Summary Responses of lentil in unsterile soils at low, medium and high levels of plant available soil P toGlomus fasciculatum inoculation were evaluated. It was observed that growth, dry matter accumulation, nodulation, and nitrogen fixation were considerably improved in VAM inoculated plants over uninoculated control at low and medium levels of plant available soil P.  相似文献

A model to predict flowering time in diverse lentil genotypes grown under widely different photothermal conditions was developed in controlled environments. The present study evaluated that model with a world germ plasm collection of 369 accessions using two field environments in Syria and two in Pakistan. Photoperiod alone accounted for 69% of the variance in 1/f, the reciprocal of time (d) from sowing to flower. In contrast, temperature alone did not account for a significant proportion of variation in flowering time due to the exposure of plants to supra-optimal temperatures in the late-sown Syrian trial. With the model mean pre-flowering values of photoperiod and temperature combined additively to account for 90.3% of the variance of 1/f over accessions. The correlation of field-derived estimates of temperature sensitivity of accessions to glass-house-derived estimates was significant at P = 0.05, but the equivalent correlation for estimates of photoperiodic sensitivity was higher at P < 0.01. Flowering in the field was better measured as time from sowing to 50% plants in flower rather than time to first bloom or its node number. Dissemination of the lentil crop following domestication in West Asia to the lower latitudes such as Ethiopia and India has depended on selection for intrinsic earliness and reduced sensitivity to photoperiod. Movement from West Asia to the higher latitudes accompanied by spring sowing has resulted in a modest reduction in photoperiod sensitivity and an increase in temperature sensitivity.  相似文献

The vesicular-arbuscular mycorrhizal fungi (VAMF) Glomus clarum (Nicol. and Schenck) isolate NT4, G. mosseae (Nicol. and Gerd.) Gerd. and Trappe isolate NT6 and G. versiforme (Karst.) Berch isolate NT7 coexist in wheat field soils in Saskatchewan. This study assessed the response of lentil (Lens esculenta L.) and wheat (Triticum aestivum L.) to monospecific and mixed cultures of these VAMF isolates. Seedlings were inoculated with 100 spores of a VAMF isolate, or an equal mixture of spores of two isolates, and grown in a sterile soil mix in a growth chamber. Both crops responded differently to these different VAMF isolates. In the case of lentil, G. clarum NT4 was more effective than G. mosseae NT6 and G. versiforme NT7, and significantly increased (P<0.05) the shoot dry weight (43%) and grain yield (57%) compared with the uninoculated control. There was a significant positive correlation between the percentage of VAMF colonized roots and shoot dry weight (r=0.672***) and shoot phosphorus concentration (r=0.608***) of lentil. In the case of wheat, G. clarum NT4 had no effect on shoot dry weight, but produced significant (P<0.08) increases in grain yield (12%) and the phosphorus concentration of the shoot and grain. Although G. clarum NT4 and G. mosseae NT6 both produced similar levels of VAM colonization in wheat, the only response of wheat to isolate NT6 was an increase in plant height at harvest. The efficacy of G. clarum NT4 on both crops appeared to be related to its ability to produce more arbuscular colonization than G. mosseae NT6. Dual inoculation of seedlings with G. clarum NT4 and G. mosseae NT6 resulted in competition between these two isolates. This was evident from a comparison of plant shoot dry weight and grain yield, and VAMF spore production on the two crops inoculated either with isolate NT4 alone or in combination with NT6. G. mosseae NT6 reduced the efficacy of G. clarum NT4 by 16% when dual inoculated on lentil, but had no effect when the host was wheat. Based on spore production, it was found that G. clarum NT4 was more competitive than G. mosseae NT6 when dual inoculated on lentil or wheat. Isolate NT4 produced ca. 2000 and 500 spores/ 100 g substrate, respectively, in the lentil and wheat pots, which was approximately 2–3 times more spores than those produced by isolate NT6 with either crop. When the plants were dual inoculated, there was a 15–19% reduction in spore production by G. clarum NT4 and a 50–70% decrease in spore production by G. mosseae NT6. Our results show that G. clarum NT4 was more competitive and effective in its ability to colonize and increase the growth and yield of lentil and wheat than G. mosseae NT6 or G. versiforme NT7. The relative performance of isolate NT4 with different host plants suggests that this VAMF isolate exhibits a host preference for lentil.  相似文献

Randomly amplified polymorphic DNA (RAPD) markers were used to estimate intra- and interspecific variations in the genus Lens (lentil). Twenty cultivars of L. culinaris ssp. culinaris, including 11 microsperma (small-seeded) and nine macrosperma (large-seeded) types, and 16 wild relatives (four accessions each of L. culinaris ssp. orientalis, L. odemensis, L. nigricans and L. ervoides), were evaluated for genetic variability using a set of 40 random 10-mer primers. Fifty reproducibly scorable DNA bands were observed from ten of the primers, 90% of which were polymorphic. Genetic distances between each of the accessions were calculated from simple matching coefficients. A dendrogram showing genetic relationships between them was constructed by an unweighted pair-group method with arithmetical averages (UPGMA). This study revealed that (1) expect for L. ervoides, the level of intraspecific variation in cultivated lentil is lower than that in wild species, (2) L. culinaris ssp. orientalis is the most likely candidate for a progenitor of the cultivated species, and (3) microsperma and macrosperma cultivars were indistinguishable by the RAPD markers identified here.  相似文献

A restriction-site analysis of chloroplast DNA (cpDNA) variation in Lens was conducted to: (1) assess the levels of variation in Lens culinaris ssp. culinaris (the domesticated lentil), (2) identify the wild progenitor of the domesticated lentil, and (3) construct a cpDNA phylogeny of the genus. We analyzed 399 restriction sites in 114 cultivated accessions and 11 wild accessions. All but three accessions of the cultivar had identical cpDNAs. Two accessions exhibited a single shared restriction-site loss, and a small insertion was observed in the cpDNA of a third accession. We detected 19 restriction-site mutations and two length mutations among accessions of the wild taxa. Three of the four accessions of L. culinaris ssp. orientalis were identical to the cultivars at every restriction site, clearly identifying ssp. orientalis as the progenitor of the cultivated lentil. Because of its limited cpDNA diversity, we conclude that either the cultivated lentil has passed through a genetic bottleneck during domestication and lost most of its cytoplasmic variability or else was domesticated from an ancestor that was naturally depauperate in cpDNA restriction-site variation. However, because we had access to only a small number of populations of the wild taxa, the levels of variation present in ssp. orientalis can only be estimated, and the extent of such a domestication bottleneck, if applicable, cannot be evaluated. The cpDNA-based phylogeny portrays Lens as quite distinct from its putative closest relative, Vicia montbretii. L. culinaris ssp. odemensis is the sister of L. nigricans; L. culinaris is therefore paraphyletic given the current taxonomic placement of ssp. odemensis. Lens nigricans ssp. nigricans is by far the most divergent taxon of the genus, exhibiting ten autapomorphic restriction-site mutations.  相似文献

The reaction with substrates and carbonyl reagents of native lentil Cu-amine oxidase and its modified forms, i.e. Cu-fully-depleted, Cu-half-reconstituted, Cu-fully-reconstituted, Co-substituted, Ni-substituted and Zn-substituted, has been studied. Upon removal of only one of the two Cu ions, the enzyme loses 50% of its enzymatic activity. Using several substrates, Co-substituted lentil amine oxidase is shown to be active but the k c value is different from that of native or Cu-fully-reconstituted enzyme, while K m is similar. On the other hand, the Ni- and Zn-substituted forms are catalytically inactive. Enzymatic activity measurements and optical spectroscopy show that only in the Co-substituted enzyme is the organic cofactor 6-hydroxydopa quinone reactive and the enzyme catalytically competent, although less efficient. The Co-substituted amine oxidase does not form the semiquinone radical as an intermediate of the catalytic reaction. While devoid or reduced of catalytic activity, all the enzyme preparations are still able to oxidise two moles of substrate and to release two moles of aldehyde per mole of dimeric enzyme. The results obtained show that although Co-substituted amine oxidase is catalytically competent, copper is essential for the catalytic mechanism. Genetic diversity and interrelationships among 31 lentil genotypes were evaluated using 10 Inter-Simple Sequence Repeat (ISSR) and 10 directed amplification of minisatellite DNA region (DAMD) primers. A total of 43 and 48 polymorphic bands were amplified by ISSR and DAMD markers, respectively. Average polymorphism information content (PIC) for ISSR and DAMD markers were 0.37 and 0.41, respectively. All 31 lentil genotypes could be distinguished by ISSR markers into three groups and by DAMD markers into two groups. Various molecular markers show a different efficiency for evaluating DNA polymorphism in lentil and indicate that the patterns of variation are clearly influenced by the genetic marker used. Comparatively, the genetic diversity of examined lentil genotypes by two different marker techniques (ISSR and DAMD) was high and indicated that ISSR and DAMD are effective and promising marker systems for fingerprinting in lentil and give useful information on its genetic relationships.  相似文献

Dwarfing genes were detected following intra- and interspecific hybridization in Lens. Dwarf phenotypes are controlled by two complementary dominant genes, Df 1 and Df 2. These two genes are suppressed by the dominant allele of the dwarf inhibitor genes, Dfi. The dominant allele of the Df gene was detected in L. ervoides from Ethiopia and Uganda and in a cultivated line of L. culinaris from Ethiopia, that of the Df 2 gene in a L. ervoides accession from Israel. The dominant allele of the Dfi gene was detected in segregating populations of hybrids between L. ervoides accessions from Israel and Uganda. Using the homozygous dwarf, dfidfi, Df 1 Df 1, Df 2 Df 2 as the parent in interspecific crosses, we detected the dominant allele of the Dfi gene in one accession of L. nigricans and another of L. lamottei. The appearance of dwarf plants in segregating populations of hybrids between the cultivated line from Ethiopia and L. ervoides from Israel indicate that the cultivated line possesses the dominant allele of the Dfi gene. Dwarf plants were characterized by short internodes, a short leaf axis and smaller, convex leaflets. Spraying the dwarf plants with gibberellic acid induced internode and lead-axis elongation but had no effect on leaflet shape and size. When the dwarfs and their parental lines were grown in the dark they had the same internode length. Received: 12 April 1997 / Accepted: 25 June 1997  相似文献

Abstract: Rapidly transported fucose-labelled glycoproteins from axons of rabbit retinal ganglion cells were solubilized with nonionic detergents. The labelled glycoproteins carried hydrophobic sites as revealed by a reversible binding to a hydrophobic matrix. A major portion of the labelled glycoproteins carried α-D-mannopyranoside (or glucopyranoside) residues, as shown by a specific interaction with the lectin from Lens culinaris. Three major groups of labelled polypeptides capable of binding to the lectin are described.  相似文献

Summary In a pot experiment with 26 calcareous soils, the critical limit of Fe in soils and plants was evaluated. DTPA-extractable Fe was found significanty correlated with Bray's per cent yield in rice. The Fe2+ (iron) in rice and lentil was also found significantly correlated with DTPA-extractable Fe as well as Bray's per cent yield showing thereby the superiority of Fe2+ (iron) in leaves over DTPA-extractable soil Fe to differentiate Fe responsive soils from non-responsive ones. The total Fe content in plant tissues does not seem correlated with the occurrence of Fe deficiency. The threshold values of DTPA-extractable soil Fe and Fe2+ (iron) in rice and lentil leaves were 6.95, 44 and 74.5 ppm, respectively below which appreciable responses to Fe application were observed. The optimum Fe level for these soils was found to be 10 ppm in which the dry matter yield response in all the 19 rice soils and 16 lentil soils ranged from 14.28 to 56.16 (Av. 25.75%) and 13.31 to 53.97 (Av. 22.47%), respectively.  相似文献
