Ultrastructural changes in isolated rat hepatocytes exposed to different CCl4 concentrations

Ultrastructural data are presented on time-course changes in isolated rat hepatocyte suspensions exposed either to 1.2 or 1.8 mM CCl4 for up to 1 h. The subcellular changes at the lower concentration, but not the higher, are shown to closely parallel those reported to occur in rat hepatocytes following ingestion of CCl4.

Charge balance in the alpha-hydroxyacid dehydrogenase vacuole: an acid test.

The proposal that the active site vacuole of NAD(+)-S-lactate dehydrogenase is unable to accommodate any imbalance in electrostatic charge was tested by genetically manipulating the cDNA coding for human muscle lactate dehydrogenase to make a protein with an aspartic acid introduced at position 140 instead of the wild-type asparagine. The Asn 140-Asp mutant enzyme has the same kcat as the wild type (Asn 140) at low pH (4.5), and at higher pH the Km for pyruvate increases 10-fold for each unit increase in pH up to pH 9. We conclude that the anion of Asp 140 is completely inactive and that it binds pyruvate with a Km that is over 1,000 times that of the Km of the neutral, protonated aspartic-140. Experimental results and molecular modeling studies indicate the pKa of the active site histidine-195 in the enzyme-NADH complex is raised to greater than 10 by the presence of the anion at position 140. Energy minimization and molecular dynamics studies over 36 ps suggest that the anion at position 140 promotes the opening of and the entry of mobile solvent beneath the polypeptide loop (98-110), which normally seals off the internal active site vacuole from external bulk solvent.     

Characterization of some East African Theileria species isolates by isoenzyme analysis, with particular reference to T. parva

Eight different isolates of Theileria parva and one isolate of T. taurotragi, in the form of intra-lymphocytic schizonts and/or purified piroplasms, were subjected to isoenzyme analysis for 24 enzymes by both isoelectric focusing in agarose and electrophoresis in starch gel. Twelve enzymes distinguished between T. parva and T. taurotragi; five enzymes (HK, GPI, PEP1, LDH and SOD) showed variations within T. parva. The metabolism of the host cell was affected by schizont infection, which masked intraspecific variations. Piroplasms were of more potential value for characterization of T. parva.     

Specific lysis of GABAergic synaptosomes by an antiserum to glutamate decarboxylase

An antiserum to pure glutamate decarboxylase (GAD) when incubated with rat cortical synaptosomes in the presence of complement caused release of 33-53% of lactate dehydrogenase (LDH) and 22-41% of total GAD. In addition most of the gamma-aminobutyrate (GABA) present was released. Anti-GAD antiserum alone, or complement alone, were without action. The antiserum plus complement had no effect on noradrenaline or choline uptake, and did not release choline acetylase (ChAT). Anti-ChAT serum plus complement released 30-37% of ChAT and 10-13% of LDH. It prevented choline uptake. This serum did not produce GAD release or prevent GABA, choline or noradrenaline uptake. When cortical synaptosomes were exposed to both antisera plus complement, their actions were strictly additive. The data indicate specific lysis of GABAergic and cholinergic synaptosomal sub-populations.     

Microsomal-catalyzed hydroperoxide-dependent C-oxidation of amines

Organic hydroperoxides are capable of supporting the C-oxidation of several different amines in the presence of hepatic microsomes. Evidence is presented that indicates that microsomal cytochrome P-450 acts as the catalyst. Removal of the NADPH-cytochrome $\text{c}$ oxidoreductase or essential phospholipid from microsomes does not significantly affect the peroxidase activity. Of the amine substrates C-oxidized by organic hydroperoxides in the presence of microsomes, only aminopyrine and dimethylaniline are rapidly oxidized by hydroperoxides in the presence of catalase. The catalase-mediated reaction can also be distinguished from the microsomal-catalyzed reaction by the use of differential inhibitors.     

聚焦色谱法测定肌型肌酸激酶亚型

报道了测定CK-MM亚型的聚焦色谱法,此法简单,快速,结果可靠,线性范围宽,最低检测限（8U／L）较正常参考值低,比国外报道的类似方法高6倍以上,分离度亦有改进.测定了20例健康人血清亚型分布,与文献报道结果相近.该法自动化程度高,已在急性心梗的诊断中实际应用.     

AST同工酶的琼脂糖凝胶电泳NBT显色法

分布于细胞内线粒体及细胞质中的天门冬氨酸氨基转移酶(AST·EC·2·6·1·1)是人血清中含有的两种同工酶,分别称为AST-m和AST-c同工酶.AST-c电泳迁移率介于血清α-球蛋白与β-球蛋白之间.AST-m电泳迁移率相似于γ-球蛋白,琼脂糖凝胶电泳固蓝B染色法对m-AST检出率较低.用NBT显色法则可得到较好效果.     

不同性别黄鳝六种组织中LDH同工酶电泳谱的初步研究

本文报告了运用聚丙烯酰胺凝胶圆盘电泳研究不同性别黄鳝的血清、心肌、骨骼肌、肝、肾和生殖腺等六种组织器官中ＬＤＨ同工酶。结果表明六种不同组织中ＬＤＨ同工酶谱各不相同,具有明显的组织特异性。在不同性别中某些同一种组织的ＬＤＨ同工酶谱也发生变化,这说明决定黄鳝ＬＤＨ同工酶表达的因素在不同性别中有差异。
