Abstract A current explanation of the mechanism of flooding injury to roots suggests that oxygen deficiency depresses the supply of respirable carbohydrates sufficiently to inhibit fermentation. However, even though it has been shown that phloem transport of assimilate is sharply reduced to anaerobic roots, inhibition of assimilate metabolism has also been suggested to be an important factor. This study examines these hypotheses by relating assimilate supply and metabolic activity in anoxic roots of alfalfa (Medicago sativa L.), a flood-intolerant species, and birdsfoot trefoil (Lotus corniculatus L.), a flood-tolerant plant. Roots were made anoxic (severe O2 deficiency) for 2, 4 or 6 d and shoots were labelled with 14CO2. Assimilate transport to the roots and metabolism to structural components were significantly decreased in both species in response to anoxia. Trefoil exhibited significantly greater 14C incorporation into the residue fraction at 4 d anoxia than did alfalfa, and this was consistent with the greater flooding tolerance of trefoil. When assimilate supply to O2-deficient roots was decreased by shoot shading, shoot fresh weight was reduced by both anoxia and light treatments. Root-soluble sugars were significantly decreased by shading but were greatly increased in response to anoxia. Root starch concentration also increased under anoxia. Root K+ concentration was reduced by anoxia only. The energy status (ATP/ADP) of roots was significantly decreased by shading; however, anoxia reduced the energy status only in unshaded plants. The data indicate that carbohydrate supply to anaerobic roots does not appear to be a limiting factor in the metabolic response of alfalfa roots. Alternatively, metabolism of assimilate in anoxic roots may be an important determinant of survival.  相似文献

Summary Earlier students of the origin of Lotus corniculatus suggested that this tetraploid species arose as an autotetraploid of the closely related diploid species L. tenuis or L. alpinus. More recent studies suggested that L. alpinus and L. japonicus could be ancestral forms. The present study of tannin content, phenolic content, cyanide production, morphology, cytogenetics, Rhizobium specificity and self-incompatibility in the corniculatus group virtually excludes the possibility that L. corniculatus could have arisen through autopolyploidy of L. tenuis or L. alpinus, and suggests that L. corniculatus arose through hybridization of L. alpinus and/or L. tenuis (probably as female parent) with L. uliginosus (probably as male parent), followed by chromosome doubling in the hybrid.  相似文献

Grasses grown in mixture with nodulated legumes often are N-limited, but N fertilization may result in reductions of N2 fixation and legume stands. We studied N-fertilizer effects on N2 fixation for three binary legume-grass mixtures in Uruguay. Replicated swards of white clover (Trifolium repens L.) (WC), red clover (Trifolium pratense L.) (RC), or birdsfoot trefoil (Lotus corniculatus L.) (BT), each in combination with tall fescue (Festuca arundinacea Schreb) (TF) at two legume proportions were sown in 1983 (Exp. 1) and 1984 (Exp. 2). In the fall of 1984, N treatments at 100 kg ha−1 and controls were randomly assigned to subplots in Exp. 1 (established swards) and in Exp. 2 (at seeding). The soil for both experiments was a fine, montmorillonitic, mesic, Typic Argiudolls. Herbage fixed-N was estimated by 15N isotope-dilution with pure stands of TF as reference. In both experiments, N fertilization reduced the proportion of legume N derived from air (% Ndfa) and increased herbage yield only during the first 18 to 20 weeks after application. Fertilizer-N reduced annual fixed-N yield from 178 to 148 kg ha−1 in Exp. 1 and from 65 to 29 kg ha−1 in Exp. 2 Fixed-N yield for BT was markedly reduced by N in both experiments (33 to 53%), whereas for the clovers reduction was lesser in Exp. 1 (9 to 13%) than in Exp. 2 (46 to 64%). Negative effects of N on % Ndfa were more evident for the high legume proportion. We conclude that fertilization with 100 kg N ha−1 reduced % Ndfa only for the immediate 18 to 20 weeks after application. Fertilizer-induced reduction of fixed-N yields lasted longer because of a more prolonged depression of legume proportion, especially for BT and for newly seeded swards. Journal Paper no. Summary A study of seed position in the pod ofLotus corniculatus L. cv. Mirabel (Fabaceae) suggested that reduced seed set after self-pollination is not due to an inability of the self-pollen tube to reach the end of the ovary. As in other cultivars, it has been demonstrated that cultivar Mirabel produced less seed per pod and shorter pods after self- than after cross-pollination. No differences were noted for percent germination of seeds produced by both types of pollination indicating that the number of seeds per pod is a reliable index of the ultimate productive potential of the pollination.  相似文献

Summary Gene flow was investigated in a natural population of Lotus corniculatus L. (Fabaceae) using a combination of pollen and seed dispersal studies and a recombinant DNA technique. The population is spatially heterogeneous and grows with Empetrum nigrum. L. corniculatus is pollinated by the pollen-collecting bumblebee Bombus lapidarius L. Most pollinator flights occurred within patches, as bees usually visit nearest-neighbour plants, show no marked directionality, and forage mostly within patches. Gene flow by seeds is also limited, reinforcing the pattern of gene flow within patches. However, 2.6% of pollinator flights are between patches and considerable pollen carryover also occurs. Thus, gene flow between patches is potentially sufficient to retard or prevent genetic differentiation in spite of the patchy sub-structuring of the population. A sub-set of the population was analysed for restriction fragment length polymorphisms (RFLPs) to document the actual gene flow pattern of the population. The DNA analysis revealed significant levels of genetic differentiation between the patches. The level of gene flow that can be inferred from the distribution of genetic variation is surprisingly restricted, as compared to gene flow inferred from pollinator behaviour, and emphasizes that stochastic processes like genetic drift and founder effects may have a strong impact on the prevailing genetic structure.  相似文献

Preplant soil applications of granular phenamiphos effectively reduced Pratytenchus penetrans in soil during the seeding year and 1 year after, and in the roots of birdsfoot trefoil 2 years after seeding. Forage yields were increased in the season following application of phenamiphos, but stands of plants/m² were not greater (P = 0.05) than those in the checks 1 and 2 years after treatment. Additional spring applications of phenamiphos 1 and 2 years after seeding further reduced numbers of nematodes in the soil but did not improve forage yields or plant stand over that of a single application. Broadcast preplant soil sprays of oxamyl followed by several foliar sprays at different rates and frequencies of application over a 3-year period restricted populations of P. penetrans in the soil and roots of birdsfoot trefoil but did not consistently result in increased forage yields. Stands of birdsfoot trefoil continued to decline each year even with oxamyl treatments.  相似文献

An efficient system for Agrobacterium tumefaciens-mediated transformation of Solanum gilo was established. The marker genes for kanamycin resistance and ß-glucuronidase expression were introduced. A comparison between cotyledon and hypocotyl explants showed that while regeneration was better from hypocotyl explants, cotyledon explants gave better transformation efficiency (46% vs. 32%). Four levels of kanamycin selection (100, 150, 200 and 250 mg/l) were tested for effect on transformation efficiency with each type of explant. Lower levels of kanamycin worked better using cotyledon explants, while higher levels of kanamycin worked better for hypocotyl explants. All nine t0 plants tested for expression of the kan r gene were positive. Pseudomonas 0–3 strain which was isolated from soil can grow on polyvinyl alcohol (PVA) as a sole carbon source. When 0.5 per cent of PVA (500, 1500 or 2000) was employed as the carbon source in the culture medium, PVA was almost completely lost from the culture fluid after a week and the concentration of total organic carbon measured by a TOC analyzer decreased from the initial value of about 2700 ppm to 250~300 ppm after 7~10 days culture. This bacterium was found to produce and secrete an inducible enzyme which degrade PVA. The way by which this enzyme degrades PVA was examined and the results were obtained which suggested that PVA was broken down oxidatively in a way of endowise splitting. However, the mechanism of PVA degradation has not been clarified yet. The optimum pH and temperature for enzyme activity were examined and they were 7.5~8.5 and 35~45°C, respectively. Morphological and biological characteristics of this bacterium were examined and it was similar to a strain of Pseudomonas boreopolis.  相似文献
