32P-Labelled tRNA was isolated from uninfected and phage phi 149-infected Vibrio cholerae cells. These tRNA preparations were then hybridised with DNA isolated from phage phi 149. Significant hybridisation was observed only with tRNA from phage phi 149-infected cells. This strongly suggests that infection of classical vibrio with phage phi 149 results in the synthesis of phage-specific tRNA molecules.  相似文献

A 28kDa chitosanase designated as Csn2 was purified from the culture broth of the fungus Gongronella sp. JG through three chromatography steps: CM-Sepharose FF, Superdex 200 and SP-Sepharose FF. Its optimal reaction pH and temperature were pH 5.6 and between 55 degrees C and 60 degrees C. The half-lives of Csn2 at 50 degrees C and 55 degrees C were estimated to be 30min and 11min, respectively. The K(m) value of Csn2 in sodium acetate buffer (pH 5.6) at 55 degrees C was 8.86mg/mL. Mn(2+), Ca(2+) and Sr(2+) were activators of Csn2; ETDA was an inhibitor. Cu(2+) stimulated Csn2 at 1mM, but inhibited Csn2 activity at 10mM. Csn2 displayed strong activity on colloidal chitosan, but did not hydrolyze colloidal chitin and carboxylmethyl cellulose. Thin layer chromatography analysis showed the end products of colloidal chitosan hydrolyzed by Csn2 were chitobiose, chitotriose and chitotetraose with chitotriose as the major product. The N terminus of Csn2 was determined to be YQLPANLKKIYDSHKSGTC. Part of the genomic DNA sequence corresponding to Csn2 was cloned. Sequence alignment showed DNA sequence of Csn2 was partly identical to chitosanase genes from Metarhizium anisopliae var. acridum, Hypocrea lixii and Aspergillus fumigatus. Based on sequence similarity, Csn2 was classified as a GH-75 chitosanase.  相似文献

The aim of this study was to estimate and analyse circadian variations of the renin-angiotensin-aldosterone system (RAA) activity in blood of goats and the influence of late pregnancy on the circadian variations of RAA system. The study was carried out on a group of 17 non-pregnant and 9 pregnant goats. The animals were kept in uniform environmental conditions, (9 h light/15 h darkness). Blood samples were collected seven times over a period of 24 h, every 4 h. Plasma renin activity (PRA), plasma aldosterone (PA), sodium, potassium and chloride concentrations were determined. PRA and PA of both groups changed during 24 h, with the highest values in the dark phase and with higher RAA system activity (especially during the night) in the pregnant goats. In the non-pregnant goats, no circadian changes in PRA and PA were observed. The circadian changes in PRA and PA found in pregnant goats had acrophases at 06:27 h and 01:13 h, respectively. Plasma electrolyte concentrations in both groups of goats also changed during 24 h. These results suggest that circadian changes of potassium concentration in plasma of goats during late pregnancy may be one of the main factors affecting the RAA system.  相似文献

The present study was designed to identify and functionally characterize potential cell surface extracellular matrix binding proteins in Hydra vulgaris. Using [3H]-laminin as a probe, radioreceptor analysis of a dissociated mixed hydra cell preparation indicated that the average number of laminin binding sites per cell was about 10,000 with a dissociation constant of 1.49 nM. These binding sites could be displaced with unlabelled laminin in a dose-dependent manner and with high concentrations (500 nM) of unlabelled fibronectin. No displacement with type-IV collagen and type-I collagen was observed. Immunoscreerting studies with a battery of antibodies raised to mammalian extracellular matrix (ECM) binding proteins indicated potential cell surface binding sites for the anti-β1 integrin monoclonal antibody, mAb JG22. Cell adhesion studies indicated that mAb JG22 blocked binding of hydra cells to laminin, but did not affect their binding to fibronectin, type-IV collagen, or type-I collagen. Light and electron microscopic immunocytochemical studies indicated that mAb JG22 localized to the basal plasma membrane of ectodermal and endodermal epithelial cells. Immunoprecipitation studies identified two major bands with masses of about 196 kDa and 150 kDa under reducing conditions, and two bands with masses of >200 kDa under non-reducing conditions. Functional studies indicated that mAb JG22 could reversibly block morphogenesis of hydra cell aggregates, and could block in vivo interstitial cell migration in hydra grafts. The Forest Stewardship Council developed the concept of High Conservation Values (HCVs) as a criteria in the forest certification process in order to promote sustainable forest management. It has six major components or values and component one and two of HCVs deal with the habitat for viable populations of “rare, endemic and threatened (RET) species” using the IUCN Red List category and other national / regional / local lists. But a consistent robust methodology for identification of these areas, does not exist. The present study tried to develop for the first time, a straight forward inclusive methodology for identification of HCVAs for the RET species on a spatio-temporal scale. A total of 50 RET and other significant species (32 flora, 10 fauna and 8 avifauna) were identified after a thorough literature review, field surveys and consultations with experts. Occurrence data of the selected species was collected from different secondary sources, field surveys, institutes and scientists who have worked on them. A 10 km grid-based approach and stratified random sampling was used for the primary GPS field surveys conducted during 2018–2019. MaxEnt species distribution model (SDM) software was used based on the occurrence data and environmental variables for identification of potential suitable habitats for the selected species. Linear support vector machine (LSVM) model was used for assessing the performance of the SDMs. The performance of each SDM has been validated through Cohen's Kappa (KAPPA), true skill statistic (TSS) and receiver operating characteristics (ROC) models. The proposed methodology addresses the urgent need for a holistic and robust set of techniques to apply the HCV toolkit. This is key to identify and map HCVAs for RET species at the landscape level and can be easily adapted to and adopted at the national, regional, state or local level in India. The methods offer an efficient, reliable approach for the application of the HCV concept, elsewhere in the world.  相似文献
