Summary Based on a dose-survival curve, a radiation dose of 3.99 C/kg was used to induce antibiotic-resistant mutants inBacteroides fragilis. Escherichia coli B/r membrane fragments were employed as a reducing agent. Antibiotic-resistant mutants ofB. fragilis were utilized to study the mechanism by which these organisms become resistant to selected chemotherapeutic agents. Decreased accumulation of tetracycline by resistant mutants ofB. fragilis suggests that the resistance to this antibiotic is associated with the outer membrane permeability. There is a marked difference in the inhibitory action of rifampicin on RNA polymerase activity in rifampicin-sensitive and-resistant strains ofB. fragilis. This enzyme is, therefore, the likely target for inhibition of bacterial growth in this anaerobe by rifampicin.  相似文献

Epithelial proliferation in the ventral surface of mouse tongue follows a pronounced circadian rhythm with a peak in mitotic activity at 10.00 a.m., preceded by a wave of DNA synthesis 8 h earlier. Nearly all cells (85%) pass through G2 and mitosis immediately after the S-phase; they subsequently divide again, usually after 2 or 3 days, indicating cohorts of cells with different G1-duration. The fraction of all nucleated cells comprised in one daily proliferation wave is about 20%, indicating a turnover time of the nucleated cell compartment of about 5 days. Cytotoxic injury by a single radiation dose of 20 Gy causes a steep decrease in cell counts, leading to complete denudation after 9–13 days. The difference between the latent period before ulceration and the tissue turnover time is explained by a marked proliferative activity of the doomed cells. The mitotic index increases steeply after day 1 to three times the control level, but most mitotic figures display gross abnormalities such as multipolar spindles or chromosome clumping. As a consequence cells with abnormal or multiple nuclei appear in the basal layers 3 days post irradiation and subsequently migrate to the upper layers. After denudation the epithelium rapidly becomes restored, with a phase of transient hyperplasia on days 13–14. Normal architecture is regained by day 15. Over the whole healing period the mitotic index remains at a high level, with most of the mitoses appearing histologically normal.  相似文献

Toxoplasma gondii: decreased resistance to intracellular bacteria in mice   总被引:1,自引:0,他引:1  
The effect of sublethal inocula of Toxoplasma gondii on the course of listeriosis and salmonellosis in mice was investigated. Intravenous injection of T. gondii 24 hr after inoculation of Listeria monocytogenes increased mortality from 16% (L. monocytogenes alone) to 68% (L. monocytogenes + T. gondii) (P less than 0.001). Multiplication of L. monocytogenes in spleens also was increased significantly in mice given T. gondii. By 3 days after infection, mice that had received T. gondii and L. monocytogenes had approximately 10 times the number of L. monocytogenes per spleen compared to mice receiving L. monocytogenes alone. Similarly, mortality and the number of bacteria in spleens were increased in mice injected with Salmonella typhimurium and then inoculated with T. gondii. An in vitro assay of macrophage listeriacidal activity was used to investigate the mechanism of this decreased resistance. Peritoneal macrophages from mice injected with T. gondii were less bactericidal than macrophages from uninfected mice. Delayed hypersensitivity responses to L. monocytogenes antigen were markedly suppressed in mice injected with T. gondii. T. gondii infection appears to suppress both macrophage and T-lymphocyte function and may result in decreased resistance to infections caused by intracellular bacteria.  相似文献

The effect of irradiation on the in vitro growth of Plasmodium falciparum was investigated. The cultured malarial parasites at selected stages of development were exposed to gamma rays and the sensitivity of each stage was determined. The stages most sensitive to irradiation were the ring forms and the early trophozoites; late trophozoites were relatively insensitive. The greatest resistance was shown when parasites were irradiated at a time of transition from the late trophozoite and schizont stages to young ring forms. The characteristics of radiosensitive variation in the parasite cycle resembled that of mammalian cells. Growth curves of parasites exposed to doses of irradiation upto 150 gray had the same slope as nonirradiated controls but parasites which were exposed to 200 gray exhibited a growth curve which was less steep than that for parasites in other groups. Less than 10 organisms survived from the 106 parasites exposed to this high dose of irradiation; the possibility exists of obtaining radiation-attenuated P. falciparum.  相似文献

The fluorescence intensity of diphenylhexatriene (DPH) and of trimethylammonium-diphenylhexatriene (TMA-DPH) is measured when these probes are embedded in vesicles of dipalmitoyl- and dioleoylphosphatidylcholine (DPPC and DOPC), in mixtures of these vesicles as well as in vesicles of the mixed phospholipids, in trout intestinal brush border membranes and in mitoplasts of rat liver cells. The intensity in DOPC vesicles is found to be significantly higher than in DPPC vesicles. When these systems are irradiated with strong ultraviolet light radiation, a decrease in the fluorescence intensity is observed; this effect is much stronger in DOPC than in DPPC vesicles. The fluorescence anisotropy values in the mixture of vesicles as well as in the membranes show an initial increase with irradiation which is followed by a significant decrease. A transfer of DPH molecules between DPPC and DOPC vesicles is observed. For TMA-DPH this transfer takes place only from DPPC to DOPC vesicles, but not vice-versa. These results are related to intensity and anisotropy measurements of these probes in cell cultures.  相似文献

用30—70GyX射线照射小麦幼苗(浸种后5天)后发现根毛区到根尖的距离缩短,根毛变密,根毛长度为对照的2—3倍。照射后2—3天就可看到该现象。根毛着生处到根尖的距离随剂量增加而减少,甚至根尖全为根毛所复盖,另外还看到有分叉的根毛。根尖纵切片表明根尖分生区随剂量增加而缩小,分生区后接着就出现输导组织。玉米和黄瓜也有类似现象。这现象说明根细胞的分裂过程对射线很敏感,而分化过程则相当耐辐射,细胞分裂停止后立即转向细胞分化过程。  相似文献

本文在首次报告二例肺结核、糖尿病患者合并感染肺微小根毛霉的基础上,进一步探讨免疫功能受损与微小根毛霉感染关系。本文采用不同剂量~(60)Co-γ全身辐照小鼠,然后以不同途径注射同剂量的微小根毛霉的孢囊孢子,观察动物的感染情况和感染后的真菌检出率,结果发现各种辐射剂量均在辐射后7-14日感染菌的检出率最高;各种脏器感染菌的检出率以脾脏最高(66.7—81.8%);淋巴结最低(0.0—25.0%);其他脏器的感染菌检出率也有不同程度的差异。  相似文献

摘要 目的:探究照射体积和时间与食管癌患者外周血淋巴细胞绝对值的相关性。方法:本研究方案将纳入2019年1月~2019年12月蚌埠医学院第一附属医院放疗科收治的放疗或同步放化疗食管癌患者84例,其中单独放疗患者24例,同步放化疗患者60例,采用血液细胞分析仪测定患者放疗期间每周复查外周血白细胞(WBC)、中性粒细胞(N)、淋巴细胞(L)、血红蛋白(HB)及血小板(PLT)计数等指标。Pearson相关性分析照射时间、剂量及体积与外周血指标之间的相关性。结果:食管癌放疗患者,包括同步放化疗及单纯放疗亚组,在治疗1-6周,照射时间与外周血指标均无相关性(P>0.05)。但在放疗第5-6周,患者放疗剂量与WBC、N、L、HB呈负相关(P<0.05),同步放化疗亚组患者照射剂量与WBC、N、L、HB呈负相关(P<0.05)。在治疗1-4周,不同照射剂量下各梯度照射剂量对应照射体积与外周血指标均无相关性(P>0.05)。但在第5-6周时,患者不同梯度照射剂量下各照射体积与WBC、N呈负相关(P<0.05),同时在20Gy-60Gy照射剂量,尤其20Gy和30Gy照射剂量下照射体积与L呈负相关(P<0.05)。同步放化疗亚组患者不同照射剂量下各照射体积与WBC、N呈负相关(P<0.05),同时在20Gy-60Gy照射剂量下照射体积与L呈负相关(P<0.05),而且在60Gy照射剂量下照射体积与HB呈负相关(P<0.05)。结论:放疗患者特别是同步放化疗亚组患者照射体积、照射剂量与食管癌患者外周血淋巴细胞计数成负相关,基线淋巴细胞与食管癌患者外周血淋巴细胞计数成正相关,而照射时间与食管癌患者外周血淋巴细胞计数无相关性。  相似文献
