The linear dichroism of single monolayers of lutein, zeaxanthin and a mixture of lutein and synthetic phosphatidylcholine has been measured. The angle of orientation of the carotenoid molecules was found to lie between 45° and 51° relative to the plane of the solid support. Although the adsorbed monolayers were mostly in a monomeric state, microscopic observations, as well as the II-A isotherms, indicated the existence of crystalline islets. The results have been interpreted in connection with Haidinger's polarization brushes.  相似文献

用原子力显微镜(AFM)研究了磷脂DMPC三层Langmuir-Blodgett(LB)膜的分子排列结构,结果表明:在磷脂LB膜的两相(液体压缩相Liquid-condensedphase和液体扩张相Liquid-expandedphase)共存时,液体压缩相中的磷脂分子排列紧密,取向一致,分子间作用力较大,因而能够得到分子图像。而液体扩张相中的磷脂分子排列松散,取向混乱。分子间的作用力较弱,难于得到分子图像。在液体压缩相中磷脂分子以单斜晶格结构排列,分子间隔为0.72nm.分子高度为2.1nm。这一结果和DMPC的单晶结构进行了比较。  相似文献

Cell growth and production of interferon-β (IFN-β) were investigated for normal human skin fibroblast cells (NB1-RGB) cultured on membranes prepared from peptide-lipids containing the arginine-glycine-aspartic acid [Arg-Gly-Asp] (RGD), tyrosine-isoleucine-glycine-serine-arginine [Tyr-Ile-Gly-Ser-Arg] (YIGSR) and arginine-glutamic acid-aspartic acid-valine [Arg-Glu-Asp-Val] (REDV) peptides. Cell density was found to be approximately the same on various peptide-lipid membranes, whereas production of IFN-β depended significantly on the peptide-lipid membranes on which NB1-RGB cells were cultured. The highest production of IFN-β was observed for NB1-RGB cells on REDV-lipid membranes prepared by a casting method (REDV-cast membranes) after 24 hr of cultivation. Specific binding between REDV of REDV-cast membranes and the receptor on the NB1-RGB cells may have caused the specific cell response for the production of IFN-β. The Langmuir-Blodgett technique and its variants (alternate layers, self-organising mixtures, the semi-amphiphilic technique, the peculiar solid state chemistry in L.B. films) are collective methods which allow physical chemists, with a very small amount of synthetic chemistry, to build up molecular assemblies exhibiting not only the properties of each of their components, but also extra properties which arise from the architecture: cooperativity, anomalous chemical properties, molecular recognition, etc. These new tailored molecular edifices are the basic “brick” of tomorrow's molecular electronics and fine chemistry. These strategies are exemplified here by two active supramolecular edifices which have been successfully designed and built up: an artificial dioxygen trap based on the same principle as hemoglobin, and one molecule thick conductors. Promising applied results have already been obtained in the field of gas sensing with these new conductors, owing to molecular architectural amplification.  相似文献

The antimicrobial activity of the anionic peptide, AP1 (GEQGALAQFGEWL), was investigated. AP1 was found to kill Staphylococcus aureus with an MLC of 3 mM and to induce maximal surface pressure changes of 3.8 mN m−1 over 1200 s in monolayers formed from lipid extract of S. aureus membranes. FTIR spectroscopy showed the peptide to be α-helical (100%) in the presence of vesicles formed from this lipid extract and to induce increases in their fluidity (Δν circa 0.5 cm−1). These combined data show that AP1 is able to function as an α-helical antimicrobial peptide against Gram-positive bacteria and suggest that the killing mechanism used by the peptide involves interactions with the membrane lipid headgroup region. Moreover, this killing mechanism differs strongly from that previously reported for AP1 against Gram-negative bacteria, indicating the importance of considering the effects of membrane lipid composition when investigating the structure/function relationships of antimicrobial peptides.  相似文献

A transition of an organic molecule from the ground into an excited state is often accompanied by the intra-molecular electronic (or ionic) charge transfer. A degree of this spatial charge transfer may be expressed in terms of the difference between electrical dipole moments of the excited and the ground states. The same is true for the inter-molecular transitions in condensed media, namely, crystals, amorphous films, liquid and solid solutions, etc. The degree of the charge transfer determines the magnitude of various physical effects such as the electro-optical (Stark) effect, optical second harmonic generation, photo-induced polarization, pyro-electric effect and others. The quadratic Stark-effect can be used for a quantitative determination of the charge transfer degree , the other effects can be understood in terms of the charge transfer. This paper is a brief review of various experimental data mostly obtained by the authors which allow the quantitative interpretation using the concept of the charge transfer.  相似文献

Amphotericin B (AmB) is considered the gold-standard in the treatment of serious systemic mycoses despite its numerous adverse effects. Both the mechanism of antifungal action and the toxicity of this drug are dependent on its molecular organization. The effect of AmB on the organization of lipid membranes formed with dipalmitoylphosphatidylcholine (DPPC) was studied with application of the Langmuir-Blodgett technique and ATR-FTIR spectroscopy. The aim of this research was to analyze the physical interactions leading to the formation of aggregated forms of AmB molecules in one-component monolayers and lipid multibilayers. Analysis of FTIR spectra of two-component multibilayers suggests the possibility the mutual reorientation of the amino-sugar moiety (mycosamine) and macrolide ring. This effect may be significant in the explanation of the aggregation processes of AmB in biological systems.  相似文献

The conductance of the BK channel was evaluated in reconstituted bilayers made of POPE/POPS (3.3:1), or POPE/POPS with an added 20% of either SPM (3.3:1:1), CER (3.3:1:1), or CHL (3.3:1:1). The presence of SPM, which is known to increase bilayer thickness, significantly reduced the conductance of the BK channel. To directly test the role of membrane thickness, the conductance of the BK channel was measured in bilayers formed from PCs with acyl chains of increasing length (C14:1-C24:1), all in the absence of SPM. Slope conductance was maximal at a chain length of (C18:1) and much reduced for both thinner (C14:1) and thicker (C24:1) bilayers, indicating that membrane thickness alone can modify slope conductance. Further, in a simplified binary mixture of DOPE/SPM that forms a confined, phase-separated bilayer, the measured conductance of BK channels shows a clear bimodal distribution. In contrast, the addition of CER, which has an acyl chain structure similar to SPM but without its bulky polar head group to POPE/POPS, was without effect, as was the addition of CHL. The surface structure of membranes made from these same lipid mixtures was examined with AFM. Incorporation of both SPM and CER resulted in the formation of microdomains in POPE/POPS monolayers, but only SPM promoted a substantial increase in the amount of the high phase observed for the corresponding bilayers. The addition of CHL to POPE/POPS eliminated the phase separation observed in the POPE/POPS bilayer. The decrease in channel conductance observed with the incorporation of SPM into POPE/POPS membranes was, therefore, attributed to larger SPM-rich domains that appear thicker than the neighboring bilayer.  相似文献

A qualitative and quantitative analysis of the conformation of Langmuir-Blodgett (LB) dried films of cytochrome C on silicon wafers was performed by Fourier transform ir (FTIR) spectroscopy. A deconvolution procedure was applied to the amide I band analysis, in order to determine the percentage of the different secondary structures. Qualitative analysis was performed by examining difference spectra. Films obtained by spreading protein solutions at pH 7.4 and 1, dried at 25 and 100°C, on silicon wafers were also examined in order to detect spectral components associated with denatured protein domains, and to compare them with cytochrome C LB films. FTIR spectroscopy showed that the following important changes characterise LB film spectra: (a) the α-helix component is higher (its percentage is 57 and 54%) than the one estimated in dried film obtained by spreading the solutions at pH 7.4 on a silicon substrate (43%), (b) there is an increase in the intensity of bands attributed to protonated carboxy group bands, involved and not involved in the formation of hydrogen bonds, and a decrease in those attributed to deprotonated carboxy groups, (c) the intensity of several bands attributed to aromatic amino acids and aliphatic chains increases, and (d) bands due to O(SINGLEBOND)H stretching vibrations of crystallization water are present. These conformational changes could be induced by protein-protein interaction caused by the close packing of molecules that occurs during LB film formation; it cannot be excluded that they may be accompanied by partial changes in the tertiary structure of the protein.
