predictions for the net energetics of the reactions
in the formation of substituted Ruhemann’s Purples, we find that a fluorine-containing analog is the most thermodynamically
feasible reaction. In light of this finding, we suggest further experimental studies to determine the kinetic feasibility
of synthesizing the fluorine-containing analog, as well as other similar molecules and to determine their spectroscopic properties. 相似文献
