目的:研究信号通路ELK-1/JNK/c-Fos在左归降糖解郁方(ZGJTJYF)对模拟糖尿病并发抑郁症(DD)环境下抗海马神经元凋亡中的作用。方法:原代培养海马神经元,加入高糖(150 mmol/L)+皮质酮(200μmol/L),构建DD体外细胞模型;将培养的海马神经元细胞随机分为5组:空白血清组、正常组、左归降糖解郁方含药血清组、阳性药(二甲双胍+氟西汀)含药血清组和模型组(每组3个复孔)。模型组和正常组给予等量培养液,其余组加入相应体积分数10%的相应血清,均干预18 h。分别采用Hoechst染色、高内涵细胞成像分析技术和RT-PCR技术分别检测海马神经元凋亡情况及检测凋亡相关ELK-1、JNK和c-Fos蛋白和基因的表达。结果:与空白组比较,模型组细胞凋亡明显,其海马神经元凋亡数量明显增多,ELK-1、JNK和c-Fos的mRNA及蛋白表达水平均显著升高(P<0.05);与模型组比较,左归降糖解郁方含药血清组和阳性药含药血清组大鼠海马神经元可见局部亮点明显减少,凋亡细胞数显著减少,ELK-1、JNK和c-Fos蛋白及mRNA表达均明显下调(P<0.05),且神经网络及树突连接情况得到明显改善。结论:左归降糖解郁方可以减低DD环境下海马神经元中Elk-1、JNK、c-fos表达而起到抗凋亡的效果。  相似文献

摘要 目的:分析自拟益气通阳活血方治疗阿尔茨海默病的临床效果。方法:选取2020年10月~2022年4月于中国人民解放军空军军医大学第一附属医院接受治疗的阿尔茨海默病患者104例,依据入院先后顺序编号,按单双号分为对照组(n=52)和研究组(n=52)。对照组进行常规西药治疗,研究组在此基础上进行自拟益气通阳活血方治疗。比较两组临床疗效、治疗前后中医证候积分、日常生活活动能力、认知功能、血清去甲肾上腺素(NE)、多巴胺(DA)、乙酰胆碱转移酶(ChAT)水平、β淀粉样蛋白前体(APP)、β淀粉样蛋白1-42(Aβ1-42)、丙二醛(MDA)、超氧化物歧化酶(SOD)水平,并比较两组安全性。结果:治疗后,两组中医证候积分均降低,且研究组显著低于对照组(P<0.05);研究组总有效率为94.23%,明显高于对照组的78.85%(P<0.05);治疗后,两组简易智能精神状态检查量表(MMSE)评分与蒙特利尔认知评估量表(MoCA)评分研究组均高于对照组(P<0.05);治疗后,两组barthel指数评分及日常生活能力量表(ADL)评分研究组均显著高于对照组(P<0.05);治疗后,两组DA、NE、ChAT水平均升高,且研究组明显高于对照组(P<0.05);治疗后,两组SOD水平升高,APP、Aβ1-42、MDA水平均降低(P<0.05),且与对照组相比研究组各指标变化更显著(P<0.05);两组治疗期间的不良反应均较轻微,经对症处理后均缓解,不良反应发生率比较无明显差异(P>0.05)。结论:自拟益气通阳活血方治疗阿尔茨海默病疗效显著,可降低中医证候积分,提高认知功能,调节NE、DA、ChAT水平,抑制APP、Aβ1-42水平,提高其日常生活活动能力,且安全性高。  相似文献

前列腺Ⅰ号抗炎镇痛作用以及抗前列腺炎的实验研究   总被引:2,自引:0,他引:2  
采用热板法和扭体法观察前列腺Ⅰ号抗炎、镇痛以及抗大鼠实验性前列腺炎的作用。采用小鼠耳廓二甲苯制炎法观察抗炎作用,前列腺炎模型采用向大鼠前列腺组织内注入角叉菜胶的方法。结果显示前列腺Ⅰ号能减少小鼠扭体反应数,延长小鼠热板法引起的痛反应潜伏期,对小鼠耳廓肿胀有明显的抑制作用,前列腺炎模型试验中,前列腺液检查卵磷脂小体明显增加,白细胞数降低。因此前列腺Ⅰ号具有明显的抗炎、镇痛以及抗前列腺炎的作用。  相似文献

Differences in iron bioavailability from human milk and milk formulas may in part be due to differences in lipid composition. We investigated the short and long term effects of diets based on different fats [corn, coconut, olive, or soy oil, human milk fat (HMF) and a formula fat blend (FF)] on iron absorption in rats. Suckling rat pups dosed with 59Fe-labeled diets containing different fat sources were killed after 6 h, and blood and individual tissues were counted. Iron availability was estimated by % 59Fe in blood. Pups dosed with a more saturated fat (coconut oil) had a higher % 59Fe in blood than those fed other fat sources. Weanling rats were used to determine iron bioavailability from fat sources using both the hemoglobin repletion method and whole body counting. Hemoglobin regeneration was significantly higher for rats fed the HMF diet (8.4 +/- 0.5 g/dl) than from the FF diet (6.5+/-0.6 g/dl) or the corn oil diet (less saturated) (6.4 +/- 0.3 g/dl). Rats fed diets based on coconut oil (more saturated) had significantly higher % 59Fe retention (61.6 +/- 1.4) than rats fed diets based on FF (49.8 +/- 3.4). There was a significant positive association between oleic acid in the diet and oleic acid in the intestinal mucosa (r = 0.95, p < 0.05) and between linoleic acid in the diet and linoleic acid in the intestinal mucosa (r = 0.97, p < 0.05) suggesting that the dietary treatment altered the fatty acid composition of the brush border membrane. Our results suggest that saturated fats may increase iron absorption and that part of this may be achieved by changes in the fatty acid composition of the intestinal mucosa. Hemoglobin regeneration and % 59Fe retention data suggest that differences in iron absorption from infant diets may in part be due to differences in fat composition. Therefore, lipid composition of infant formulas should also be taken into consideration as a factor influencing iron bioavailability.  相似文献

促甲状腺素(TSH)通过甲状腺细胞膜上的TSH受体(TSHR),产生第二信使cAMP,从而激活cAMP反应性起动子,而使相应的基因获得表达.实验结果表明,在构建有TSHR和糖蛋白激素cAMP反应性起动子以及萤光素基因(Luc)的转染细胞中,经补肾益精中药固真方提取液(1×10~(-4)稀释)处理3~5d后,可下调TSHR基因的表达,并使TSHR数目减少.提示固真方可调整甲状腺细胞的功能,或许有利于调整甲状腺机能亢进.  相似文献

In vehicle dynamics studies, the tire behaviour plays an important role in planar motion of the vehicle. Therefore, a correct representation of tire is a necessity. This paper describes a mathematical model for wheelchair tire based on the Magic Formula model. This model is widely used to represent forces and moments between the tire and the ground; however some experimental parameters must be determined. The purpose of this work is to identify the tire parameters for the wheelchair tire model, implementing them in a dynamic model of the wheelchair. For this, we developed an experimental test rig to measure the tires parameters for the lateral dynamics of a wheelchair. This dynamic model was made using a multi-body software and the wheelchair behaviour was analysed and discussed according to the tire parameters. The result of this work is one step further towards the understanding of wheelchair dynamics.  相似文献

To investigate the pharmacological effects of Danggui Buxue Tang (DBT) on immune-mediated aplasia anemia mice. The model of immune-mediated aplasia anemia mice was induced by means of 60Co γ-ray irradiation and mixed cells of thymus and lymphnode of DBA/2 mice infusion through tail vein, the parameters tested indices were as following: blood picture, bone marrow nucleated cell count (BMNC), murine colony-forming unit-megakaryocytes (CFU-GM) of bone marrow cells, murine colony-forming unit-erthroid (CFU-E) and burst forming unit-erythroid (BFU-E). The results showed that DBT could not only withstand significantly decreation of blood cells by immune-mediated, but also stimulate on the growth of bone marrow colony cell and increase the weight of hemopoietic progenitor of bone marrow. Therefore, DBT had an obvious treat effect on immune-mediated aplasia anemia models mice.  相似文献

目的:观察脑卒中后大鼠海马的形态学演变趋势及颐脑解郁方对其的干预作用。方法:雄性Wistar大鼠随机分为正常组、假手术组、MCI组、中药组、西药组、PSD组和VD组。采用同种系微栓子体外注入法制备MCI模型;PSD组术后7 d予21 d慢性不可预知的温和应激加孤养复制PSD模型;VD组术后28 d经水迷宫筛选后进入实验。假手术组、中药组及西药组分别予蒸馏水、颐脑解郁方、尼莫地平干预给药4周。于第1、2、4周光镜下分析海马的形态学结构变化。结果:MCI组海马锥体细胞数量显著减少,细胞排列不整齐。PSD组及VD组锥体细胞数量显著减少,胞体不同程度缩小、核固缩、核碎裂、核溶解,细胞排列不整齐,且呈进行性发展。中药组锥体细胞缺失较少,细胞排列整齐,核仁明显,西药组细胞变性有所改善,部分细胞核固缩,不如中药组改善明显。结论:颐脑解郁方可能通过益肾调气的主治功效改善Papez回路的病理改变,阻断病情的进一步发展。  相似文献
