Bacteria communicate at high cell density through quorum sensing, however, there are no reports about this mechanism in archaea. The archaeon Natronococcus occultus produces an extracellular protease at the end of growth. Early production of protease activity was observed when a low density culture was incubated with late exponential conditioned medium suggesting the presence of factor(s) inducing this activity. Conditioned medium and ethyl acetate extracts corresponding to the transition from exponential to stationary phase showed a positive signal in Agrobacterium biosensor. We report the detection of potential autoinducer molecules of the acylated homoserine lactone type in the archaeon N. occultus. These molecules may be responsible for the production/activation of extracellular protease.  相似文献

Isolated membranes of the extreme haloalkaliphilic archaeon Natronococcus occultus were able to hydrolyze ATP via an ATPase, which required the presence of Mg(2+), high concentrations of NaCl, and a pH value of 9. The native molecular mass of the purified ATPase was 130 kDa and was composed of 74- and 61-kDa subunits. Enzyme activity was specific for the hydrolysis of ATP with slight activity towards GTP, CTP, and ITP. The enzyme required NaCl for maximal activity but Na(2)SO(4) and (NH(4))(2)SO(4) could substitute. The enzyme showed no activity if Na(2)SO(3) or sodium citrate was substituted for NaCl. The ATPase from N. occultus was inhibited by NBD-Cl, NaN(3), and ouabain, and was sensitive to nitrate, vanadate, DCCD, and bafilomycin A(1). It was not inhibited by NEM in contrast to other previously characterized halophile ATPases. The ATPase had a K(M) of 0.5 mM and appeared to be non-competitively inhibited by NaN(3) with a K(I) of 3.1 mM.  相似文献

Natronococcus occultus, a haloalkaliphilic archaeon, was examined for its resistance to potassium tellurite. Cells grown in the presence of 1 mM potassium tellurite reduced it to metallic tellurium resulting in the deposition of intracellular crystals in the cytoplasm. The minimal inhibitory concentration for potassium tellurite was 10 mM. N. occultus had an inducible tellurite reductase activity. Cell-free extracts catalyzed the enzymatic reduction of potassium tellurite in a reaction which was dependent on NADH oxidation and a reduced environment.  相似文献

In this review, the history of the classification of the family Halobacteriaceae, the extremely halophilic aerobic Archaea, is reviewed with some emphasis on the recently described new genera Halobaculum, Halorubrum, Natrialba, Natronomonas, and "Haloterrigena." Speculation is made about the evolutionary relationship between members of the Halobacteriaceae and the extremely halophilic, anaerobic methanogens of the genera Methanohalobium and Methanohalophilus. Ultrastructure and morphogenesis of extremely halophilic neutrophilic (Halobacteriam distributum, Halococcus turkmenicus) and alkaliphilic (Natronobacterium pharaonis, Natronococcus occultus) archaeobacteria were studied. The H. distributum culture was rather polymorphous and produced cells of four types. Due to the irregular cell fission in different planes packages of various numbers of cells surrounded by a common capsule were formed. Resting forms (halocysts) with multilayer covers were present in the population. The N. pharaonis culture consisted of rod-like cells and cyst-like forms. Besides, under conditions of carbon limitation, multicellular aggregated forms were found in the culture. Encapsulated single cells and aggregated forms with a common capsule were observed in H. turkmenicus and N. occultus cultures.  相似文献

根据经典微生物划线纯培养方法从湛江一个盐田泥样中分离获得1个嗜盐碱古菌菌株SCSIOCD13T,通过形态、细胞化学分类及分子发育等对其进行系统分类鉴定研究。经鉴定该菌细胞球形,好氧,革兰染色不定,在最适pH 8.5和37℃培养条件下,能够耐受2.5~5.5 mol/L盐浓度生长。16S rRNA基因序列比对分析显示,该菌株与嗜盐碱古菌Natronococcus属成员N.jeotgali B1T(99.2%),N.occultus NCMB 2192T(96.8%)和N.amylolyticus Ah-36T(95.8%)最相近,且系统发育树和细胞化学特征与Natronococcus属相一致。然而,系统发育树上显示该菌株与最相似菌种形成单独分支,且生理学特性显示出与最相似的3个物种具有明显的差异,包括盐浓度、pH耐受,镁离子需求,H2S产生,淀粉、明胶水解,抗生素敏感性等。另外,杂交结果显示,菌株SCSIO CD13T与最相似菌种Natronococcus jeotgali B1T的杂交值为23%;综合多相分类结果,菌株SCSIO CD13T应为Natronococcus属的一个新成员,将其命名为湛江嗜盐碱球菌新种(Natronococcus zhanjian-gensis),典型菌株SCSIO CD13T。  相似文献

A serine protease secreted by the haloalkaliphilic archaeon Natrialba magadii at the end of the exponential growth phase was isolated. This enzyme was purified 83 fold with a total yield of 25% by ethanol precipitation, affinity chromatography, and gel filtration. The native molecular mass of the enzyme determined by gel filtration was 45 kDa. Na. magadii extracellular protease was dependent on high salt concentrations for activity and stability, and it had an optimum temperature of 60°C in the presence of 1.5 M NaCl. The enzyme was stable and had a broad pH profile (6–12) with an optimum pH of 8–10 for azocasein hydrolysis. The protease was strongly inhibited by diisopropyl fluorophosphate (DFP), phenylmethyl sulfonylfluoride (PMSF), and chymostatin, indicating that it is a serine protease. It was sensitive to denaturing agents such as SDS, urea, and guanidine HCl and activated by thiol-containing reducing agents such as dithiotreitol (DTT) and 2-mercaptoethanol. This protease degraded casein and gelatin and showed substrate specificity for synthetic peptides containing Phe, Tyr, and Leu at the carboxyl terminus, showing that it has chymotrypsin-like activity. Na. magadii protease presented no cross-reactivity with polyclonal antibodies raised against the extracellular protease of Natronococcus occultus, suggesting that although these proteases share several biochemical traits, they might be antigenically unrelated. Strains of Halobacteria from an Algerian culture collection were screened for their lipolytic activity against p-nitrophenyl butyrate (PNPB) and p-nitrophenyl palmitate (PNPP). Most strains were active on both esters and 12% hydrolyzed olive oil. A strain identified as Natronococcus sp. was further studied. It grew optimally at 3.5 M NaCl, pH 8 and 40 degrees C. An increase in temperature shifted the optimum salt concentration range for growth from a wider range of 2-4 M, obtained at 25-30 degrees C, to a narrower range of 3.5-4 M, obtained at 35-40 degrees C. At 45 degrees C the optimum salt concentration was 2 M. These results show a clear correlation between salt and temperature requirement. The optimum conditions for the production of hydrolytic activity during growth were: 3.5 M NaCl and pH 8 for PNPB hydrolytic activity and 4 M NaCl and pH 7.5 for PNPP hydrolytic activity; both at 40 degrees C. The clear supernatant of cells grown at 4 M NaCl showed olive oil hydrolysis activity (in presence of 4 M NaCl) demonstrating the occurrence of a lipase activity in this strain. To our knowledge, this is the first report of a lipase activity at such high salt concentration.  相似文献

Extracellular proteolytic activity was detected in the haloalkaliphilic archaeon Natronococcus occultus as the culture reached the stationary growth phase. Proteolytic activity was precipitated with ethanol and subjected to a preliminary characterization. Optimal conditions for activity were attained at 60° C and 1–2 M NaCl or KCl. Gelatin zymography in the presence of 4 M betaine revealed a complex pattern of active species with apparent molecular masses ranging from 50 to 120 kDa. Experiments performed with inhibitors of the various groups of proteases indicated that the extracellular proteolytic enzymes of N. occultus are of the serine type. Abstract Archaebacterial halophiles dominate naturally occurring brines as the concentration of salts approaches saturation. Although only 4 genera of these bacteria have been recognised ( Halobacterium, Halococcus, Natronobacterium, Natronococcus ), chemotaxonomic studies indicate that there are 4 or 5 additional distinct groups. Archaebacterial halophiles are extremely physiologically versatile, some examples being capable of an anaerobic fermentative mode of growth, others of growth linked to the reduction of sulphur compounds. Many strains contain retinal-based pigments producing light-mediated movements of ions across the cell membrane, that in some cases can be harnessed for energy generation. However, it is likely that under normal conditions, an aerobic, chemoorganotrophic mode of nutrition is adopted.  相似文献
