Most Boletaceae form ectomycorrhizas but species in two sister groups (subfamily Chalciporoideae and the genus Pseudoboletus) have unclear trophic modes and are putatively mycoparasitic. Chalciporoideae is the earliest diverging lineage in Boletaceae and includes Chalciporus and Buchwaldoboletus. Buchwaldoboletus species fruit on wood and were thought to be saprobes, but the type species Buchwaldoboletus lignicola is mycoparasitic on the wood decay fungus Phaeolus schweinitzii. However, the nutritional mode and host range of B. lignicola and other Buchwaldoboletus remain unclear. We obtained fresh collections and axenic cultures of B. lignicola, B. hemichrysus and B. sphaerocephalus from the southeastern USA. We use cultures to examine the phylogenetic relationships among taxa in Chalciporoideae, elucidate the trophic mode of Buchwaldoboletus species and test the effect that Buchwaldoboletus species have on wood decay. Our analysis supports the hypothesis that the Chalciporus lineage is sister to all other Boletaceae but suggests that Buchwaldoboletus is not monophyletic. Confrontation experiments on agar suggest that Buchwaldoboletus species might parasitize a wide range of decay fungi. Buchwaldoboletus hemichrysus and B. sphaerocephalus produce abundant conidia in culture whereas the more distantly related B. lignicola produces sclerotia. Wood decay assays show that Buchwaldoboletus species did not have a significant effect on the decay caused by wood decay fungi. Interestingly, when B. lignicola or B. hemichrysus were inoculated alone on wood both species decayed wood, producing a brown rot that can cause 35–65% mass loss over 6 months. Our results suggest that Buchwaldoboletus species have a dual trophic mode as saprobes and mycoparasites.  相似文献

Attini are the only ants that use fresh plant material to cultivate species of Leucoagaricus, which are their source of nutrition. Escovopsis species are specialized mycoparasites of Leucoagaricus sp. and Escovopsis parasitism has a negative impact on the health of the ants' colonies. The goals of this work were: to test if the virulence of different isolates of Escovopsis weberi were the same across Leucoagaricus sp. and to analyze if structural mechanisms were related to variation in the virulence of E. weberi isolates. All E. weberi isolates were able to parasitize isolates of Leucoagaricus spp. but with striking differences in virulence, and it was shown that the contact between hyphae of both fungi was the main process that generates the degradation of Leucoagaricus isolates. Additionally, the two most virulent isolates produced hook-like protuberances, increasing the damage caused to its target. Finally, E. weberi was re-classified as a destructive biotrophic parasite.  相似文献

The functional genomics project “TrichoEST” was developed focused on different taxonomic groups of Trichoderma with biocontrol potential. Four cDNA libraries were constructed, using similar growth conditions, from four different Trichoderma strains: Trichoderma longibrachiatum T52, Trichoderma asperellum T53, Trichoderma virens T59, and Trichoderma sp. T78. In this study, we present the analysis of the 8,160 expressed sequence tags (ESTs) generated. Each EST library was independently assembled and 1,000–1,300 unique sequences were identified in each strain. First, we queried our collection of ESTs against the NCBI nonredundant database using the BLASTX algorithm. Moreover, using the Gene Ontology hierarchy, we performed the annotation of 40.9% of the unique sequences. Later, based on the EST abundance, we examined the highly expressed genes in the four strains. A hydrophobin was found as the gene expressed at the highest level in two of the strains, but we also found that other unique sequences similar to the HEX1, QID3, and NMT1 proteins were highly represented in at least two of the Trichoderma strains. Witches’ broom is the most devastating disease of cacao in Brazil, and losses to it entail serious socio-economical and environmental problems. Biological control of the causal agent Moniliophthora perniciosa (Mp) using the antagonistic fungus Trichoderma stromaticum (Ts) is promising, although the identification of superior isolates is necessary. Here, we report a study on the selection of more effective Ts isolates based on field experiments. Sixty-three Ts isolates from a local collection were applied on brooms and placed under typical conditions of shaded-cacao plantations in southeastern Bahia State (Brazil), during two periods of three months each. The percentages of Ts sporulation and incidence and severity of Mp were the parameters used for biocontrol assessments. The results from both experiments were very distinct, indicating a high phenotypic variation in this collection and suggesting a significant effect of the environment in the TsMp interaction. Ts-sporulation rates were negatively correlated with the presence of Mp in the brooms and a number of isolates reduced Mp incidence more efficiently than the reference isolate. Contrasting isolates in their efficiency of reducing Mp incidence were selected and further tested in four subsequent field trials for validation purposes. The results partially confirmed their biocontrol phenotypes but also suggested isolate-specific responses to environmental variations. Inhibition of Mp-basidiospore germination by total protein secreted in culture supernatants of Ts isolates correlated well with field results and revealed a potentially useful procedure for pre-screening of large collections towards selection of better biological control isolates. The characteristics and efficiency of the method as a reliable protocol for identification of superior BCAs in the witches’ broom—cacao pathosystem is discussed.  相似文献

Fungal chitinases are hydrolytic enzymes responsible for degradation of chitin. Chitinases are involved in several aspects of fungal biology, including cell wall remodelling during hyphal growth, conidial germination, autolysis, mycoparasitism and nutrient acquisition. They are divided into three distinct phylogenetic groups; A, B and C. Chitinases from the C group show structural similarities with the killer toxin zymocin produced by the yeast Kluyveromyces lactis and it is speculated that they have a similar function in filamentous ascomycetes, by facilitating penetration of toxins into cells of competing individuals. Genome analyses show that certain fungal species with a mycoparasitic lifestyle contain high numbers of killer toxin-like chitinases, compared with specialized saprotrophs and plant pathogens. Recent developments within this research field have revealed considerable variation in the modular structure and regulation of killer toxin-like chitinases, suggesting more diverse roles than merely fungal-fungal interactions. In this review, we summarize the current knowledge about this intriguing class of chitinases, including their modular structure, evolution, gene regulation, and functional analyses in mycoparasitic as well as in saprotrophic species. Summary Experiments on nutrient and staled agar were carried out to investigate the mycoparasitic activity of some fusaria againstRhizoctonia solani Kühn. Penetration and coiling byFusarium oxysporum Sch.,F. semitectum Berk & Rav. andF. udum Butler in and around theR. solani hyphae was observed. Lysis ofF. udum hyphae was observed inside theR. solani hyphae showing the reverse of the normal direction of necrotrophic mycoparasitic relationships. The involvement of the G-alpha protein GNA3 in the production of cell wall-degrading enzymes (CWDEs) by Trichoderma reesei during antagonism against Pythium ultimum was investigated. cAMP content was 2.8-fold higher in the T. reesei mutant gna3QL than in the parental TU-6. The gna3QL, like TU-6, inhibited the growth of P. ultimum in dual culture assays. Scanning electron microscopy showed that the gna3QL promoted more morphological alterations of P. ultimum cell wall than TU-6. In general, gna3QL produced higher activities of CWDEs than TU-6. We therefore suggest that CWDEs production during mycoparasitism by T. reesei against P. ultimum may be associated with the level of GNA3 activity.  相似文献

Restriction enzyme mediated integration (REMI) and Agrobacterium-mediated transformation (ATMT) were used to transform protoplasts or germinated conidia of the mycoparasite Coniothyrium minitans to hygromycin resistance. Using REMI, up to 32 transformants mug DNA(-1) were obtained, while 37.8 transformants 5 x 10(5) germlings(-1) were obtained using ATMT. Single-copy integrations occurred in 8% and 40% of REMI and ATMT transformants, respectively. A novel microtitre plate-based test was developed to expedite screening of 4000 REMI and ATMT C. minitans transformants. Nine pathogenicity mutants that displayed reduced or no pathogenicity on sclerotia of Sclerotinia sclerotiorum were identified.  相似文献

Antagonistic Trichoderma spp. are used throughout the world for the biological control of soil-borne plant diseases. This approach has stimulated an on-going search for more efficient mycoparasitic strains with a high potential for producing extracellular lytic enzymes. This study compares the production of lytic enzymes by native strains of Trichoderma asperellum and Trichoderma longibrachiatum on substrates of differing complexity. The quantity of protein induced by Agaricus bisporus-based medium was higher than that induced by Phymatotrichopsis omnivora-based medium. In P. omnivora medium, T. asperellum exhibited higher chitinolytic and β-1,3-glucanolytic activities than T. longibrachiatum. The enzyme profile was related to the previously reported ability of these strains to inhibit the growth of several soil-borne plant pathogens. NAGase production was similar among the tested indigenous strains of T. longibrachiatum; T479 and T359 produced more endochitinase, T479 produced more glucanase, and T341 and T359 produced more β-1,3-glucanase. The detected variations in glucanase and β-1,3-glucanase activities suggest that the production of these enzymes is strongly influenced by the substrate. Strains T397 and T359 exhibited xylanase activity, which triggers defence mechanisms in plants. Thus, these strains may utilise an additional mechanism of biocontrol.  相似文献

The production of proteolytic enzymes by several strains of ectomycorrhizal fungi i.e., Amanita muscaria (16-3), Laccaria laccata (9-12), L. laccata (9-1), Suillus bovinus (15-4), Suillus bovinus (15-3), Suillus luteus (14-7) on mycelia of Trichoderma harzianum, Trichoderma virens and Mucor hiemalis and sodium caseinate, yeast extract was evaluated. The strains of A. muscaria (16-3) and L. laccata (9-12) were characterized by the highest activity of the acidic and neutral proteases. Taking the mycelia of saprotrophic fungi into consideration, the mycelium of M. hiemalis was the best inductor for proteolytic activity. The examined ectomycorrhizal fungi exhibited higher activity of acidic proteases than neutral ones on the mycelia of saprotrophic fungi, which may imply the participation of acidic proteases in nutrition.  相似文献
