Metabolism is recognized as an important driver of cancer progression and other complex diseases, but global metabolite profiling remains a challenge. Protein expression profiling is often a poor proxy since existing pathway enrichment models provide an incomplete mapping between the proteome and metabolism. To overcome these gaps, we introduce multiomic metabolic enrichment network analysis (MOMENTA), an integrative multiomic data analysis framework for more accurately deducing metabolic pathway changes from proteomics data alone in a gene set analysis context by leveraging protein interaction networks to extend annotated metabolic models. We apply MOMENTA to proteomic data from diverse cancer cell lines and human tumors to demonstrate its utility at revealing variation in metabolic pathway activity across cancer types, which we verify using independent metabolomics measurements. The novel metabolic networks we uncover in breast cancer and other tumors are linked to clinical outcomes, underscoring the pathophysiological relevance of the findings.  相似文献

Questions: Is change in cover of dominant species driving the velocity of succession or is it species turnover (1)? Is the length of the time‐step chosen in sampling affecting our recognition of the long‐term rate of change (2)1 Location: 74 permanent plots located in the Swiss National Park, SE Switzerland, ca. 1900 m a.s.l. Methods: We superimpose several time‐series from permanent plots to one single series solely based on compositional dissimilarity. As shown earlier (Wildi & Schütz 2000) this results in a synthetic series covering about 400 to 650 yr length. Continuous power transformation of cover‐percentage scores is used to test if the dominance or the presence‐absence of species is governing secondary succession from pasture to forest. The effect of time step length is tested by sub‐samples of the time series. Results: Altering the weight of presence‐absence versus dominance of species affects the emerging time frame, while altering time step length is uncritical. Where species turnover is fast, different performance scales yield similar results. When cover change in dominant species prevails, the solutions vary considerably. Ordinations reveal that the synthetic time series seek for shortest paths of the temporal pattern whereas in the real system longer lasting alternatives exist. Conclusions: Superimposing time series differs from the classical space‐for‐time substitution approach. It is a computation‐based method to investigate temporal patterns of hundreds of years fitting between direct monitoring (usually limited to decades) and the analysis of proxy‐data (for time spans of thousands of years and more).  相似文献

The red-striped soft scale insect Pulvinaria tenuivalvata (Newstead) (Hemiptera: Coccidae) started to infest sugarcane plants (Saccharum officinarum L.) in different districts in Egypt during the last decade. The percentage of infestation was recorded in El-Wakf area, Qena Governorate (Naghhamadi mill zone) Upper Egypt in some fields. There are three levels of infestation, low, intermediate and high. From these fields, samples were selected for physical and chemical studies. The results obtained show that the stalks of infested plants decreased in weight, the sugar content (glucose and sucrose) drastically reduced and as the percentage of infestation increased the percentage of glucose and sucrose content significantly decreased. The primary and secondary humidity and the cellulose content also increased in the healthy plants compared to the infested ones. All the physical character of the infested plants was significantly affected in comparison with the healthy ones.  相似文献

Summary Nitrogen (N2) fixed by Rhizobium bacteroids in the legume nodule is excreted as ammonia to the surrounding host cell where it is efficiently assimilated into the amide group of glutamine. Generally glutamine is a minor exported solute of nitrogen, being further metabolised to asparagine in temperate species and to the ureides, allantoin and allantoic acid in tropical species. These solutes serve as the principal translocated forms of nitrogen in xylem. Compartmentalisation of the pathways of nitrogen metabolism and the role of ammonia in regulation of their activity is examined in nodules of both asparagine-forming (Lupinus albus L.) and ureide-forming (Vigna unguiculata L. 水洞沟第四纪地层可与萨拉乌苏河组上段对比。水洞沟遗址包含有新石器和旧石器晚期文化,其旧石器晚期文化发生在距今20,000多年至10,000多年间的黄土状粉砂土中,即发生在末次冰期主冰期的干冷环境下,比发育在距今约35,000—50,000年间的萨拉乌苏河旧石器文化为晚。  相似文献

The human platelet integrin GPIIb/IIIa (228 kDa), a Ca-dependent heterodimer formed by the IIb subunit (GPIIb, 136 kDa) and the 3 subunit (GPIIIa, 92 kDa), serves as the fibrinogen receptor at the surface of activated platelets. The degree of dissociation of the GPIIb/IIIa heterodimer (s°20 *, 8.9 S) into its constituent glycoproteins (GPIIb, 5.8 S; and GPIIIa, 3.9 S) has been assessed by analytical ultracentrifugation in Triton X100 buffers, and its Ca2+- and temperature-dependence correlated with Ca2+-binding to GPIIb/IIIa and its temperature dependence. At 21°C half-maximal dissociation of GPIIb/IIIa occurs at 5.5 ± 2.5 × 10–8 M Ca2+, very close to the dissociation constant of the high affinity Ca-binding site of GPIIb/IIIa (Kd1 8 ± 3 × 10–8 M) (Rivas and González-Rodríguez, 1991) and much lower than the Kd of the 3.4 medium affinity Ca-binding sites (Kd2 4 ± 1.5 × 10–5 M), which seems to demonstrate that the stability of the heterodimer in solution at room temperature is regulated by the degree of saturation of the high-affinity Ca-binding site. At 4°C, the stability of the heterodimer is apparently Ca2+-independent, while at room and physiological temperatures (15–37°C) the degree of dissociation of the heterodimer is regulated by the degree of dissociation of the high- and medium-affinity Ca-binding sites, respectively. On increasing the Ca2+ concentration up to 1 × 10–4 M after dissociation in Triton X100 solutions, the reconstitution of the GPIIb/IIIa heterodimer depends on the time and temperature at which the dissociated heterodimer was maintained, being almost complete within the first 5–10 min at 37°C and within the first 1–2 h at 21°C. After this time, a time- and temperature-dependent irreversible autoassociation of GPIIb (covalent) and GPIIIa (non-covalent) occurs, which hinders both the isolation of permanently stable monoamers of GPIIb and GPIIIa and the reconstitution of the GPIIb/IIIa heterodimer in Triton X100 solutions. Analysis of nine characteristics on 1,205 males and 932 females from 12 tribes or groups of tribes indicated a poor relationship between morphology and language, as well as moderate agreement with the variability expected considering geography only. Two samples in the Xingu area studied during an interval of half a century (1897-1947) showed remarkable similarity. The conformity of the Caingang morphology with those of other tribes and the distinctiveness of the Xavante and Tenetehara has been amply confirmed.  相似文献

Résumé En vue d'une étude, au laboratoire, de la nodulation et de la symbiose plante-bactérie des préparations de protoplastes de mésophylle et de nodule racinaire ont été effectuées chez le haricot (Phaseolus vulgaris variété Constant). Pour chaque préparation, les conditions opératoires et les concentrations optimales de cellulase et de pectinase en solution, dans du mannitol 0,6M ont été définies.Áprès 15 jours, 10 à 15% des protoplastes de mésophylle mis en culture dans le milieu B5 de Gamborg se divisent 2 à 3 fois. Des essais de fusion réalisés avec des faibles densités cellulaires entre protoplastes de mésophylle et protoplastes de nodule ont, donné des taux de fusion de l'ordre de 2,43%.
Study of the symbiosis Rhizobium-legume: Preparation of isolated cells forin vitro study
Summary With a view to studyin vitro the processes of nodulation and plant-bacteria symbiosis, preparation of mesophyll protoplasts and root nodule protoplasts have been realized fromPhaseolus vulgaris (Constant var.).For every preparation the process of experimentation and optimum concentrations of cellulase and pectinase diluted in mannitol 0,6M have been defined.After a 15 days' period, 10 to 15% of mesophyll protoplasts divided twice or even three times in B5 Gamborg's medium. Although we used low rate cell concentrations, root nodule protoplasts and mesophyll protoplasts have been able to fuse in the proportion of 2,43%.
